Langerhans cells (LCs) are the sole professional antigen-presenting cell normally found in the human epidermal compartment. Research into their physiological role is hindered by the fact that they are invariably activated during isolation from the skin. To overcome this challenge, we turned to a monocyte-derived LC model (moLC), which we characterized with RNASeq, and compared the transcriptome of moLCs to donor-matched immature dendritic cells. We found that moLCs express markers characteristic of LC2 cells, as well as transient receptor potential vanilloid 4 (TRPV4). TRPV4 is especially important in skin, as it has been linked to conservation of the skin barrier, immunological responses as well as acute and chronic itch, but we know little about its function on LCs. Our results show that TRPV4 activation increased the expression of Langerin and led to increased intracellular calcium concentration in moLCs. Regarding the functionality of moLCs, we found that TRPV4 agonism had a mitigating effect on their inflammatory responses, since it decreased their cytokine production, and T cell activating capability. As TRPV4 has emerged as a potential therapeutic target in dermatological conditions, it is important to highlight LCs as a novel target of these therapies.

The transcriptional basis of sebocyte differentiation and lipid production is mostly unclear. Peroxisome proliferator-activated receptor gamma (PPAR?), a lipid activated transcription factor, has been implicated in differentiation and lipid metabolism of various cell types. Here, we show that PPAR? is differentially expressed in normal and pathological human sebocytes, and appears to have roles in their differentiation and lipid production. We used laser microdissected normal and pathologic human sebaceous glands (SG) and SZ95 cells (immortalized sebocyte cell line) analyzed by RT-qPCR and immunohistochemistry. Lipids were analyzed by quantitative fluorimetry and mass spectrometry based approaches. We have observed that PPAR? and its target genes, ADRP and PGAR, are expressed in sebocytes and show association with their level of differentiation. Also, PPAR? is present in normal and hyperplastic SG, whereas its expression levels are decreased in SG adenoma and SG carcinoma cells, reflecting a maturation-linked expression pattern. Furthermore, in SZ95 sebocytes, naturally occurring lipids including arachidonic acid and arachidonic acid keto-metabolites (eg 5-KETE, 12-KETE) appear to regulate PPAR? signaling pathways, which in turn modulate phospholipid biosynthesis and induce neutral lipid synthesis. Collectively, our findings highlight the importance of endogenous ligand-activated PPAR? signaling in human sebocyte biology and suggest that PPAR? might be a promising candidate for the clinical management of SG disorders.

The in situ control of redox insult in human organs is of major clinical relevance, yet remains incompletely understood. Activation of nuclear factor (erythroid-derived 2)-like 2 (Nrf2), the "master regulator" of genes controlling cellular redox homeostasis, is advocated as a therapeutic strategy for diseases with severely impaired redox balance. It remains to be shown whether this strategy is effective in human organs, rather than only in isolated human cell types. We have therefore explored the role of Nrf2 in a uniquely accessible human (mini-) organ: scalp hair follicles. Microarray and qRT-PCR analysis of human hair follicles after Nrf2 activation using sulforaphane identified the modulation of phase II metabolism, reactive oxygen species clearance, the pentose phosphate pathway, and glutathione homeostasis. Nrf2 knockdown (small interfering RNA) in cultured human hair follicles confirmed the regulation of key Nrf2 target genes (i.e., heme oxygenase-1, NAD(P)H dehydrogenase, quinone 1, glutathione reductase, glutamate-cysteine ligase catalytic subunit, ABCC1, peroxiredoxin 1). Importantly, Nrf2 activation significantly reduced reactive oxygen species levels and associated lipid peroxidation. Nrf2 preactivation reduced premature catagen and hair growth inhibition induced by oxidative stress (H2O2 or menadione), significantly ameliorated the H2O2-dependent increase in matrix keratinocyte apoptosis and reversed the reactive oxygen species-induced reduction in hair matrix proliferation. This study thus provides direct evidence for the crucial role of Nrf2 in protecting human organ function (i.e., scalp hair follicles) against redox insult.

The pathogenesis of hidradenitis suppurativa/ acne inversa (HS) is still unclear, but is likely initiated by follicular occlusion of the pilosebaceousapocrine units (Hoffman et al., 2017) in the apocrine gland-rich (AGR) skin (armpits, intramammary fold, genital groin, perianal areas, and buttocks). The outstanding role of keratinocytes in the initiation phase was also highlighted by recently described signaling pathways (Notch and mTOR signaling, IL-36 pathway) related to HS pathophysiology (Balato et al., 2018; Di Caprio et al., 2017; Scala et al., 2018). Following keratinocyte activation, proliferation and follicular occlusion, immune cell infiltration, pro-inflammatory cytokine, and chemokine and antimicrobial peptide (AMP) production resulting in severe destruction of the surrounding tissue lead to fully developed immunemediated inflammation.

We have previously shown that endocannabinoids promote sebaceous lipogenesis, and sebocytes are involved in the metabolism of the "endocannabinoid-like" substance oleoylethanolamide (OEA). OEA is an endogenous activator of GPR119, a recently de-orphanized receptor, which is currently being investigated as a promising anti-diabetic drug target. Thus, in the current study, we investigated the effects of OEA as well as the expression and role of GPR119 in human sebocytes. We found that OEA promoted differentiation of human SZ95 sebocytes (elevated lipogenesis, enhanced granulation, and the induction of early apoptotic events), and it switched the cells to a pro-inflammatory phenotype (increased expression and release of several pro-inflammatory cytokines). Moreover, we could also demonstrate that GPR119 was expressed in human sebocytes, and its siRNA-mediated gene silencing suppressed OEA-induced sebaceous lipogenesis, which was mediated via JNK, ERK1/2, Akt/PKB, and CREB activation. Finally, our pilot data demonstrated that GPR119 was down-regulated in sebaceous glands of acne patients, arguing that GPR119 signaling may indeed be disturbed in acne. Collectively, our findings introduce the OEA?GPR119 signaling as a new positive regulator of sebocyte differentiation, and highlight the possibility that dysregulation of this pathway may contribute to the development of seborrhea and acne.
