Megjegyzések:Grapevine Trunk Diseases (GTD) are amongst the most important diseases in vineyards. GTD have been characterized only recently because of a worrying increase of the frequency of their symptom expression worldwide. Esca-, Petri- and black-foot disease, Eutypa- and Botryosphaeria- dieback are considered as major GTDs, are caused by a variety of pathogens which attack the woody perennial organs of the vine and ultimately lead to the death of the plant. Control of GTD is problematic, no grapevine cultivar is known to be resistant to GTD and there are no highly effective treatments available. The present knowledge of epidemiology, host-pathogen interaction and microbial ecology is still poor. The objective of this study was to isolate and identify fungi associated with symptoms of GTD and from canes of healthy grapevines in vineyards from the Tokaj wine region. The plantation studied is 21 years old of grapevine variety called Furmint. Four rows of grapevine were chosen for the collection of samples. Pure fungal cultures were used for morphological and molecular identifications of fungal species. Chips were cut, disinfected and incubated on potato dextrose agar medium at room temperature. Fungal DNA was obtained from pure fungal colonies after 3 days of growth on 50 ml potato dextrose broth on a rotary shaker (125 rpm). Mycelia were harvested by vacuum filtration. Total genomic DNA was extracted from freeze-dried mycelium and isolated using NucleoSpin Plant II (Macherey-Nagel) according to the protocol, followed the manufacturer's instructions. DNA concentrations were measured by NanoDrop (Thermo Scientific) and used in PCR amplification of the internal transcribed spacer region (ITS) and the 5.8S ribosomal subunit with primers ITS4 and ITS5 [1]. The PCR products were purified by using Nucleospin Gel and PCR Clean-up (Macherey-Nagel). Purified amplification products were sequenced by MWG Biotech Company in Germany. Sequences obtained were assembled and manually corrected. Reference sequences for the Botryosphaeriaceae were obtained from GenBank. Phylogenetic analyses were performed with MEGA. One thousand boostrap analyses were run to determine confidence levels at the branching points. The principal aim of this work is to assess the GTD types and their occurrence in the Tokaj wine region as well as the identification of the pathogenic fungi (isolated from symptomatic and asymptomatic grapevine trunks) on morphological and genetic basis. In this study, 101 fungal isolates were identified from grapevine woody tissues. The majority of them (72.3% ) were determined as member of Botryosphaeriaceae, 66 sample were determined as Diplodia seriata , 6 Botryosphaeria stevensii. Other fungi, like Alternaria, Mucor, Phoma, Phomopsis, Diaporthe and Xylaria also were identified from grapevine samples.

Megjegyzések:Grapevine trunk diseases (GTD), caused by different phytopathogenic fungi threaten the vineyards worldwide and are one of the most important diseases in the Hungarian vineyards as well. They are caused by several fungal pathogens and the environmental factors has probable role as well. GTD pathogens can be detected only from the woody tissues of the plants. These fungi can often be found on asymptomatic plants as well. Currently there is no adequate and environmentally friendly protection against GTD pathogens. The aim of our work was to detect the pathogenic and non-GTD pathogenic endosymbiotic fungi from the grapevines, and to search for potential biocontrol agents from the healthy, asymptomatic plants.Pure fungal cultures isolated from wood tissues were used for morphological and molecular identifications. Different endophytic and GTD pathogen fungi could be isolated from the grapevines from the Tokaj wine region between 2013 and 2014. Diplodia seriata and Diaporthe species were identified as GTD pathogen fungi from the diseased woody tissue. The presence of D. seriata pathogen was 72%, in 2013, while in 2014 it was only 48%. The Diaporthe sp. pathogens had identified in 4% and 3% in 2013 and 2014 in the diseased trunks. The identified non-GTD pathogen endophytic fungi were Alternaria sp., Mucor sp., Penicillium sp., Epicoccum sp., Fusarium sp., Botrytis and Aspergillus sp. The rate of these fungi was 72% in 2013, while in 52% in 2014.Several Trichoderma species were also isolated from the asymptomatic grapevines. Endophytic, mycoparasitic fungi, like Trichoderma species have huge potential against grapevine plant pathogens colonizing woody tissues. Testing new isolates with biocontrol potential and ability to grow under different environmental conditions may provide effective biopesticide to control GTD pathogens. The Trichoderma isolates were identified based on their ITS1,2 and tef1 marker sequences. The ten isolates belonged to three species: Trichoderma harzianum (eight isolates), Trichoderma orientalis (one isolate) and Trichoderma viride (one isolate). Their mycelial growth was determined from average colony diameters on PDA at different temperatures between 5 and 37?C. T. orientalis and one of T. harzianum isolates showed the highest growth rates within the whole temperature range, and their growth rates were especially high at 30 and 37?C. The mycoparasitic ability was tested in vitro against GTD pathogens (D. seriata and Neofusicoccum parvum). All Trichoderma overgrew the GTD pathogens, and sporulated on their colonies, therefore their Biocontrol Index was 100%. Some GTD symptomatic plants were treated with Trichoderma spore suspension in Tokaj wineyard in 2014. The treated trunks were checked monthly. The symptoms decreased on all treated samples, except one. Moreover the Trichoderma strains could be isolate again from their woody tissues 10 month following the treatment.

Megjegyzések:Trichoderma harzianum is mycoparasite with antagonistic and mycotoxic metabolite production properties. Trichoderma species can be used as biopesticides against pathogenic fungi. Currently there is no effective fungicide against Fusarium oxysporum in grapevine trunk; therefore our aim was to find potential biopesticide against it.Eight mycoparasitic Tichoderma harzianum isolates (TR01-TR05; TR07; TR09; TR10) were tested in vitro to examine their mycelial growth rate. Those were isolated from grapevine trunk disease (GTD) asymptomatic grapevine trunks from the Tokaj wine region, Hungary. The average linear growth rate (ALG) of the isolates was tested at five different temperatures (18.5-37C). The mycoparasitic ability of the Trichoderma isolates against Fusarium oxysporum isolated from GTD symptomatic plants was also determined. 5 mm mycelial disks from the margin of seven days old colonies were cut, and put in 3 cm distance on PDA plates, than incubated at room temperature for 6 days. Finally growth inhibition value was calculated to express the mycoparasitic potential.Significant difference was found between the mycelial growth of the isolates. TR03 and TR08 isolates had significant lower growth rate, than the other Trichoderma isolates. The TR07, TR09 and TR10 isolates grew rapidly in all examined temperatures.The interaction between Trichoderma harzianum and Fusarium oxysporum f. sp. ciceris showed different inhibition ratio on the fourth and sixth day. The TR08 - F. oxysporum had the highest inhibition zone (4 day: 63.6%; 6. day: 82.6%), while TR10 - Fusarium oxysporum had the lowest one (4. day: 40.9%; 6. day: 47.3%). The TR09 isolate showed low (<50%) inhibition zone at the examined days, while TR07, the most intensively growing isolate also had high (> 50%) inhibition rate.
