Megjegyzések:The ascomycete fungus, Cryphonectria parasitica (Murr.) Barr, is an important fungal pathogen of chestnut in Europe and North America. The fungus grows into the bark up to the cambium, causing typical diffuse cancers and the death of distal parts of the trunks or branches. The pathogen kills the infected tree branches and the rapid death of the entire tree take place which is causing high environmental and economic concerns. In this study we employed a part of the translation elongation factor 1 subunit alpha (EF-1α=tef1) and ITS region, as a potential genetic markers to infer phylogenetic relationships among Cryphonectria parasitica isolates. The translation elongation factor 1 subunit alpha (EF-1α=tef1) containing both introns and exons and ITS region containing the internal transcribed spacer regions 1 and 2 and the 5.8S rDNA, as a potential genetic markers to infer phylogenetic relationships among Cryphonectria parasitica isolates. Translation elongation factor 1 subunit alpha (EF1α=tef1) is part of the cytosolic EF1 complex, whose primary function is to promote the binding of aminoacyl-tRNA to the ribosome in a GTP-dependent process. Simultaneously, elongation factor 1α (EF-1α) is a highly conserved ubiquitous protein that has been suggested to have desirable properties for phylogenetic inference and population genetic studies. Ribosomal DNA (rDNA) has long been used as a potential marker for phylogenetic studies. rRNA genes are organized in clusters of tandemly repeated units, each of which consists of coding regions (18S, 5.8S, and 28S) and 2 internal transcribed spacers (ITS) and intergenic spacer (formerly called the Non-Transcibed Spacer, NTS) region. While the coding regions are evolutionarily conserved and have been utilized for phylogenetic inferences for major phyla, the 2 ITS regions are appropriate for detecting differences between co-specific individuals and are hence potentially useful markers to study the relationships of populations and closely related species in fungal, plant, and animal taxa due to their relatively rapid evolutionary rates.

Megjegyzések:Chestnut blight disease caused by the fungus Cryphonectria parasitica (Murr.) Barr is a devastating tree disease in forests and other areas with chestnut cultivation in central Europe. The virulent strain of the pathogen is responsible for the deadly necrotic lesions or cankers on infected chestnut trees. The discovery of Cryphonectria hypovirus 1 (CHV1) in Europe has opened wide possibilities for biological control of C. parasitica in sweet chestnut populations (Castanea sativa) once considered lost. Inoculation of virulent chestnut cankers with hypovirulent isolates of the fungus carrying CHV1 leads to non-lethal and inactive cankers. Since the hypovirus can only be transmitted by hyphal anastomoses among fungal individuals sharing the same vegetative compatibility type, it is imperative to analyse the diversity of vegetative compatibility types of C. parasitica to develop improved, site-specific biological control applications and disease management strategies. Infected bark samples of 12 chestnut trees were collected (in 2022) from the site Farkasgyepű (Veszprém county, Hungary). Isolates were cultured in the laboratory and vegetative compatibility between isolates was evaluated on PDA medium by a co-culturing assay. An important task of the laboratory work was also to identify local hypovirulent fungal strains and to determine their usability (conversion capacity) in biological control applications. The main aim of our research was to base of chestnut blight disease control with special emphasis on hypovirulent strains of C. parasitica. During our surveys, we visited a production site in west-Hungary (Farkasgyepű/Veszprém county) where infected bark samples had not yet been collected or hypovirulent fungal strains had not been used earlier. With our tests, we wanted to establish the successful field treatments. Based on the results, at least 2 vegetative compatibility groups of C. parasitica isolates were found on a relatively small plot. The first group (EU12) contains FG3, FG8, FG10 and FG12. The second group (EU13) contains FG5, FG6 and FG7 whereas FG2 and FG4 belongs to both groups ("bridge" isolates). At the same time, among the hypovirulent strains collected from the area, only 1 (FG2) was found to be suitable for in vitro conversion of all local, virulent isolates. It was also proven with a wound inoculation test on apple fruits that FG2, FG5, FG6 and FG7 hypovirulent isolates have the strongest hypovirulent effect on virulent isolates in in vitro conversions.

Megjegyzések:Chestnut blight caused by Cryphonectria parasitica, is among the most important fungal diseases of European chestnut. However the aggressiveness of the pathogen to other tree species of the Fagaceae family (oaks, beech) is increasing in relevance. Blight symptoms were detected on the bark of oak species (Quercus spp.) in many countries since 1960. In Hungary first observation of symptoms of the disease was reported on sessile oak in 2002. Then the fungus was reported also on oaks in Slovakia and in Romania. The main goal of our study was to examine the susceptibility of oak species to C. parasitica. Virulence of different isolates was examined on dormant sticks of 9 different oak species and European chestnut as a control. Shoots were infected with virulent isolates of C. parasitica. The size of the caused ellipse shaped cankers on the inoculated branches was measured. Necroses on red oak were significally larger in size than those on the chestnut. Q. rubra and C. sativa were more susceptible than the other species, and sessile oak (Q. petraea) and pedunculate oak (Q. robur) also showed a high susceptibility. Results demonstrate that most oak species are susceptible to C. parasitica (at various levels), red oak (Q. rubra) being the most susceptible to the chestnut blight fungus.
