Megjegyzések:he effects of Cr(VI) were investigated on the growth rate, element, photosyn-thetic pigment and amino acid composition of Chlorella pyrenoidosa. Cr(VI) is toxic to Chlorella pyrenoidosa. The influence of chromium on cell density and cell number followed very similar trends, indicating that these growth responses might be correlated. The EC50 value for Cr(VI) were 2.0 mg dm-3, the lethal concentration of chromium appears to be approximately 20 mg dm-3for Chlorella pyrenoidosa. We have investigated the concentrations of chromium, calcium, mag-nesium and iron. The cells were fractionated into three fractions: cell wall fraction, membrane fraction, soluble fraction after the cells were disrupted. The amount of metals in whole cells and in each cell fraction was determined. Chromium uptake at each concentration was high within 3 days. Chlorella pyrenoidosa can accumulate chromium mainly (approximately 70%) in the cell wall. The concentrations of chromium and calcium show parallel changes with each other. A higher calcium concentration can be observed along with an increasing chromium concentration, both in the cell wall system and in the whole cells. Iron and magnesium concentration show a decreasing tendency. Cr(VI) caused a changes both in free amino acids and proline content. Both free amino acids and proline content increase with the increasing concentration of chro-mium. Chlorophyll a and b content show a decreasing , while OH-chlorophylls show increasing tendency. Rate of carotene ? and ? change grows with the increasing chromium concentration. The toxic properties of Cr(VI) can arise from the possibly free diffusion across cell membranes and strong oxidative potential. The toxicological impact of Cr(VI) originates from the action of this form itself as an oxidizing agent, as well as from the formation of free radicals during the reduction of Cr(VI) to Cr(III) occuring inside the cell.

Megjegyzések:In this work small-scale variation of leaf morphological, phenological and isoenzyme pattern was investigated win trees belonging to the Lepidobalanus sub-genus in a sessile oak-Turkey oak stand in order to make an exact taxonomic identification for further studies on physiological tolerance to climatic fluctuations. Classification functions based onleaf morphological traits suggest that most of the trees belongs to Q.petraea, but the introgression effects of Q. dalechampii and Q. polycarpa is also significant in the forest stand which might favour the drought tolerance in these groups of trees. Two representatives (Q. pubescens, Q. virgiliana) of Lanuginosae series are also present in the foreststand. The maximum difference in budburst time was two weeks among the trees. Representatives of Lanuginosae series can be described by late budburst. The isoenzyme analysis did not reveal taxon-specific allels, but some loci exhibited considerable differences in the frequency of allels among the different series and budburst groups.

Megjegyzések:In this study the variability of leaf growth traits was investigated within the canopy ABSTRACT of the two dominant tree species of Quercetum petraeae-cerris forest community, covering 30 % of the present forested area in Hungary. The investigation were performed in the "Sikf©okut Project" LTER site (Jakucs 1985). We compared the present leaf growth traits of species with those reported by Virágh (1985) for years of 1973-1975 before the decline of sessile oak in the site. After bud break leaves of the two canopy layers exhibited different rate in the increase of leaf area (La) and leaf mass (Lm). In spring shade leaves showed more intense increase in leaf area than in leaf mass. This resulted in a transitional large reduction of SLM in the shade leaves. In case of sessile oak the full leaf size was achieved within the same period in 2003 and 2004 but the area of mature leaves were larger in both canopy layers in 2004 than in 2003. In case of sessile oak the area of mature leaves in growing seasons of 2003 and 2004 increased significantly as compared to the period before the forest decline (Virágh 1985). In the upper canopy layer of Austrian oak the leaf area and leaf mass did not differ significantly from those reported for the period before the forest decline. In the lower layer of the canopy, however, the increase of leaf area was observed but the leaf mass exhibited similar values. In 2004 the loss of leaf area in the sun layer was 17% and in the shade layer 6 % as a consequnce of photophagous damages. In case of Austrian oak the loss of leaf area was smaller and amounted only to 1-5%.

Megjegyzések:The aim of this study is to reveal the histological structure of galls induced by Neu-roterus quercusbaccarum and to identify accumulated secondary metabolism products and stor-age materials in gall tissue by histochemical methods. The galls induced by N. quercusbaccarum showed a concentrical layer tissue structure under light microsope. Directly next to the chamber, a protein and lipid containing nutritive tissue can be found, which is covered by sclerenchyma layer consisted of two large cell plates in the lenticular galls of the unisexual generation, while the bisexual generation induced grape shaped galls lack lignifed sheaths. Our results confirm previous findings. The outer layer of the gall is the voluminous parenchymatic cortex with a supplying vascular network and covered epidermis. We proved with histological methods, that the galls really contain accumulated nutritives -proteins, lipids and starch- in large quantities. The concentration of these nutrients from the chamber toward the border of inner-gall, show a decreasing gradient in the case of proteins and lipids, and an increasing gradient for starch. We present the differences and similarities in histological structure among galls induced by two generations of N. quercusbaccarum and the well studied Biorrhiza pallida.

Megjegyzések:The effects of exclusion of UV-B radiation on leaf physiology of beech seedlings regenerating in a deforested area were studied in field. Under UV-B exclusion the chlorophyll content of leaves was higher, the chlorophyll a/b ratio and the carotenoid content was lower than in leaves under the ambient level of UV-B radiation. In leaves developed under the exclusion of UV-B a smaller degree of conversion of violaxantin to zeaxantin and atheraxantin was found at noon. The results showed that the ambient level of UV-B radiation decreased the maximum photochemical efficiency (Fv/Fm) of the dark-adapted leaves, especially at noon. Beech leaves contained significantly higher quantities of UV-B absorbing compounds on the basis of leaf area at the ambient UV-B than under the UV-B exclusion. Flavonoid accumulation took place in close correlation with the increase of specific leaf weight as compared to the leaves developed under UV-B exclusion.

Megjegyzések:Responses of beech seedlings to supplemental UV-B radiation were investigated ABSTRACT during three consecutive seasons, in three repeated experiments. Our attention was paid on the alteration of the photosynthetic pigment composition - especially on the xanthophyll cycle pigments - chlorophyll fluorescence parameters, furthermore the accumulation of UV-B absorbing compounds in leaves, the specific leaf mass and leaf water content. The enhanced UV-B radiation generally affected significantly neither photochemical efficiency of PSII, nor photosynthetic pigment composition. UV-B radiation induced some protective mechanisms, thus VAZ-pool increased in beech leaves in every experiment, parallel with the enhancement of non-photochemical quenching. Amount of UV-B absorbing compounds in leaves increased under enhanced UV-B, but no significant changes were observed in the specific leaf mass. Sensitivity of plants to UV-B is largely influenced by other environmental factors and experimental conditions.

Megjegyzések:We present a list of tissue culture systems developed at the Department of Botany,University of Debrecen. Several of tissue cultures were developed for the first time in our laboratory.These include micropropagation of Elatine hungarica, callus cultures of several oak genotypesof Hungarian origin and of Crocus species characteristic for the Carpathian Basin. Calluscultures were either organogenic (e.g. Crocus scepusiensis) or embryogenic (Quercus petraea,Galanthus and Crocus species, Phragmites australis). In case of embryogenic cultures, somaticembryos showed either normal, bipolar development (e.g. Crocus heuffelianus, Phragmitesaustralis) or they have lost this bipolarity at the maturation of embryos (C. banaticus, C. sativus,Galanthus). The type of callus is yet to be identified for several cultures (e.g. Plantago lanceolata,Vicia faba). Most of our in vitro cultures proved to have plant regeneration potential. Severalof them derived from endangered/ red list plants (Crocus species, Elatine hungarica, Galanthusnivalis), therefore they are suitable for germplasm preservation. Others (Quercus, Phragmitesaustralis) proved to be suitable for stress physiology and/ or cell biology experiments. Culturessuch as Crocus sativus or Plantago lanceolata derived from plants of medicinal importance,therefore are of potential pharmacological use.

Megjegyzések:In this work pigment composition and chlorophyll fluorescence characteristics were studied in seedlings of Fagus sylvatica regenerating in contrasting light environments of forest understory, forest edge and deforested area. The results have shown that the survival of beech seedlings under high light conditions in the forest edge and deforested area, as well as their acclimation to excess light require effective functional plasticity of the photosynthetic apparatus and protective mechanisms. Physiological acclimation of beech seedlings to high light involves significantly the carotenoids both in the short-term through the reversible conversion of violaxanthin to zeaxanthin and in the long-term through the increase of total carotenoid content and the size of xanthophyll cycle pool.

Megjegyzések:The aim of this work was to compare the photosynthetic responses of two tree species (Quercus petraea, Quercus robur) and to determine the leaf level protection under enhanced UV-B radiation in outdoor experiments. Three year old seedlings of both species were exposed to enhanced UV-B radiation (supplementing the ambient level with 40%) from bud break until leaf senescence. Under enhanced UV-B, increases of specific leaf mass (SLM) and reduction of leaf chlorophyll content per dry weight basis were observed but the total carotenoid content remained high. Both oak species responded with an increase of VAZ pool to enhanced UV-B. High values of potential photochemical efficiency of PS2 (Fv/Fm) were observed in both species during the growing season. On clear sunny days loss of Fv/Fm was observed from morning till midday which was larger under enhanced UV-B for both species suggesting that UV-B exposure enhanced the sensitivity of seedlings to photoinhibition. The enhanced UV-B induced increases in the amount of UV-B absorbing compounds in leaves of Q. robur but it slightly affected these traits of leaves of Q. petraea.

Megjegyzések:The effects of drought and heat wave appeared in growing season of 2003 on some physiological traits of Quercus petraea were studied at the Sikfőkút research site. There were comparative measurements in the subsequent humid growing season of 2004. Drought reduced significantly the leaf mass increment of Quercus petraea in the dry year (2003) which was reflected in lower SLM too as compared to 2004. Chlorophyll content showed inter-annual and within-canopy variability. Carotenoid content was 1,5-2 times larger in spring than in summer. In summer of 2003 VAZ pool was larger especially in sun leaves than in 2004. Values of Fv/Fm were lower in 2004 than 2003 which indicated lasting drought effects on the photochemical activity of leaves of sessile oak.

Megjegyzések:In the inland saline area two structurally different sands of Lepidio- Puccinellietum limosae community can be distinguised: open stand with low plant cover (mostly by Lepidium crassifolium) and closed stand with dense plant cover (mostly by Puccinellia limosa). The open habitat can be characterised by higher salinity degree. The leaf area of L. crassifolium is significantly lower on the open stand. The high water content per unit leaf area indicates the succulent character of the species and is involved in the avoidance of high salt (Na+) concentrations in the most saline habitat. There was remarkable accumulation of proline in leaves that can act as an osmotically active compound in the citoplasm. The osmotic potential of the tissue sap follows the changes of the proline concentrations, both diurnally and with the differences of habitats. Significant midday depression of Fv/Fm was found in the open habitat due to the unfavourable soil conditions and other additional abiotic factors such as higher irradiance and temperature.

Megjegyzések:Spirodela polyrhiza (L.) Schleiden (giant duckweed) is an extensively studied representative of duckweeds (Lemnoideae subfamily, Araceae). Despite its widespread application in numerous research areas and well-known capability for producing turions, the effects of various toxicants on turion production are sporadically discussed in the scientific literature. Our aim was to study the growth of S. polyrhiza under cadmium stress and to test its potential interference with turion production. Effects of cadmium were assessed in axenic cultures of a local S. polyrhiza clone in 7-days-long duckweed growth inhibition tests. Our results indicated that higher cadmium concentrations decreased the relative growth rates of cultures within 3 days and growth inhibition reached its maximum between the 3rd and 5th days with 0.080 mg l-1 IC50. Parallel with slowed frond production - by mirror image pattern - induction of turion formation was observed. The first turions protruded by the 5th day of treatments in the 0.075-0.75 mg l-1 concentration range. Such rapid onset of turion formation is indicative of an almost immediate switch between normal and turion mode of meristem function upon cadmium stress. The consideration of this mechanism in practical applications of S. polyrhiza is recommended.
