Megjegyzések:The present study was designed to determinethe effect of anisocytosis on the association of MCVvalues with HbA1c and reticulocyte counts as markersof red cell age. Normo-, micro- and macrocytic samples,fractionated by counterflow centrifugal elutriation werestudied. The previously described correlation betweenMCV and HbA1c was only observed in normal samplesand in the middle fractions of samples with anisocytosis.At both extremes of the elutriation profile, curvesfor HbA1c content and reticulocyte count levelled out.Furthermore, in fractions containing the largest redcells of the microcytic series and the smallest red cellsof normo- and macrocytic samples, reticulocyte countdecreased while HbA1c content increased with increasingMCV. From these data it is concluded that MCV isnot an absolute determinant of red cell age in case ofanisocytosis.Key words Centrifugal elutriation 7 Meancorpuscular volume 7 Glycosylated hemoglobin 7Reticulocyte count 7 Red cell ageIntroductionTo reveal the aging process of red blood cells (RBC) amethod is required that reliably isolates them accordingto age. Previous investigations mostly used erythrocytesseparated on the basis of cell density [3]. Since the introductionof the counterflow centrifugation separationchamber, it is possible to isolate red cell fractions basedon differences in RBC volume [7, 9].The discovery that hemoglobin is non-enzymaticallyglycosylated during RBC life span has led to the conclusionthat HbA1c is a marker of red cell age [4]. Inmost studies based on counterflow centrifugal elutriationit was established that red cell age as marked byHbA1c content, decreased in parallel to an increase inMCV. Thus, it was concluded that cell age determinedRBC volume [8]. To investigate whether MCV was anabsolute determinant of red cell age, we studied a seriesof samples with anisocytosis and characterized red cellfractions of different MCV according to their HbA1cand reticulocyte content.
