Megjegyzések:Atopic dermatitis (AD) is a chronic infl ammatory skin disease resulted partly of type I andtype IV hypersensitivity reactions triggered by T helper 2 (Th2) cells. The explanations behindthe Th2 dominance in the disease are still controversial. We addressed the question whetherthe Th2 cell domination is associated with a reduction and/or functional impairment ofCD4+CD25+FOXP3+ regulatory T cells (Treg) essential for the maintenance of Th1- Th2balance. Peripheral blood of AD patients with severe clinical symptoms and extremely highserum IgE levels (>2000 U/ml) were investigated. Flow cytometry was utilised to determinethe percentage of CD4+CD25brightFOXP3+ Tregs and CLA+CD4+CD25brightFOXP3+ Tregs in thesamples of AD patients and healthy controls. For detection of suppressor activity of CD4+CD25+Tregs, they were cocultured with CD4+CD25- effector T cells, and for T cell stimulation,anti-CD3/CD28 microbeads were applied alone or with Staphylococcus Enterotoxin B(SEB). The proliferation of the cells was measured by EZ4U colorimetric cell proliferationassay. Signifi cantly increased number of CD4+CD25brightFOXP3+ Tregs and also of CLA+CD4+CD25brightFOXP3+ Tregs were found in the peripheral blood of AD patients compared tohealthy individuals. The degree of suppressor activity of CD4+CD25+ regulatory T cells weredecreased in both anti-CD3/CD28 and SEB stimulated subgroups compared to control. Ourdata suggest coincidently with the literature that the pathogenesis of AD cannot be explainedwith the absence of CD4+CD25+FOXP3+ Treg cells, moreover the number of these cells isincreased in the peripheral blood. At the same time the function of Tregs may be impaired.
