Megjegyzések:Intrinsic structural disorder is a widespread phenomenon in eukaryotic proteomes. Proteins devoid of a well-defined three-dimensional structure carry out important functions most often related to signal transduction and transcription regulation. In terms of the underlying molecular mechanisms, the action of intrinsically disordered proteins (IDPs) or intrinsically disordered regions (IDR) of proteins either stems directly from the disordered structural state (entropic chain functions) or is realized via molecular recognition that leads to transient or permanent interactions with the partner molecule(s). The highly specific, fast, yet low-affinity binding by disordered proteins however, raises serious kinetic and thermodynamic issues, which need to be addressed to understand the function of IDPs in full detail. The effectivity in IDP recognition can be rationalized by the involvement of transient structural elements at the secondary and/or tertiary structural level. This model suggests that many IDPs are not fully disordered and exploit structural pre-organisation for partner binding. In this review the seven distinct functional modes of IDPs are surveyed, and are linked with experimental evidence for pre-formed structure of functional relevance. This treatise allows to interpret such structural and functional data in terms of various binding-related concepts, such as primary contact sites, molecular recognition elements, and pre-formed structural elements. The unifying model of "facile binding" is proposed to elucidate how IDPs combine extreme flexibility and limited structural order for fulfilling effective binding functions.
