Megjegyzések:Nuclear receptors are transcription factors that bind directly to DNA. Upon activation by their lipid soluble ligands they alter the expression of their target genes, playing important roles in metabolic processes, proliferation, differentiation, apoptosis, and immune responses of the cells on the level of gene expression regulation. Our aim was to examine the role of nuclear receptors that are known to be expressed in the skin such as Peroxisome Proliferator- Activated Receptors (PPARs), Retinoic Acid Receptors (RARs), Retinoid X Receptors (RXRs) and Liver X Receptors (LXRs) in in vitro differentiated and in UV exposed keratinocytes.In the experiments we used primary keratinocytes differentiated by raising the Ca2+ levels of the culturing medium to 1.2mM in a 0-6 days period and exposed to UVB (30mJ/cm2) on the last day of differentiation. The changes in the expression levels of the marker genes of differentiation (TGM1, INV, PI3) and in the expression levels of the nuclear receptors and their target genes were followed by Real Time Quantitative RT-PCR technique. After verifying the differentiation of the keratinocytes by detecting the induction of the marker genes we measured the mRNA expression levels of nuclear receptors. We found that mRNA levels of PPAR alpha, RAR beta/delta, RAR gamma, RAR alpha, RXR alpha RXR beta and LXR apha did not change markedly. Measuring the expression levels of the known target genes for the PPAR pathways (ADRP, CPT1, FABP4, ABCG1) it was found that while PPAR beta/delta and alpha could be activated, PPAR gamma was inactive when using specifi c, synthetic activators. Although in vitro UV exposure of the keratinocyte cultures resulted in decrease both in the expression levels of PPAR isotypes and in the expression levels of their target genes, the responsiveness of PPAR beta/delta to its activator was still at high levels. Our results further confi rm the role of PPAR beta/delta in keratinocyte homeostasis.

Megjegyzések:Background The healthy human skin with its effective antimicrobial defense system forms an efficient barrier againstinvading pathogens. There is evidence suggesting that the composition of this chemical barrier varies between diseases,making the easily collected sweat an ideal candidate for biomarker discoveries.Objective Our aim was to provide information about the normal composition of the sweat, and to study the chemicalbarrier found at the surface of skin.Methods Sweat samples from healthy individuals were collected during sauna bathing, and the global protein panelwas analysed by label-free mass spectrometry. SRM-based targeted proteomic methods were designed and stableisotope labelled reference peptides were used for method validation.Results Ninety-five sweat proteins were identified, 20 of them were novel proteins. It was shown that dermcidin is themost abundant sweat protein, and along with apolipoprotein D, clusterin, prolactin-inducible protein and serum albumin,they make up 91% of secreted sweat proteins. The roles of these highly abundant proteins were reviewed; all of whichhave protective functions, highlighting the importance of sweat glands in composing the first line of innate immunedefense system, and maintaining the epidermal barrier integrity.Conclusion Our findings with regard to the proteins forming the chemical barrier of the skin as determined by label-freequantification and targeted proteomics methods are in accordance with previous studies, and can be further used as astarting point for non-invasive sweat biomarker research.

Megjegyzések:Little is known about the altered lipid metabolism-related transcriptional events occuring in sebaceous glands of patients with acne vulgaris. Peroxisome proliferator-activated receptor (PPAR)?, a lipid-activated transcription factor, is implicated in differentiation and lipid metabolism of sebocytes. We have observed that PPAR? and its target genes, ADRP (adipose differentiation related protein) and PGAR (PPAR? angioprotein related protein) are expressed at lower levels in sebocytes from patients with acne than in those from healthy controls (HCs) Furthermore, endogenous PPAR? activator lipids such as arachidonic acid-derived keto-metabolites (e.g. 5KETE, 12KETE) are increased in acne-involved and nonacne-involved skin of patients with acne, compared with skin from healthy individuals. Our findings highlight the possible anti-inflammatory role of endogenous ligand-activated PPAR? signaling in human sebocyte biology, and suggest that modulating PPAR?- expression and thereby signaling might be a promising strategy for the clinical management of acne vulgaris.

Megjegyzések:Peroxisome proliferation activated receptor gamma (PPAR?) has been implicated in lipid metabolismand also in infl ammation. PPAR? is expressed in lipid rich human sebocytes and it appears to haveroles in their functions. The details of PPAR?-regulated lipid metabolism in sebocytes are, however,not well understood. Therefore our aim was to characterize the expression patterns of PPAR?, itsheterodimeric partner RXR and target genes in normal and pathologic sebaceous glands in tissuesand in the SZ95 sebocytes . Therefore we applied formaldehyde fi xed parraffi n embedded skinsamples of patients with sebaceous hyperplasia, sebaceous carcinoma for immunohistochemistry.From fresh frozen skin samples, we laser microdissected normal sebaceous glands and isolatedRNA for RT-PCR. We studied function of PPAR? molecule in SZ95 sebocyte culture by addingPPAR? agonist rosiglitazone (RSG), RXRalfa agonist LG268 or arachidonic acid (AA). Viabilityof the cells was measured by MTT test. Nile red staining and quantitative fl uorimetric analysiswere used to detect changes in polar and neutral lipid content levels as a differentiation markerof sebocytes. We found that PPAR? and RXR? proteins are present in normal and hyperplasticsebaceous glands, immortalized SZ95 cell line, but PPAR? is barely expressed in sebaceouscarcinoma. We also demonstrated that mRNA of PPAR? and its lipid-metabolism associatedtarget genes, ADRP and PGAR were present in both models, but only sebaceous glands in situexpressed FABP4. Target genes responded to activation or inhibition of PPAR? and RXR? inSZ95 sebocytes. These data suggest that PPAR? as a transcription factor is likely to play a role innormal and pathological sebaceous gland biology, thus possibly serving as a relevant target forfurther investigations in sebaceous gland-associated dermatoses and a potential target of therapy.

Megjegyzések:Sebaceous gland is a holocrine gland with accelerated lipid metabolism. On the otherhand, peroxisome proliferation activated receptor gamma (PPAR?) is a nuclear receptor,functioning as a transcriptional factor. Together with retinoid X receptor alpha (RXR?) it formsa heterodimer and regulates gene expression. Certain lipid ligands activate or inactivate thisreceptor complex. PPAR? has a key role in adipocyte lipid metabolism and differentiation;sebaceous glands were also reported to express PPAR?. Our goal is to assess if PPAR? hasa role in sebocyte function and/or diseases associated with this cell type. Human SZ95sebocytes and skin samples of healthy individuals and patients with dermatoses were usedfor immunhistochemistry to determine the presence of the receptor proteins. In addition,we isolated mRNA from laser microdissected sebaceous glands to detect gene expressionwith RTqPCR. The function of the receptor was studied in SZ95 sebocytes by adding thePPAR? agonist rosiglitazone (RSG), the PPAR? antagonist GW9668, interleukin4 (IL4), theRXR? agonist LG268 or arachidonic acid (AA). Quantitative fluorimetric analyses wereused to detect changes in lipids. We detected PPAR? and RXR? proteins in samples fromheathy individuals, patients with sebaceous hyperplasia, sebaceous carcinoma and inSZ95 sebocytes. The expression of PPARs in SZ95 sebocytes and also expression of targetgenes (ie. ADRP, LXR?) were detectable. By adding RSG, IL4 and AA no change in certainPPAR? target genes was detected, however changes in sebocyte lipid content occurred.GW9668 treatment decreased sebaceous lipids. Our data suggest that PPAR? is likelyto play a role in sebaceous lipid synthesis, thus it is possibly a relevant target for furtherinvestigation in sebaceous gland-associated dermatoses and a potential target of therapy.

Megjegyzések:Blepharochalasis is a rare disorder characterized by recurrent painless periorbital oedema, which leads to atrophy of the periorbital skin. Pathomechanism of the disease is probably immunological with a nearly complete loss of elastic fibres. The authors describe a 17-year-old woman, who was followed for 7 years. IgA deposits were found in the periorbital tissues, which confirm the immunological background of the condition. Electron microscopy results show that not only elastic but also collagen fibres were affected.

Megjegyzések:The transcriptional basis of sebocyte differentiation and lipid production is mostly unclear. Peroxisome proliferator-activated receptor gamma (PPAR?), a lipid activated transcription factor, has been implicated in differentiation and lipid metabolism of various cell types. Here, we show that PPAR? is differentially expressed in normal and pathological human sebocytes, and appears to have roles in their differentiation and lipid production. We used laser microdissected normal and pathologic human sebaceous glands (SG) and SZ95 cells (immortalized sebocyte cell line) analyzed by RT-qPCR and immunohistochemistry. Lipids were analyzed by quantitative fluorimetry and mass spectrometry based approaches. We have observed that PPAR? and its target genes, ADRP and PGAR, are expressed in sebocytes and show association with their level of differentiation. Also, PPAR? is present in normal and hyperplastic SG, whereas its expression levels are decreased in SG adenoma and SG carcinoma cells, reflecting a maturation-linked expression pattern. Furthermore, in SZ95 sebocytes, naturally occurring lipids including arachidonic acid and arachidonic acid keto-metabolites (eg 5-KETE, 12-KETE) appear to regulate PPAR? signaling pathways, which in turn modulate phospholipid biosynthesis and induce neutral lipid synthesis. Collectively, our findings highlight the importance of endogenous ligand-activated PPAR? signaling in human sebocyte biology and suggest that PPAR? might be a promising candidate for the clinical management of SG disorders.
