Megjegyzések:Venous thrombosis (VT) is multifactorial trait that attributed to the burden of cardiovascular diseases worldwide. Although abundant single nucleotide polymorphisms (SNPs) provoke the susceptibility of an individual to VT; it seems five most strongly associated SNPs play the greatest role. Association and risk prediction models are rarely established by using merely 5-strongly associated SNPs. We aimed to explore five SNPs' [rs6025 (F5 Leiden), rs2066865 (FGG), rs2036914 (F11), rs8176719 (ABO), and rs1799963 (F2)] combined risk prediction ability in the Hungar ian population. SNPs were genotyped in clinically confirmed VT patients (n=298) and controls (n=400). Associations were estab lished using standard genetic models. Genetic risk scores (unweighted-unGRS, weighted- wGRS) were computed as well. Correspondingly, area under receiver operating characteristic curves (AUCs) for individual SNP, for 5 SNPs, and for non-genetic risk factors were estimated to explore VT risk predictability. The risk allele frequency of rs6025 was the highest for VT patient (OR=3.2 [95% CI: 2.50; 4.95]. The rs6025, rs2036914, and rs8176719 were associated with the trait using all genetic models. The rs2066865 was associated with VT us ing a multiplicative, additive, and recessive model only before the multiple correction testing. Conversely, rs1799963 did not show any significant association with VT. The wGRS was a better predictor than the unGRS (AUC=0.67 [95% CI: 0.63, 0.71] vs. AUC=0.65 [95% CI: 0.60, 0.69], respectively). Furthermore, the com bination of genetic and non-genetic (eg. age, sex, BMI) VT risk factors significantly increased the AUC to 0.87 [95% CI: 0.84, 0.89, p<0.0001). Conclusions: Our study revealed that strongly associated five SNPs combined with non-genetic factors could efficiently predict individual VT risk susceptibility.
