Megjegyzések:The catalase enzyme is one of the earliest known and well studied enzymes. In the decades following its discovery in 1819, the enzyme catalase has been studied and detected in many types of living organisms, organs and tissues. Its substrate hydrogen peroxide was discovered in 1818. In the body, the catalase enzyme is responsible for eliminating hydrogen peroxide in toxic concentrations. The enzyme is encoded by a single gene (33114 Kb, 13 exons, 12 introns). Several polymorphisms and mutations have been identified for the catalase gene. Red blood cells contain 99.9% of blood catalase. The activity of the blood catalase can be easily determined by a widely used spectrophotometric method. It is a combination of optimized enzymatic conditions and the spectrophotometric assay of hydrogen peroxide based on formation of its stable complex with ammonium molybdate which could be measured at 405 nm. Acatalasemia occurs when the activity of the enzyme blood catalase is less than 10%. Hypocatalasemia is when blood cell activity is below 50% of the average. We have reported patients with acatalasemia and hypocatalasemia in diseases such as diabetes mellitus, microcyter anemia, vitiligo, and thalassemia. We've identified mutations and polymorphisms in the catalase gene that may be responsible for the decrease in enzyme activity. Mutations and polymorphisms are in exons and introns. The effects of the polymorphisms in the introns are currently unknown. According to the exon mutations of catalase gene eleven types of acatalasemia were established in Hungary. Enzyme catalase plays an important role in defense against the toxic effects of free oxygen species by the removal of hydrogen peroxide. Hydrogen peroxide may be formed in several pathologic conditions such as inflammation, cancer, erythrocyte metabolism, homeostasis and platelet activation. In this chapter we are discussing: the catalase enzyme (discovery, history, the relationship with hydrogen peroxide and reactive oxygen species, the blood and erythrocyte catalase), the catalase gene (its deficiency acatalasemia and hypocatalasemia, its mutations), the blood catalase in various diseases (diabetes mellitus, anemia, thalassemia, vitiligo) and the possible future of catalase research.
