Megjegyzések:After-effects of seed ultrasonication (30 kHz, 70 W for 5 min) of winter wheat were investigated in present study. DNA methylation and mRNA transcriptomic patterns were studied and compared in 1-week-old seedlings. Whole genome bisulfide sequencing and mRNA seq. were used for identifying the differentially expressed genes (DEGs) and differentially methylated genes (DMGs) or regions (DMRs) . Seed ultrasonication caused increased growth of seedlings. Behinds this growth enhancement process, the global hypomethylation of DNA, and increased mRNA transcription related mainly to starch biosynthesis, IAA biosynthesis, photosynthesis and TCA cycle pathways were detected. Furthermore, the detected changes in gene transcription suggested that after ultrasonication there were alterations in IAA and sucrose signalling and thereby in crosstalk between them, in growth enhancement processes, and in transcription of genes that have the motif of CCAAT in their promoter. DNA methylation changes in DMGs were associated with the main pathways that were detected to be up-regulated in mRNA transcription. Our results suggested that seed ultrasonication acted as a priming technique that resulted in DNA hypomethylation leading to modification of mRNA transcription in seedlings causing finally an enhanced growth of seedlings.
