It has been demonstrated that in the superficial spinal dorsal horn, Lamotrigine, which is known to block voltage-sensitive Na+ and Ntype Ca2+ channels, depresses neural activities evoked by sustained activation of nociceptive primary afferent fibres. In the present experiments, we study how Lamotrigine exerts its inhibitory effect on spinal nociceptive information-processing mechanisms. We show that Lamotrigine in an in vitro slice preparation effectively blocks synaptic transmission between primary afferents and secondary sensory neurons. Together with the robust increase in the failure rate and reduction in the amplitude of excitatory post-synaptic potentials (EPSPs) evoked by stimulation of nociceptive primary afferents, Lamotrigine causes a marked decrease in the number and amplitude of spontaneous EPSPs and a gradual shift of the resting membrane potential towards hyperpolarization. In addition, Lamotrigine treatment also changes the intrinsic firing pattern of superficial dorsal horn neurons. The results suggest that the effect of Lamotrigine on spinal nociceptive information-processing mechanisms is multiple: it depresses synaptic inputs from nociceptive primary afferents to secondary spinal sensory neurons and also weakens the intrinsic activities of nociceptive spinal neural circuits in the superficial spinal dorsal horn.

Meng et al. conjecture that pinwheel density scales with body and brain size. Our data, spanning a 40-fold range of body sizes in Laurasiatheria and Euarchonta, do not support this conclusion. The noncolumnar layout in Glires also appears size-insensitive. Thus, body and brain size may be understood as a constraint on the evolution of visual cortical circuitry, but not as a determining factor.

PURPOSE: To demonstrate local cortical activations in the primary visual cortex of the cat as a result of retinal electrical stimulation by means of a completely wireless-controlled, implantable retinal prosthesis in a series of acute experiments. METHODS: The transfer of energy to drive the device and signals to activate any combination of 25 retinal electrodes was achieved completely wirelessly by an external transmitter positioned in front of the eye. Individually configured electrical stimuli were applied via any combination of 25 electrodes, on sending the necessary pulse parameters to the implant. Placement of the implant onto the retinal surface was achieved after lensectomy and vitrectomy in the cat. Fixation was performed with a retinal tack. Cortical activation patterns were recorded by means of optical imaging of intrinsic signals. RESULTS: Implantation and fixation were successfully performed in three cats. Wireless activation of the implant by radiofrequency was demonstrated by recording of stimulus artifacts from the sclera. Local activation of the visual cortex measured by optical imaging of intrinsic signals revealed a shift of cortical response that was well correlated with a change in the position of the activated retinal electrodes. CONCLUSIONS: The results demonstrate the retinotopic activation of the visual cortex using a completely wireless, remote-controlled retinal implant.
