Megjegyzések:The main goal of our research was to study comprehensively the differences between the two phenological forms of the socially parasitic and globally threatened Large Blue (Maculinea arion)in the Carpathian Basin using four character sets (mitochondrial sequences, allozymes, male genitalia and wing morphometrics). Comparative analyses of distance matrices, phylogenetic trees and ordination patterns have been applied. The genetic and morphometric patterns revealed by our studies were disordant. While we experienced a significant differentiation between the 'spring' and 'summer type' of M. arion in both wing and genital traits, the two phenological forms did not show any genetic differentiation on two mitochondrial loci and in allozymes. At the same time, all individuals were infected by Wolbachia. Although certain wing traits may not represent reliable tracers of phylogeny because of the particular adaptive significance, the wing characteristics involved in our research are probably determined genetically. Additionally, the significant differentiation of male genitalia also indicates incipient prezygotic isolation arising from phenological differentiation between the 'spring' and summer arion'. It is possible that all extant differences between the two forms are attributable to (1) different host-ant use, (2) incipient speciation, (3) cytoplasmatic incompatibility (CI) by Wolbachia or the combination of these factors. In addition, discordant results indicate that the combined use of different approaches and data sets is strictly necessary to clarify systematic and evolutionary relationships.

Cím:Pattern of genetic differentiation in the Maculinea alcon species group (Lepidoptera, Lycaenidae) in Central Europe / J. Bereczki, K. Pecsenye, L. Peregovits, Z. Varga
Dátum:2005
ISSN:0947-5745
Megjegyzések:The taxonomic status of Alcon Blues living in Central and Western Europe (conventionally: Maculinea alcon and Maculinea rebel11 is quite confused. Some authors distinguish them as separate species of the M. alcon species group, while others consider them as subspecies or simply ecological forms. Our aim was to study the pattern of genetic differentiation among several populations of these taxa with reference to their taxonomic position.Imagoes were collected from 27 localities in Central Europe between 1999 and 2003. Samples originated from four regions: northern Hungary, central Hungary, Romania: Transylvania and Slovenia. Enzyme polymorphism was analysed using polyacrylamide gel electrophoresis. In all samples 16 enzyme loci were studied. In the analysis of the data, F-statistics was computed and the total genetic variation was partitioned into within and between population components. Nei's genetic distances were calculated and UPGMA dendrogram was constructed on the basis of the distance matrix. Hierarchical F-statistics and AMOVA was computed to study the pattern of genetic differentiation among the samples. Principal component analysis (PCA) was also carried out using the allele frequencies of the samples. The results of all analyses indicated a high level of differentiation among the samples. The differences among the samples collected in different years from the same population accounted for a considerable amount of the between sample variation. The distribution of the between sample variation did not exhibit a species pattern. The results of AMOVA and PCA indicated that the geographic pattern was slightly more expressed in the between sample variation compared with the species pattern of it. These results seem to contradict with the conventional taxonomical subdivision

Cím:Taxonomic composition and ploidy level among European water frogs (Anura: Ranidae: Pelophylax) in eastern Hungary / Dávid Herczeg, Judit Vörös, Ditte G. Christiansen, Michal Benovics, Peter Mikulíček
Dátum:2017
ISSN:0947-5745
Megjegyzések:The western Palaearctic water frogs in the genus Pelophylax comprise several distinct species and three hybridogenetic hybrid forms. In this study, wefocus on the Pelophylax esculentus complex, which consists of two sexual species, Pelophylax ridibundus and Pelophylax lessonae, and their hybridogenetichybrid, Pelophylax esculentus. Specifically, we investigated taxonomic composition and ploidy level of water frogs sampled in three differenttypes of wetland habitats in the Hortob agy National Park (HNP), eastern Hungary. Using variation in serum albumin intron 1 (SAI-1) and 15microsatellite loci, we detected the presence of all members of the P. esculentus complex in the studied localities. In one locality, all three taxaoccurred syntopically, while in others water frog populations consisted of P. ridibundus and P. esculentus exclusively. The genomic composition ofthe 63 examined hybrid specimens analysed with microsatellites showed the occurrence of diploid genotypes only. We used a population geneticapproach (allelic richness, gene diversity, multilocus genotypes and multilocus disequilibrium) to infer the breeding system of water frogs at HNP. Ourdata indicate that at least in two populations, hybrids form gametes with clonally transmitted P. ridibundus genome and produce a new hybrid generationby mating with P. lessonae.

Megjegyzések:Since genetic variation is the basis of evolutionary potential of a species, its structure needs to be understood. Thus, the aim of this study was to analyze and contrast the structure of genetic and phenotypic variation in the Euphydryas aurinia populations of southeastern central Europe. Genetic variation was studied by two types of molecular genetic markers: mtDNA COI sequences and allozymes. As the great hiatus in the European distribution of E. aurinia is located in the central part of the Carpathian Basin, we expected that the populations East and West to this gap would be highly differentiated. Populations of Central Transdanubia actually represent the easternmost margin of the West European distribution of E. aurinia. In view of the peripheral position of these populations, we supposed to find some genetic sign of local adaptation, as a consequence of diversifying selection and an increased level of fluctuating asymmetry as a result of environmental stress. The analyses of the molecular genetic markers revealed a basic East-West differentiation among the populations of southeastern central Europe which was further structured in the western part of the study area. The results suggested that the genetic differentiation between the two western regions is probably the consequence of diversifying selection. The pattern of phenotypic differentiation among the western populations, however, was different. A geographic cline was revealed (decreasing wing size) toward the eastern margin of the distribution in parallel with increasing fluctuating asymmetry. The conservation inferences of the results are considered.

Cím:Long-term survival and diversification of an endemic Melitaea species in mountains of Iran and adjacent areas / János P. Tóth, Zoltán Varga, Judit Bereczki
Dátum:2016
ISSN:0947-5745 1439-0469
Megjegyzések:Disjunct distribution patterns regularly resulted in the separation of different genetic lineages in glacial refugia. Recent patterns of survival and expansion have been often revealed by climatic niche modelling. We used the combination of genetic markers, geometric morphometry and climatic niche modelling to clear up the taxonomy and reconstruct the potential range of an endemic Iranian, taxonomically disputed Melitaea species in climatically different epochs. Our results show that this species (Melitaea abbas Gross and Ebert 1975, comb. n. = M. zagrosi T oth and Varga syn. nova) is clearly separated from all taxa of the Melitaea phoebe species group and only occurs in Iran and Azerbaijan but were also predicted for some adjacent regions. Molecular markers and distribution modelling show consistently that this species should have had a longterm survival in this area, and its range could have been slightly larger during the last glacial maximum than currently. Based on the studied molecular markers, three main groups in M. abbas can be recognized: those of steppic area of Azerbaijan, western Iran and northeastern Iran. Each group is characterized by own mitochondrial haplotypes, but also a high level of genetic diversity appears in the central part of the distribution area (Zagros Mts.).
