Megjegyzések:Based on our previous works involving two 1,4,7-triazacyclononane (tacn)-based ligands Hno2py1pa (1-Picolinic acid-4,7-bis(pyridin-2-ylmethyl)-1,4,7-triazacyclononane) and Hno1pa (1-Picolinic acid-1,4,7-triazacyclononane), we report here the synthesis of analogues bearing picolinate-based rz-conjugated ILCT (IntraLigand Charge Transfer) transition antenna (HL1, HL2), using regiospecific N-functionalization of the tacn skeleton and their related transition metal complexes (e.g. Cu2+, Zn2+ and Mn2+). Coordination properties as well as their photophysical and electrochemical properties were investigated in order to quantify the impact of such antenna on the luminescent or relaxometric properties of the complexes. The spectroscopic properties of the targeted ligands and metal complexes have been studied using UV-Vis absorption and fluorescence spectrocopies. While the zinc complex formed with HL1 possesses a moderate quantum yield of 5%, complexation of Cu2+ led to an extinction of the luminescence putatively attributed to a photo-induced electron transfer, as supported by spectroscopic and electrochemical evidences. The [Mn(L2)](+) complex is characterized by a fluorescence quantum yield close to 8% in CH2Cl2. The potential interest of such systems as bimodal probes has been assessed from radiolabeling experiments conducted on HL1 and( 64)Cu(2+) as well as confocal microscopy analyses and from relaxometric studies carried out on the cationic [Mn(L2)](+) complex. These results showed that HL1 can be used for radiolabeling, with a radiochemical conversion of 40% in 15 min at 100 degrees C. Finally, the relaxivity values obtained for [Mn(L2)](+), r(1p) = 4.80 mM(-1).s(-1) and r(2p) = 8.72 mM(-1).s(-1), make the Mn(II) complex an ideal candidate as a probe for Magnetic Resonance Imaging.

Megjegyzések:In order to assess the Al(III)-binding abilities of phosphorylated proteins and peptides, the interactions of Al(III) with the building blocks O-phosphoserine (PSer) and O-phosphotyrosine (PTyr) were studied. pH-metric and 31P NMR measurements were carried out to determine the stoichiometries and stability constants of the complexes formed, and to establish the most probable binding sites of the metal ion. PSer was found to bind Al(III) in a monodentate manner at the phosphate moiety, and in a tridentate manner with the simultaneous coordination of all donor groups. PTyr binds Al(III) only at the separate phosphate function. Citric acid (Cit) proved to be a stronger Al(III) binder at the physiological pH, though, it was able to displace PSer only in a rather slow process through formation of the trinuclear species Al3(Cit)3(OH). The results were used to evaluate the potential role of Al(III) in inducing the formation of neurofilamentous aggregates in neurological disorders.

Megjegyzések:Stability constants of the complexes formed in the reaction of [Pd(bpma)]2+ [bpma=bis(pyridin-2-ylmethyl)amine] with monodentate nitrogen and thioether ligands including uridine, MeUH, cytidine, MeC, EtGH, AcHis, AcHm, AcLys and AcMet were determined by potentiometric method. The coordination chemistry of [Pd(bpma)]2+ shows a significant similarity to that of [Pd(terpy)]2+, but it is different from [Pd(dien)]2+. The formation of hydroxo and dinuclear complexes is especially enhanced in the case of [Pd(bpma)]2+ and [Pd(terpy)]2+, but the affinity of palladium(II) ions for the coordination of thioether residues is reduced in the presence of pyridine nitrogen atoms. Stopped-flow kinetic measurements reveal that the substitution reactions of the thioether ligand AcMet are much faster than those of the N-donor cytidine. The presence of the two pyridyl residues significantly enhances the kinetic reactivity of [Pd(bpma)]2+ as compared to that of [Pd(dien)]2+. The Pd-S(thioether) bonded species can be important intermediates in multicomponent systems, but the equilibrium state is characterised by the formation of Pd-N bonded species. The complex [Pd(bpma)NO3]NO3 has been prepared in solid state and its structure was elucidated by single crystal X-ray diffraction method.

Cím:A new, model-free calculation method to determine the coordination modes and distribution of copper(II) among the metal binding sites of multihistidine peptides using circular dichroism spectroscopy / Ősz Katalin

Megjegyzések:A new calculation method to determine microscopic protonation processes from CD spectra measured at different pH and Cu(II):ligand ratios was developed and used to give the relative binding strengths for the three histidines of hsPrP(84-114), a 31-mer polypeptide modeling the N-terminal copper(II) binding region of human (homo sapiens) prion protein. Mutants of hsPrP(84-114) with two or one histidyl residues have also been synthesized and their copper(II) complexes studied by CD spectroscopy. The 1-His models were analyzed first, and the molar CD spectra for the different coordination modes on the different histidines were calculated using the general computational program PSEQUAD. These spectra were deconvoluted into the sum of Gaussian curves and used as a first parameter set to calculate the molar spectra for the different coordination modes (3N and 4N coordination) and coordination positions (His85, His96 and His111) of the 2-His peptides. The calculation method therefore does not require the direct use of CD spectra measured in the smaller peptide models. This is a significant improvement over earlier calculation methods. In the same runs, the stepwise deprotonation pKmic values were refined and the pH-dependent distribution of copper(II) between the two histidines was determined. The results revealed the high, but different copper(II) binding affinities of the three separate histidines in the following order: His85 << His96 <= His111. The calculation also showed that molar CD spectra which belong to the same coordination mode and coordination position in different ligands have very similar transition energies but different intensities. For this reason, direct transfer of molar CD spectra between different ligands may be a source of error, but the pKmic values and the copper(II) binding preferences are transferable from the 2-His peptides to the 3-His hsPrP(84-114).

Megjegyzések:Complex forming capabilities of [(?6-p-cymene)Ru(H2O)3]2+ with aminohydroxamates (2-amino-N-hydroxyacetamide (?-alahaH), 3-amino-N-hydroxypropanamide (?-alahaH) and 4-amino-N-hydroxybutanamide?(?-abhaH)) having the primary amino group in different chelatable position to the hydroxamic function were studied by pH-potentiometry, NMR and MS methods. Formation of stable [O,O] and mixed [O,O][N,N] chelated mono- and dinuclear species is detected in partially slow with ?-alahaH and ?-alahaH or in fast processes with ?-abhaH and the formation constants of the complexes present in aqueous solution are reported. Synthesis, spectral (NMR, IR) and ESI mass spectrometric characterization of novel dinuclear ?-alaninehydroximato complexes containing the half-sandwich type Ru(II) core is described. The crystal and molecular structure of [{(?6-p-cymene)Ru}2(?2-?-alahaH?1)(H2O)Br]Br?H2O (1) and [{(?6-p-cymene)Ru}2(?2-?-alahaH?1)(H2O)Cl]BF4?H2O (2) was determined by single crystal X-ray diffraction method. In the complexes one half-sandwich core is coordinated by a hydroxamate [O,O] chelate while the other one by [Namino,Nhydroxamate] fashion of the bridging ligand. In both cases the remaining coordination sites of one of the Ru cores are taken by a halide ion whiles the other one by a water molecule. Reaction of 2 with 9-methylguanine indicates the N7 coordination of this simple DNA model. Complexes 1 and 2 were tested for their in vitro cytotoxicity using human-derived cancer cell lines (A2780, MCF-7, SKOV-3, HCT116, HeLa) and showed no anti-proliferative activity in the micromolar concentration range.

Megjegyzések:Rigidification of the ligand scaffolds has been a particular mechanism of interest employed to achieve properties suitable for MRI contrast, catalysis, or other applications of metal complexes. Towards the goal of targeting a 15-anePyN5Pip type ligand, a serendipitous isolation of a 30-anePy2N10Pip2 aza-macrocycle was achieved, instead. X-ray diffraction and determination of pKa events were carried out and compared to 17-anePyN5Pip. Furthermore, the X-ray diffraction of the Cu(II) and Zn(II) complexes of 17-anePyN5Pip was achieved and compared to previous reports of other first-row transition metal derivatives of this ligand. Determination of the log beta with both 30-anePy2N10Pip2 and 17-anePyN5Pip with the divalent Mnsingle bondZn metal-ion series was used to demonstrate the impact that the piperazine ring plays compared to other, less rigid macrocycles reported to date.

Megjegyzések:Three triaza macrocyclic ligands, H6NOTP (1,4,7-triazacyclononane-N,N·,Nı-trimethylene phosphonic acid), H4NO2AP (1,4,7-triazacyclononane-N-methylenephosphonic acid-N·,Nı-dimethylenecarboxylic acid), and H5NOA2P (1,4,7-triazacyclononane-N,N·-bis(methylenephosphonic acid)-Nı-methylene carboxylic acid), and their gallium(III) chelates were studied in view of their potential interest as scintigraphic and PET (Positron Emission Tomography) imaging agents. A 1H, 31P and 71Ga multinuclear NMR study gave an insight on the structure, internal dynamics and stability of the chelates in aqueous solution. In particular, the analysis of 71Ga NMR spectra gave information on the symmetry of the Ga3+ coordination sphere and the stability of the chelates towards hydrolysis. The 31P NMR spectra afforded information on the protonation of the non-coordinated oxygen atoms from the pendant phosphonate groups and on the number of species in solution. The 1H NMR spectra allowed the analysis of the structure and the number of species in solution. 31P and 1H NMR titrations combined with potentiometry afforded the measurement of the protonation constants (log KHi) and the microscopic protonation scheme of the triaza macrocyclic ligands. The remarkably high thermodynamic stability constant (log KGaL =34.44 (0.04) and stepwise protonation constants of Ga (NOA2P)2? were determined by potentiometry and 69Ga and 31P NMR titrations. Biodistribution and gamma imaging studies have been performed on Wistar rats using the radiolabeled 67Ga(NO2AP)? and 67Ga (NOA2P)2?chelates, having both demonstrated to have renal excretion. The correlation of the molecular properties of the chelates with their pharmacokinetic properties has been analysed.
