Megjegyzések:The relaxivity of Gd(HP-DO3A) was studied as a function of pH and buffer composition in order to identify the main factors of the observed relaxation enhancement due to the exchange of the coordinated hydroxyl proton. It was established that the paramagnetic relaxation time, T1M, of the coordinated hydroxyl proton is about 50% shorter than that of the protons in the coordinated water molecule. The control of the pK of the coordinated alcoholic ?OH moiety in the ligand is fundamental to utilize the proton exchange enhanced relaxivity under physio/pathologic conditions. A new derivative of Gd(HP-DO3A) was synthesized by replacing the ?CH3 group with a ?CF3 moiety. In this complex, the ?OH group becomes more acidic. Consequently, the maximum contribution of the proton exchange to the relaxivity is shifted to a lower pH region with the fluorinated ligand.

Megjegyzések:The chemistry of polyamino carboxylates and their use as ligands for Ln3+ ions is of considerable interest from the point of view of the development of new imaging agents. Of particular interest is the chemistry of the macrocyclic ligand 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) and its derivatives. Herein we report that the tetramethylated DOTA derivative, DOTMA, possess several properties that, from an imaging agent development point of view, are more advantageous than those of the parent DOTA. In particular, the Ln3+ chelates of DOTMA exhibit a marked preference for the monocapped twisted square antiprismatic coordination isomer which imparts more rapid water exchange kinetics on the chelates; ?M298 was determined to be 85 ns for GdDOTMA. Differential analysis of the 17O R2? temperature profiles of both GdDOTA and GdDOTMA afforded the ?M298 values for the square (SAP) and twisted square antiprismatic (TSAP) isomers of each chelate that were almost identical: 365 ns (SAP) and 52 ns (TSAP). The origin of this accelerated water exchange in the TSAP isomer appears to be the slightly longer Gd?OH2 bond distance (2.50 Å) that is observed in the crystal structure of GdDOTMA which crystallizes in the P2 space group as a TSAP isomer. The Ln3+ chelates of DOTMA also exhibit high thermodynamic stabilities ranging from log KML = 20.5 for CeDOTMA, 23.5 for EuDOTMA and YbDOTMA comparable to, but a shade lower than, those of DOTA.

Megjegyzések:An advanced experimental protocol is reported for studying the kinetics and mechanism of the complex redox reaction between chlorite ion and hypochlorous acid under acidic condition. The formation of ClO2 is followed directly by the classical two-component stopped-flow method. In sequential stopped-flow experiments, the target reaction is chemically quenched using NaI solution and the concentration of each reactant and product is monitored as a function of time by utilizing the principles of kinetic discrimination. Thus, in contrast to earlier studies, not only the formation of one of the products but the decay of the reactants was also directly followed. This approach provides a firm basis for postulating a detailed mechanism for the interpretation of the experimental results under a variety of conditions. The intimate details of the reaction are explored by simultaneously fitting 78 kinetic traces, i.e., the concentration vs. time profiles of ClO2 ?, HOCl, and ClO2, to an 11-step kinetic model. The most important reaction steps were identified, and it was shown that two reactive intermediates have a pivotal role in the mechanism. While chlorate ion predominantly forms via the reaction of Cl2O, chlorine dioxide is exclusively produced in reaction steps involving Cl2O2. This study leads to clear conclusions on how to control the stoichiometry of the reaction and achieve optimum conditions to produce chlorine dioxide and to reduce the formation of the toxic chlorate ion in practical applications.

Megjegyzések:We have synthesized and structurally characterized the first discrete thallium-containing polyoxometalate, [Tl2{B-β-SiW8O30(OH)}2]12- (1). Polyanion 1 was characterized in the solid-state and shown to be solution-stable by 203/205Tl NMR, electrospray ionization mass spectrometry, and electrochemical studies. The antibacterial activity of 1 was also investigated.

Megjegyzések:Radiometals play an important role in the diagnosis of physiological processes in vivo by means of imaging technologies, but also increasingly contribute to the treatment of diseases. The by far dominant way to apply the advantages of nuclear transformations of radiometals is the use of radiometal-ligand complexes. In this context, it turns out that DOTA (and its derivatives) currently is the most potent chelator. This article reviews the physico-chemical properties of DOTA and its [*M(DOTA)](-) complexes, which made the DOTA moiety the dominant chelator for theranostics in radiopharmaceutical chemistry and nuclear medicine. Interestingly, and beyond nuclear medicine applications, the physico-chemical properties of [*M(DOTA)] complexes open access to unique radiochemical separation strategies, which for decades appeared to be impossible not only within conventional chemistry, but also within modern radiochemistry. We will discuss three groups of such separations: i) the radiochemical separation of two chemically very similar elements such as two neighbor lanthanides, ii) the radiochemical separation of two isotopes of the same element, and even iii) the radiochemical separation of two nuclear levels of one and the same isotope.

Megjegyzések:A complete potentiometric and NMR relaxometric solution study on the heptadentate 2,2·,2ı,2·ı-[(6-piperidinyl-1,3,5-triazine-2,4-diyl)dihydrazin-2-yl-1-ylidene]tetraacetic acid (PTDITA) ligand has been carried out. This ligand is based on the 1,3,5-triazine ring with two hydrazine-N,N-diacetate groups in positions 2 and 4 and a piperidine moiety in position 6. The introduction of the triazine ring into the ligand backbone is expected to modify its flexibility and then to affect the stability of the corresponding complexes with transition-metal and lanthanide ions. Thermodynamic stabilities have been determined by pH potentiometry, UV spectrophotometry, and 1H NMR spectroscopy for formation of the complexes with Mg2+, Ca2+, Cu2+, Zn2+, La3+, Gd3+, and Lu3+ ions. PTDITA shows a good binding affinity for Gd3+ (logK = 18.49, pGd = 18.6) and an optimal selectivity for Gd3+ over the endogenous Ca2+, Zn2+, and Cu2+ (Ksel = 6.78 ? 107), which is 3 orders of magnitude higher that that reported for Gd(DTPA) (Ksel = 2.85 ? 104). This is mainly due to the lower stability of the CuII- and ZnII(PTDITA) complexes compared to the corresponding DTPA complexes, which suggests an important role of the triazine ring on the selectivity for the Gd3+ ion. The relaxometric properties of Gd(PTDITA) have been investigated in aqueous solution by measuring the 1H relaxivity as a function of the pH, temperature, and magnetic field strength (nuclear magnetic relaxation dispersion profile). Variable-temperature 17O NMR data have provided direct information on the kinetic parameters for exchange of the coordinated water molecules. A simultaneous fit of the data suggests that the high relaxivity value (r1 = 10.2 mM?1 s?1) is a result of the presence of two inner-sphere water molecules along with the occurrence of relatively slow rotation and electronic relaxation. The water residence lifetime, 298?M = 299 ns, is quite comparable to that of clinically approved magnetic resonance imaging contrast agents. The displacement of the inner-sphere water molecules by bidentate endogeneous anions (citrate, phosphate, and carbonate) has also been evaluated by 1H relaxometry. In general, the binding interaction is markedly weak, and only in the case of citrate, a ca. 35% decrease in relaxivity was observed in the presence of 60 equiv of the anion. Phosphate and carbonate also interact with the paramagnetic ion, likely as monodentate ligands, but formation of the ternary complex is accompanied by a modest increase of r1 due to the contribution of second-sphere water molecules.

Megjegyzések:A detailed study of the structures, thermodynamic stabilities and kinetics of the dissociation of Ga3+, In3+ and Cu2+ complexes formed with the heptadentate ligand AAZTA is reported. The stability constants (logKML) of the AAZTA complexes of Ga3+, In3+ and Cu2+ are 22.18, 29.58 and 22.27, respectively, which suggests that the seven-membered-ring skeleton is suited to the accommodation of these metal ions. The solid-state structure of Cu(H2AAZTA)]?H2O shows a distorted octahedral coordination. The equatorial coordination sites of Cu2+ are occupied by one of the ring N atoms, a water O atom, one of the carboxylate O atoms and the N atom of the iminodiacetate moiety. The other ring N atom and thecarboxylate O atom of the iminodiacetate moiety coordinate to the Cu2+ in the axial positions. In the pH range 4.5?8.5, Ga3+ is present in the form of the highly stable [Ga(AAZTA)OH]2? (log ?GaLH?1 = 17.69) The exchange reactions of [Ga(AAZTA)OH]2? with Cu2+ and transferrin are very slow and mainly occur through the spontaneous dissociation of the complex close to physiological conditions. The half-life for the dissociation of [Ga(AAZTA)OH]2? is t1/2 = 23 h at pH = 7.5 and 25 ?C in 0.025 M NaHCO3 and 0.15 M NaCl. The high conditional stability, fast formation and sufficiently slow dissociation of [Ga(AAZTA)OH]2? represent promising properties for the complexation and diagnostic applications of radioactive Ga isotopes.

Megjegyzések:The stability and protonation constants of the complexes formed between the ligand BOPTA [H(5)BOPTA: 4-carboxy-5,8,11-tris(carboxymethyl)-1-phenyl-2-oxa-5,8,11-triazatrideca n-13-oic acid] and Gd3+, Cu2+, and Zn2+ have been determined by pH potentiometry (Gd3+, Zn2+) and spectrophotometry (Cu2+) at 25 degrees C and at constant ionic strength maintained by 0.15 M NaCl. For comparison, the stability and protonation constants of the complexes of Gd3+, Cu2+, and Zn2+ formed with DTPA and DTPA-BMA {H(5)DTPA: diethylenetriamine-N,N,N',N '',N ''-pentaacetic acid, H(3)DTPA-BMA: 2,2'-[(carboxymethyl)imino]bis[ethylenebis(methylcarbamoyl-methyl)imino] diacetic acid} have also been determined under similar conditions. The stability constants (log K-ML) of the complexes of BOPTA and DTPA are very similar, but in 0.15 m NaCl the protonation constants of the ligands (log K-j(H)) and the log K-ML values are lower than those obtained in 0.1 M KCl or Me4NCl by 0.3-0.9 log K units. The order of selectivity of the ligands for Gd3+ over Zn2+ is BOPTA > DTPA > DTPA-BMA. The complex [Cu(DTPA-BMA)](-) deprotonates in the pH range 7-10 with the dissociation of an amide NH group and the coordination of the amide N atom. The kinetics of the exchange/transmetallation reactions between the complex [Gd(BOPTA)](2-) and the metal ions Cu2+, Zn2+, and Eu3+ have been studied by spectrophotometry (Cu2+, Eu3+) and relaxometry (Zn2+) in the pH range 3.3-6 at 25 degrees C in 0.15 m NaCl. The reactions with Cu2+ and Zn2+ occur predominantly with direct attack of the metal ions on the [Gd(BOPTA)](2-) complex. The kinetic activity of Eu3+ is lower, and in the exchange reactions with Eu3+ the proton-assisted dissociation of [Gd(BOPTA)](2-) (which is followed by fast reaction between the free ligand and Eu3+) could be also investigated. The rate constants, characterizing the proton-assisted dissociation of [Gd(BOPTA)](2-) and the exchange reactions occurring with the direct attack of Cu2+, Zn2+, and Eu3+ on the complex, are lower by about 30-90% than the rate constants obtained earlier for similar transmetallation reactions of [Gd(DTPA)](2-). The half-time for the dissociation of [Gd(BOPTA)](2-) at pH 7.4 and at 25 degrees C in the presence of 1 x 10(-5) m Zn2+ and 1 x 10(-5) M Cu2+ is 169 h.
