Megjegyzések:AbstractEndogenous free radicals continuously form in theskin as a result of auto-, enzymatic oxidation (xanthineoxidase-superoxide dismutase system, inducible nitricoxide synthase), metabolic activity of subcellularorganelles (mitochondria, microsome, peroxisome),respiratory burst in leukocytes infiltrating the skin layersand transition metal ions. The skin is situated at theinterface between the body and the environment,functioning as a physical, chemical and biologicalbarrier. As a consequence of its localization it is oneof the organs most exposed to exogenous agents. Some exogenous noxa or molecules by themselves are202Eva Remenyik et al.capable of directly producing free radicals in the skin, but others are convertedinto highly reactive species by endogenous enzymes. The most importantexogenous factors of skin damage comprise are sunlight, cigarette smoke, airpollution, infections, cosmetics, topical and systemic drugs and nutrients.The molecular products of the oxidative stress serve as biomarkers and also caninitiate pathological conditions: sunburn, aging, photoaging, carcinogenesis,inflammation, thermal bum, chronic wound, psoriasis, acne, atopic dermatitis,scleroderma, vitiligo, cutaneous drug reactions, and porphyrias.Some of the therapeutic modalities used by dermatologists to treat thedifferent skin disease and conditions are based on free radicals mechanisms(phototherapies, irritants). Topicaly or systemic application of antioxidants,avoidance of exogenous compounds increasing free radicals generation, andtheoretically reduced calorie intake are approaches to prevent or eliminatefree radicals production, and avoid the initiation of pathological conditions.This paper discusses the oxidant and defense mechanisms of the skin andalso the deleterious effects of exogenous agents on the skin structure and funcions.
