Megjegyzések:Background: Status epilepticus, the most severe and damaging form of epilepsy, and spontaneous termination of seizures are poorly understood at present. Mechanisms were studied in a well-known model nervous system showing properties which correspond in every respect to those of a mammalian nervous system including man. Method: Identified giant neuronal individuals in the buccal ganglia of helix pomatia were intracellularly recorded for up to 24 hours. Structurally and functionally intact ganglia were kept under in-vitro conditions. Epileptiform activity was induced by bath application of pentylenetetrazol. Results: Paroxysmal depolarization shifts (PDS) and short interictal- like potentials appeared reliably in neuron B3 after begin of application of 40 mM of pentylenetetrazol. With ongoing drug application PDS-activity spontaneously decreased during 4h to 18h and it re-increased spontaneously thereafter. With lower concentration of pentylenetetrazol (25 mM) this PDS-decrease took several minutes only. To study whetherPDS induce PDS-decrease, one of both neurons B3 was blocked by injection of hyperpolarizing current into the soma. Maximal PDS-activity and subsequent PDS-decrease appeared only if the neuron was released from hyperpolarization. Injection of slow ramp-current into the soma showed that during PDS-decrease threshold of PDS slowly shifted from hyperpolarized to depolarized membrane potentials. PDS-decrease was accelerated with [Mg2+] which could not be replaced by [Sr2+]. Conclusion: It is concluded that termination of epileptic activity and avoidance of status epilepticus are primarily no network phenomena but result from cellular defence mechanisms triggered by PDS. ♭Zero Mg' does not introduce epileptogenicity but status epilepticus.
