Megjegyzések:Previous studies showed that some lactones have beta-lactamase inhibitory or antibacterial effects, others--like A-factor (a gamma-butyrolactone) and its derivatives--stimulate sporulation in Streptomyces griseus strains. Our experiments were aimed at exploring whether synthetic gamma-lactones had such effects. None of the seven gamma-lactones studied showed antibacterial activity, but two of them inhibited beta-lactamases isolated from various bacteria. These two gamma-lactones did not reduce colony formation of murine bone marrow cells in vitro, indicating that they were not toxic to proliferating mammalian cells. Four gamma-lactones, including the two inhibiting beta-lactamase, stimulated sporulation in the non-sporulating S. griseus bald 7 mutant. Further studies of gamma-lactones as potential inhibitors of beta-lactamase seem to be warranted.

Megjegyzések:The borderline methicillin resistance in Staphylococcus aureus is due to ?-lactamase overproduction and/or specific methicillinases.Methods: beta-lactamase activity in culture supernatants and in cytoplasmic membrane fractions were estimated by bioassay and by SDS-PAGE combined with nitrocefin assay.Results: During the investigation of borderline methicillin-resistant Staphylococcus aureus (BORSA) strains VU94 and 822 two beta-lactamases were detected in the membranes, with molecular weights of 13 and 30 kDa. The second one could be found in the culture supernatants as well. In the presence of globomycin this enzyme disappeared from the membrane, and the oxacillin-hydrolysing activity of the membrane decreased to the level of susceptible strains. Both beta-lactamases were detected in the studied methicillin-resistant Staphylococcus aureus (MRSA) strain, but the susceptible strains possessed only the first enzyme.Conclusions: The 30 kDa ?-lactamase proved to be a methicillinase, and it can be one of the main causes of the borderline phenotype of BORSA strains. The other enzyme is one of the smallest published beta-lactamases until recently.

Megjegyzések:We read with interest the article of Munoz ~ et al.1 describing the performance of the T2Candida MR assay in patients receiving empirical antifungal therapy for suspected invasive candidiasis (IC).1 The study was well designed and fully warranted in attempting to address the demands of managing patients at risk of fungal disease and provides much needed real-life data on the performance of the T2 platform. However, unlike the authors, we feel the performance of the T2 assay in this study was insufficient to discontinue antifungal therapy.
