Megjegyzések:	In the circulation one of the major sources of reactive-oxygen-species (ROS) production including superoxide anion (O2) are neutrophils. Enhanced O2 production by neutrophils were observed in several oxidative-stress-mediated diseases. Most natural antioxidant, however, did not scavenge O2 radical. In spite of that they - including silybin and its structural analogues inhibited O2 production in phorbol ester stimulated conditions via inhibition of protein kinase C (PKC). Maximal inhibition in PKC activity was achieved by silybin skeleton, which contained only a 4-keto group in ring C. Albeit, silybin-derived structures differentially affected NADPH oxidase activity in cell-free system, the final output in intact cells; e.g. reduction in O2 production depended only degree of PKC inhibition. These results suggest that silybin and its derivatives decreased ROS production in stimulated neutrophils by the inhibiton of the first step in phorbol-ester signaling that is the activation of PKC - possibly a competition for phorbol-ester binding sites in PKC.
