Megjegyzések:In this paper our aim is to examine whether research conducted on human participants with LSD-25 (lysergic acid diethylamide) raises unique research ethical questions or demands particular concerns with regard to the design, conduct and follow-up of these studies, and should this be the case, explore and describe those issues. Our analysis is based on reviewing publications up to date which examine the clinical, research and other uses of LSD and those addressing ethical and methodological concerns of these applications, just as some historical examinations of this subject. The first chapters of the paper give an overview regarding the history of LSD-research with human participants, healthy volunteers and patients alike. The remaining chapters have a focus on questions regarding the potential ethical issues of such human trials in the contemporary research ethics framework. We also consider briefly political and regulatory issues regarding this substance that possibly affect its clinical and research applications.

Megjegyzések:The importance of optimal blood pressure control for preventing or reducing the impairment of vascular and cognitive functions is well known. However, the reversibility of early alterations in vascular and cognitive functions through antihypertensive agents is under-investigated. In this study, we evaluated the influence of 3 months of angiotensin-converting enzyme (ACE) inhibition treatment on the morphological and functional arterial wall and cognitive performance changes in 30 newly diagnosed primary hypertensive patients. Common carotid intima-media thickness (IMT) and brachial artery flow-mediated dilatation (FMD) were detected by ultrasonography. Arterial stiffness indicated by augmentation index (AIx) and pulse wave velocity (PWV) was assessed by arteriography. Cognitive functions were assessed by neuropsychological examination. The executive function overall score was significantly higher at 3-month follow-up than at baseline (median, 0.233 (IQR, 0.447) vs ?0.038 (0.936); P?=?.001). Three-month ACE inhibition did not produce significant improvement in IMT, FMD, AIx and PWV values. Significant negative associations were revealed between IMT and complex attention (r?=??0.598, P?=?.0008), executive function (r?=??0.617, P?=?.0005), and immediate memory (r?=??0.420, P?=?.026) overall scores at follow-up. AIx had significant negative correlations with complex attention (r?=??0.568, P?=?.001), executive function (r?=??0.374, P?=?.046), and immediate memory (r?=??0.507, P?=?.005). PWV correlated significantly and negatively with complex attention (r?=??0.490, P?=?.007). Timely and effective antihypertensive therapy with ACE inhibitors has significant beneficial effects on cognitive performance in as few as 3 months. Early ACE inhibition may have an important role in the reversal of initial impairments of cognitive function associated with hypertension-induced vascular alterations.

Megjegyzések:Purpose/Introduction: High level functioning autism (HLFA) is a distinctentity among autism spectrum disorders (ASD) characterized by autistic traitsunaccompanied by mental retardation. Our aim was to employ resting statefunctional MRI (r-fMRI) and diffusion tensor imaging (DTI) augmentedwith a graph theoretical approach to describe the pathological connectomein HLFA subjects.Subjects and Methods: Subjects were sampled from the a public dataset(n=209). Inclusion criteria for the HLFA group were adult age, elevated AutismSpectrum Questionaire (ASSQ) values and a marked discrepancy between theperformance IQ and verbal IQ measurements, 9 such subjects were found.Controls (n=40) with normal ASSQ values were age and gender matched tothis group while preserving the criteria of the IQ mismatch. Using the func-tional connectivity of r-fMRI data, individual graphs were created. For eachbrain region, network properties (strength, efficiency) were calculated. DTIwas used to determine the diffusion anisotropy in cortical, subcortical andmajor white matter regions.Results: Anisotropy patterns were characterized by significantly elevated valuespredominantly in frontal, temporal regions, gyrus cinguli and the thalamus.Graph analysis revealed reduced local efficiency and strength of the associ-ated networks with a pattern similar to DTI results. No differences in globalnetwork parameters were found.Discussion/Conclusion: A widely accepted hypothesis is that the autisticconnectome is dominated by a reduction of long-range cortico-cortical con-nections and an excessive presence of short fibers. We provided a comparisonof local attributes of structural and functional connectivity. The spatial distribution of the HLFA connectome phenotype is in accordance with previouslysuggested models.

Megjegyzések:We will analyse the representations and conceptualisation of genetics and genetic information in bioethical discourse. Genetics and genetic information is widely believed to be revolutionizing medicine and is sometimes misconceived as having a high predictive value compared to traditional diagnostics. We will attempt to present the inherent limitations of genetic information within its health care context. We will also argue against the exceptional treatment of genetic information that seems to govern bioethical reflection and regulatory approaches. And finally, we will make the claim that geneticization should be seen as a more serious threat to our privacy, autonomy and freedom, than genetic discrimination.
