Cím:A simple technique for the estimation of regional blood content in brain / László Csiba, Dániel Bereczki
Dátum:1994
ISSN:0165-0270
Megjegyzések:A simple and inexpensive technique is described for the assessment of regional cerebral blood volume (CBV) using a color reaction based on the pseudoperoxidase activity of hemoglobin. Cryostat sections are placed on a commercially available reagent strip originally designed for quantitative estimation of blood content in urine. After melting of the brain section, a blue color reaction appears which is darker in areas with higher hemoglobin content. The image is analyzed by a computerized image-processing system and evaluated by standards prepared from brain homogenate with different hemoglobin content. From the optical density data, regional CBV is calculated with correction for the difference between microvascular and large vessel hematocrit. The method was validated by a radiolabel procedure. Values of regional CBV are reported in gray matter areas of control rabbits and in brains embolized by autologous clot. This simple inexpensive technique is a useful supplementary tool in multiparametric metabolic studies.

Cím:Visualization of the ischemic core on native human brain slices by potassium staining method / Csiba Laszlo, Farkas Szabolcs, Kollar Jozsef, Berenyi Ervin, Nagy Katalin, Bereczki Daniel
Dátum:2010
ISSN:0165-0270
Megjegyzések:The potassium staining method is based on the formation of potassiumcobaltnitrite crystals after the treatment by sodium cobaltnitrite ofbrain tissue. The degree of staining correlates with the distinctpotassium content of infracted and non-infarcted brain areas. The aim of the present study was to prove that potassium stainingtechnique is a reliable method for localization of ischemic core onnative whole hemisphere cryosections of stroke patients. Furthermore, potassium stained sections have been compated with appropriatepostmortem MRI images of respective brains.Brains of stroke patients were removed within 24 h after death andpostmortem MRI scanning was performed. Horizontal cryosections offrozen brains were taken and potassium staining was performed. Usingthe stained whole hemisphere sections as "map" tissue sampling has beenmade in order to determine water and potassium content.Potassium content of infarcted samples was significantly decreased incomparison with intact regions (0.7346 +/- 0.2142 mg/L and 1.928 +/-0.447 mg/L, respectively, p<0.01) (mean values +/-SD). Water content ofaffected areas (expressed in percents) has been found to be abovenon-infarcted regions (81.657% +/- 4.07 and 72.96% +/- 6.37,respectively, p<0.01).According to our results the potassium staining method of human wholehemisphere brain sections reliably differentiates focal ischemic areasfrom intact brain regions. In conclusion, the postmortem examination ofischemic brain could be started with making the potassium map ofinfarcted whole hemisphere cryosections providing guidance for targetedtissue sampling and base of comparison for further examinations. Cím:Ischemia induces a translocation of the splicing factor tra2-beta 1 and changes alternative splicing patterns in the brain / Daoud R., Mies G., Smialowska A., Oláh L., Hossmann K. A., Stamm S.
Dátum:2002
Megjegyzések:Alternative splice-site selection is regulated by the relative concentration of individual members of the serine-arginine family of proteins and heterogeneous nuclear ribonucleoproteins. Most of these proteins accumulate predominantly in the nucleus, and a subset of them shuttles continuously between nucleus and cytosol. We demonstrate that in primary neuronal cultures, a rise in intracellular calcium concentration induced by thapsigargin leads to a translocation of the splicing regulatory protein tra2-beta1 and a consequent change in splice-site selection. To investigate this phenomenon under physiological conditions, we used an ischemia model. Ischemia induced in the brain causes a cytoplasmic accumulation and hyperphosphorylation of tra2-beta1. In addition, several of the proteins binding to tra2-beta1, such as src associated in mitosis 68 and serine/arginine-rich proteins, accumulate in the cytosol. Concomitant with this subcellular relocalization, we observed a change in alternative splice-site usage of the ICH-1 gene. The increased usage of its alternative exons is in agreement with previous studies demonstrating its repression by a high concentration of proteins with serine/arginine-rich domains. Our findings suggest that a change in the calcium concentration associated with ischemia is part of a signaling event, which changes pre-mRNA splicing pathways by causing relocalization of proteins that regulate splice-site selection.
