The aim of this study was to examine the impact of the probiotics Saccharomyces cerevisiae (SC) and Pediococcus acidilactici (PA), as well as the prebiotic yeast cell wall extract (Cyberlindnera jadinii and S. cerevisiae) (YP), on the growth parameters, intestinal histomorphology, liver and gills normality, and gut microbiome of common carp (Cyprinus carpio L.). These feed supplements were subjected to a 60-day experimental period, during which 120 fish (26.4 ? 5.2 g) were distributed into four groups, with each group comprising 30 fish. The feed supplements were administered at a rate of 1 g/kg of body weight. Before the trial, the fish were acclimatized for two weeks, then injected with Passive Integrated Transponder (PIT) tags. The results showed that none of the feed supplements exhibited a significant effect (p > 0.05) on growth indices. In contrast, the villi length, villi width, muscular thickness, and crypt depth in the anterior, mid, and posterior intestine of the SC group exhibited significant (p < 0.05) improvements compared to the other groups. No alteration or abnormal growth were noticed in the gills and livers. The most dominant microbe genera in all groups, with abundances greater than 60% of the total, were Cetobacterium and Aeromonas. However, Polynucleobacter, Nordella, Mycoplasma, Romboutsia, and Staphylococcus species were present at lower abundances. The presence of Actinobacteria has been observed only in the intestine of fish that have been fed a diet supplemented with PA. It can be concluded that the tested probiotics and the yeast cell wall prebiotic have the potential to produce a remarkable improvement in intestinal morphology and a considerable change in the gut microbiome without notable effect on growth, livers, or gills of common carp.

Alteration in Gut Microbiome of Common Carp (Cyprinus carpio L., 1758) Mediated by Probiotics and Yeast Prebiotic / Elshafia Ali Hamid Mohammed; Milán Fehér; Péter Bársony; Károly Pál
Dátum:

Megjegyzések:
The objective of the present study was to examine the impact of dietary supplementation with probiotics and yeast cell wall prebiotics on the intestinal microbiota of common carp (Cyprinus carpio). A total of 96 carp, with an average body weight of 932 ? 161 g, were distributed into 12 fish tanks (800 L), with 8 fish/tank. The fish were fed a variety of experimental diets, including a basal diet only (CD) or a basal diet supplemented with the probiotic Pediococcus acidilactici (PA), the yeast probiotic Saccharomyces cerevisiae (SC), or the yeast cell wall prebiotic (YANG) at a concentration of 0.1% (1 g/kg) for a duration of 42 days. At the end of the trial, fish digesta were withdrawn, and the total bacterial community of the gut of common carp was analyzed using Illumina's NGS targeting the 16S rRNA gene. A Krona phyla richness pie chart showed that 11 bacterial phyla were recorded in fish fed YANG, with the top three phyla being Fusobacteria, Firmicutes, and Proteobacteria. In addition, 10 phyla were identified in fecal samples from carp fed PA, with the top three phyla being Proteobacteria, Firmicutes, and Fusobacteria. Furthermore, nine phyla were recorded for carp fed SC, with the top three phyla being Fusobacteria, Firmicutes, and Proteobacteria. However, carp fed a basal diet exhibited 14 phyla, with the most abundant phyla being Fusobacteriota, Bacteroidota, and Proteobacteria. This study concluded that the tested feed supplements could cause considerable alterations in the composition of the gut microbiome of carps reared in recirculating systems.

Megjegyzések:
The current study aimed to examine the relative impact of dietary supplementation on the intestinal microbiota of common carp, Cyprinus carpio L., 1758. Different types of dietary supplements, including the probiotic Pediococcus acidilactici, the yeast probiotic Saccharomyces cerevisiae, and a yeast-based multi-strain prebiotic, were tested at a concentration of 0.1% (1.0 g/1 kg). A total of 96 carps with an average body weight of 932?163 g were distributed in 12 fish tanks (1000 L), with 8 fish/tank. Fish received the experimental diet for 40 days. At the end of the trial, fish feces were withdrawn and the total bacterial community in the gut of common carp was analyzed using Illumina's NGS targeting the 16S rRNA gene. The Krona phyla richness pie chart showed that 11 bacterial phyla were recorded in fish fed the yeast prebiotic, with the top 3 phyla being Fusobacteria, Firmicutes, and Proteobacteria. Similarly, 10 phyla were found in fecal samples from carp fed P. acidilactici, with the top 3 phyla being Proteobacteria, Firmicutes, and Fusobacteria. Also, carp fed S. cerevisiae recorded nine phyla, the top three phyla being Fusobacteria, Firmicutes, and Proteobacteria. However, carp fed the basal diet only showed 14 phyla, with the most abundant phyla being Fusobacteriota, Bacteroidota, and Proteobacteria. This study concluded that the examined feed supplements can cause alteration in the composition of the gut microbiome of the carps reared in a recirculating system.

Megjegyzések:
Limited studies discussed the impact of probiotics and prebiotics on fish microbiome. This research aimed to examine the effects of different feed additives on the intestinal microbiota of the common carp (Cyprinus carpio). Three different types of feed additives including live lactic acid bacterium Pediococcus acidilactici, Saccharomyces cerevisiae and YANG prebiotic were tested at the concentration of 1 g/ kg. Fish with average body weight of 932?163 g were fed with supplemented diet of each type for 41 days in a recirculation system containing 12 units (size=1000 liter each unit, 8 fish per unit). Water temperature, pH, TDS, dissolved oxygen, NO- 2, NO-3, and NH+4 concentrations were maintained to be at the optimum levels in all fish tanks. At the end of the experiment, fish feces samples were collected and the bacterial communities structure found in the intestines of common carp were analyzed through a metagenomic approach using 16S rRNA sequencing. The results of the metagenomic analysis of carp fed with a diet supplemented with YANG showed 11 different groups of bacterial phyla detected with the top three phyla, Fusobacteria, Firmicutes, and Proteobacteria. In contrast, fecal samples from carp fed with P. acidilactici recorded 10 phyla, the top three phyla were Proteobacteria, Fusobacteria, and Firmicutes. Fish fed with S. cerevisiae recorded 9 phyla, the top three phyla were Fusobacteria, Proteobacteria, and Firmicutes. However, samples from carp fed with the control diet recorded 14 phyla, the top three phyla were Fusobacteriota, Proteobacteria, and Bacteroidota.

Effect of selected feed additives on growth performance and the intestinal microbiota of the Common carp (Cyprinus carpio) / Elshafia Ali Hamid Mohammed, Milán Fehér, Péter Bársony, Abdelhakam Esmaeil Mohamed Ahmed, Ayaz Mukarram Shaikh, Károly Pál
Dátum:

Megjegyzések:
The research aimed to examine the impact of various feed additives on the growth performance and intestinal microbiota of the common carp (Cyprinus carpio). Three different feed additives including Bactocell (live lactic acid bacterium Pediococcus acidilactici CNCM I-4622 MA 18/5M), Levucell (probiotic derived from the fungus Saccharomyces cerevisiae boulardii CNCM I-1079) and YANG prebiotic (Saccharomyces cerevisiae AQP1260, S. cerevisiae AQP 12988 and Cyberlindnera jandinii AQP 12549) were tested at the concentration of 1% (10 g/1 kg of basic diets) in addition to the control (basic feed only). Fish with average body weight (g) of 932?163 were fed with supplemented diet for 45 days in a recirculation system (RAS). The RAS contains 12 units (size=1000 liter each unit, 8 fish per unit). Water temperature, pH, total dissolved solids, dissolved oxygen, NO-2 , NO-3 , and NH+4 concentrations were maintained within optimal levels. After the end of the feeding experiment, fish feces samples were collected and the bacterial community structure found in the intestines of common carp was analyzed through a metagenomic approach using 16S rDNA sequencing. The results indicated that there was no significant effect (P<0.05) of the tested feed additives (Bactocell, Levucell and YANG) on growth performance (final body weight), feed conversion ratio (FCR) and specific growth rate (SGR). The results of the metagenomic data of carp fed with a diet supplemented with YANG showed 11 different groups of bacterial phyla detected with the top three phyla, namely Fusobacteria (55%), Firmicutes (36%), and Proteobacteria (6%). In contrast, fecal samples from carp fed with 1% Bactocell recorded 10 phyla. The top three phyla were Proteobacteria (70%), Fusobacteria (21%), and Firmicutes (5%). A sample of carp fed with Levucell recorded 9 phyla, the top three phyla were Fusobacteria (50%), Proteobacteria (35%), and Firmicutes (14%). Finally, samples from carp fed with the basal diet (the control) recorded 14 phyla. The top three phyla recorded were Fusobacteriota (68%), Proteobacteria (27%), and Bacteroidota (3%). The study found that the tested feed additives did not significantly affect the growth performance of common carp. However, they did lead to variations in the composition of the intestinal microbiota of the common carp.
