Megjegyzések:Epigenetic modifications play a vital role in the preservation of genome integrity and in the regulation of gene expression. DNA methylation, one of the key mechanisms of epigenetic control, impacts growth, development, stress response and adaptability of all organisms, including plants. The detection of DNA methylation marks is crucial for understanding the mechanisms underlying these processes and for developing strategies to improve productivity and stress resistance of crop plants. There are different methods for detecting plant DNA methylation, such as bisulfite sequencing, methylation-sensitive amplified polymorphism, genome-wide DNA methylation analysis, methylated DNA immunoprecipitation sequencing, reduced representation bisulfite sequencing, MS and immuno-based techniques. These profiling approaches vary in many aspects, including DNA input, resolution, genomic region coverage, and bioinformatics analysis. Selecting an appropriate methylation screening approach requires an understanding of all these techniques. This review provides an overview of DNA methylation profiling methods in crop plants, along with comparisons of the efficacy of these techniques between model and crop plants. The strengths and limitations of each methodological approach are outlined, and the importance of considering both technical and biological factors are highlighted. Additionally, methods for modulating DNA methylation in model and crop species are presented. Overall, this review will assist scientists in making informed decisions when selecting an appropriate DNA methylation profiling method. Introduction

Megjegyzések:Callus was induced and plants were regenerated for Cuscuta trifolii, the obligatory parasite of Medicago sativa. Callus was initiated on young seedlings grown in liquid culture medium consisting of Murashige and Skoog basal salts, Gamborg's vitamins (MSB5), 5 mg/l kinetin and 0.75 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D). The structure of callus tissue depended on the carbohydrates supplied, Friable, fast growing calluses were obtained in the presence of maltose and sorbitol combined (1.5-1.5%) and was maintained on corresponding agar-solidified medium for over a year. Plants were regenerated from calluses grown on solidified MSB5 medium that either lacked growth regulators or contained 2 mg/l kinetin. Embryo-like forms developed continuously from this culture for over 5 months. The detached embryo-like structures were placed in liquid MSB5 medium containing 2 mg/l kinetin and they grew as seedlings would under the same conditions. Both embryo-like structures and seedlings developed aggregate structures from which shoots grew. In this way, Cuscuta plants could be maintained in vitro without the presence of a host.

Megjegyzések:A greenhouse experiment was conducted on a calcareous chernozem soil to investigate the effects of nitrogen (N) and sulfur (S) fertilization and their ratios on the yield and quality of white mustard (Sinapis alba L.). Four levels of N [0.5, 1, 1.5, and 2 g pot21 N as ammonium nitrate (NH4NO3) and ammonium phosphate (NH4H2PO4)] in combination with three levels of applied N?S ratios [8, 4, 2; S as potassium sulfate (K2SO4) and sodium sulphate (Na2SO4)] were tested as treatments. Results indicated that a significant response to seed yield was observed for N and S application. Maximum yield, 24.8 g pot21 , was found when full doses of N and S were applied. Increasing N supply from 0.5 g pot21 to 1 g pot21 had little increasing effect in the oil content of the seed. Higher application of N doses (.1 g pot21 ) decreased these values significantly. The maximum oil content (28%) was achieved with the 1 g N pot21 treatment; the lowest values were observed in the pots applied with the highest N doses. Changes in the N content of the seed and straw showed a statistically significant increase with increasing N and S fertilization. Highest values in the seed and straw (5.96% and 0.87%, respectively) were observed by applying highest N and S doses. Seed and straw S levels were also observed to increase with increasing N rates and decreasing N?S ratio. Nitrogen doses significantly improved the quantities of essential amino acids with the exception of threonine, tyrosine, phenylalanine, and methionine. The amounts of these amino acids decreased with increasing N supply. When the N?S ratio decreased by increasing S, the quantities of valine, isoleucine, leucine, lysine, methionine, and cysteine increased significantly and the amount of tyrosine decreased. The quantities of nonessential amino acids with the exception of proline, histidine, and glycine increased with increasing N doses. In contrast, N rates decreased the amount of glycine. Decreasing the N?S ratio lowered the proline and arginine contents. The total amounts of essential amino acids slightly increased with increasing N rates and decreasing N?S ratio.

Megjegyzések:Calcium chloride-extractable (1:10 soil/solution ratio, w/v) nitrate nitrogen (NO3-N), ammonium nitrogen (NH4-N), organic nitrogen (EON), and Ntotal fractions were measured in two long-term experiments. Both experiments are located in the Great Hungarian Plain. The first experiment was established on a Luvic Phaeozem soil and the second experiment was on a Calcic Chernozem soil. Researchers investigated the effects of long-term fertilization and irrigation on the amounts of nitrogen (N) fractions. Nitrogen rates caused significant increase in NO3-N and Ntotal fractions. These fractions were in close correlation (r = 0.87; r = 0.88) with nitrogen balance. The nitrate content of the 200-cm-deep soil layer of soils under different nitrogen- and water-supply conditions were also studied. As an effect of N overfertilization, the maximum nitrate accumulation on nonirrigated plots occurred in the 200-cm depth, whereas on irrigated plots a nitrate accumulation layer could not be observed in the 200-cm-deep soil layer

Megjegyzések:Recent evidence sheds light on the peculiar type of plant intelligence. Plants have developed complex molecular networks that allow them to remember, optionally choose and make decisions depending on the stress stimulus, although they lack a nervous system. Being sessile, plants can exploit these networks to optimize their resources cost-effectively and maximize their fitness in response to multiple environmental stresses. Even more interesting, is the capability to transmit this experience to the next generation(s) through epigenetic modifications that add to the classical genetic inheritance. In this opinion article, we present concepts and perspectives regarding the capabilities of plants to sense, perceive, remember, re-elaborate, respond and to some extent transmit to their progeny information to adapt more efficiently to climate change.

Megjegyzések:Sulphur is an essential element for both plants and animals. Decreasing sulphur deposition from the air, and the use of more concentrated phosphate fertilizers that contain less sulphur, has led to reports of sulphur deficiencies for winter wheat. Sulphur deficiency significantly effects production and quality of winter wheat. Sulphur deficiency decreases grain size and baking quality because of formation of disulfide bonds formed from the sulphydryl groups of cysteine. This effects the viscoelasticity of dough. The purpose of this study was to evaluate the effect of NPK fertilization on sulphur concentration in winter wheat grain on different soil types. We analyzed wheat grain, grown at various locations where fertiliser had been applied for many years in places throughout Hungary, for sulphur content. These experiments were conducted using the identical varieties and fertiliser treatments at eight locations. At a separate location, additional varieties were also evaluated. The sulphur contents of the grains were determined by using ICP-AES equipment. The sulphur contents of winter wheat varieties in Hungary averaged 1500 mg/kg and the difference between the varieties was as much as 120 mg/kg. The sulphur contents are influenced not only by sulphur fertilisers, but nitrogen fertilisers as well. Due to the strong N-S relationships, fluctuations in protein contents also caused decreases/increases in the sulphur contents of the wheat grain. The N-S relationship could be described by using a mathematical linear function. Annual applications of 100 kg/ha superphosphate, containing more than 10 kg sulphur, provide sufficient sulphur to produce a 5 t/ha grain yield. This study suggests that a 1500 mg/kg value for sulphur content in winter wheat grain is sufficient to maintain good yield and grain quality.

Megjegyzések:Nowadays, there is increasing interest in environmentally friendly growing systems, and attention has also been paid to the environmental impacts of natural and artificial fertilizations. However, seasonal dynamics of macronutrient status needs further investigations in environmentally friendly apple orchards. Therefore, this study was aimed to determine nitrogen, phosphorus, and potassium contents of leaves in every month during the season and of soil in April and October at Debrecen?Pallag, Hungary, from 2002 to 2004. The study was performed in integrated and organic apple orchards on three apple cultivars (Mutsu, Elstar, and Reka). All easily soluble (0.01 M CaCl2) and plant?available nitrogen fractions (nitrate nitrogen, ammonium nitrogen, and total soluble nitrogen) in the soil were significantly higher (P = 0.05) in the integrated apple orchard compared to the organic one. The 0.01 M calcium chloride (CaCl2) soluble ortho?phosphate and total phosphate fractions of the soil were significantly higher (P = 0.05), whereas organic phosphate and soil pH showed no significant difference in the integrated apple orchard compared to the organic one. Potassium contents of the integrated soils were significantly higher (P = 0.05) compared to those of the organic one. Nitrogen, phosphorus, and potassium contents of leaves were higher in the integrated orchard compared to the organic one. The dynamics of P uptake corresponded to the phenological phases of apple. Potassium content of apple leaves decreased continuously until July, after that it slightly increased, and then it decreased again. In conclusion, results suggest that better mobility of artificial fertilizers in integrated orchards provided higher macronutrient uptake than in organic orchards, where only natural nutrient sources are allowed.

Megjegyzések:The production/consumption of greenhouse gases (GHG) in soils are of great importance in global warming, but the involved soil physico-chemical and biological characteristics affecting GHG production and consumption potentials are poorly understood in different land-use types. Carbon dioxide (CO2), nitrous oxide (N2O) and methane (CH4) production/consumption potentials from four land-use types and 10 soil types in central Japan and eastern Hungary, and their relationships with soil characteristics, were investigated. The average of CO2 production in Japanese soils was significantly higher than that of Hungarian soils due to the relatively higher microbial biomass carbon (MBC) content. N2O production from both countries' soils did not exhibit a significant difference. Most soils except Japanese paddy and soybean soils showed the potentials for CH4 consumption. Forest and grassland soils had relatively higher CO2 and N2O production than orchard and cropland soils for both countries. From regression analyses, it could be concluded that soil total nitrogen (TN) and ammonium-nitrogen (NH4+-N) account for 40.8% and 25.5% variations of the soils' CO2 and N2O productions, respectively. The CH4 consumption was positively correlated with soil carbon/nitrogen (C/N) ratio, while soil MBC availability could account for 15% variation of CH4 consumption under aerobic conditions.
