Megjegyzések:Objective: Polymers (Polymethyl methacrylate; PMMA) are one the most frequently used material in dentistry, which is often causingallergic reactions. The aim of our study was to investigate peptide structures bound to PMMA in order to deduct molecules may bind toPMMA and responsible for allergic reactions.Method: The investigated heat cured polymethyl methacrylate was Orthocryl (Dentaurum, Germany). We used phage display technologyto select peptides on PMMA surface. The phage work was carried out by linear peptide library. Ph.D.-7T M and Ph.D.-12T M Peptide LibraryKits (Biolabs, England) were based on a combinatorial library of random peptide 7-mers and random peptide 12-mers fused to a minorcoat protein of M13 Phage respectively. The phage selection was carried out five times on the PMMA surface. C olonies of the third roundsof each phage selection were used to isolate DNS for amino acid sequence determination. The binding of the selected peptides wasquantitated by ELISA and by calculation of binding constants.Result: Efficacy of the phage selections resulted in 2.33x107 phages/ml on the titer plate. At the estimation of binding affinities of theselected phages it was found that the phages bound to the surface with saturation kinetic. The amino acid sequence of the peptide insertsof selected phages showed that mostly apolar, non-aromatic amino acid occurred among them the proline is the most frequent. TheNVQTRQPEKPVL sequence gave a relative low E-value at the comparison of protein data bank.C onclusion: Oligopeptides bound to the PMMA surface and among PMMA specific phages the proline and leucine is the most oftenoccurred amino acid. The NVQTRQPEKPVL sequence may occur in that haptens or immunmediators which can be responsible for theallergic reaction.
