The mesencephalic locomotor region (MLR) is a functionally-defined area located in the midbrain that serves as an interface between higher-order motor systems (e.g. basal ganglia and cortex) and lower motor neurons (brainstem and spinal cord). The excitatory module of the MLR is composed of the pedunculopontine nucleus (PPN) and the cuneiform nucleus (CnF) and their activation has been proposed to elicit different modalities of movement, but whether differences in connectivity and physiological properties explain their contributions to motor activity is not known. Here we report that CnF-glutamatergic neurons are functionally homogeneous and have short-range axonal projections, whereas PPN-glutamatergic neurons are heterogeneous and maintain long-range connections with other motor circuits, most notably the basal ganglia. Optogenetic activation of CnF-neurons produced short-lasting muscle activation and fast-onset, involuntary motor activity, whereas activation of PPN-neurons produced long-lasting muscle activation, decreased locomotion and disrupted gait. Our results thus reveal a differential contribution to motor behavior in the structures that compose the MLR.

Precise control of protein turnover is essential for cellular homeostasis. The ubiquitin-proteasome system is well established as a major regulator of protein degradation, but an understanding of how inherent structural features influence the lifetimes of proteins is lacking. We report that yeast, mouse, and human proteins with terminal or internal intrinsically disordered segments have significantly shorter half-lives than proteins without these features. The lengths of the disordered segments that affect protein half-life are compatible with the structure of the proteasome. Divergence in terminal and internal disordered segments in yeast proteins originating from gene duplication leads to significantly altered half-life. Many paralogs that are affected by such changes participate in signaling, where altered protein half-life will directly impact cellular processes and function. Thus, natural variation in the length and position of disordered segments may affect protein half-life and could serve as an underappreciated source of genetic variation with important phenotypic consequences.

Osteoclasts play a central role in cancer-cell-induced osteolysis, but the molecular mechanisms of osteoclast activation during bone metastasis formation are incompletely understood. By performing RNA sequencing on a mouse breast carcinoma cell line with higher bone-metastatic potential, here we identify the enzyme CYP11A1 strongly upregulated in osteotropic tumor cells. Genetic deletion of Cyp11a1 in tumor cells leads to a decreased number of bone metastases but does not alter primary tumor growth and lung metastasis formation in mice. The product of CYP11A1 activity, pregnenolone, increases the number and function of mouse and human osteoclasts in vitro but does not alter osteoclast-specific gene expression. Instead, tumor-derived pregnenolone strongly enhances the fusion of pre-osteoclasts via prolyl 4-hydroxylase subunit beta (P4HB), identified as a potential interaction partner of pregnenolone. Taken together, our results demonstrate that Cyp11a1-expressing tumor cells produce pregnenolone, which is capable of promoting bone metastasis formation and osteoclast development via P4HB.
