Megjegyzések:Subclasses of cells in different compartments of the immune system possesses all those attributes, that make them suitable though somewhat limited models for the investigation of cellular processes during aging. Blood samples provide relative easily high amount of cells belonging to the same subclass, all of them having complex cascade processes in their signal transduction mechanisms, therefore being excellent targets for such investigations. One such subclass comprises peripheral blood lymphocytes. The signal-transduction cascade across the plasma membrane of lymphocytes displays many of the general features enabling us to draw conclusions for other cellular signaling problems that may arise during aging in other cell types not directly related to the immune system. The advantage of this approach lies in the fact that sometimes it is extremely difficult to study signal transduction processes in certain cell types under physiological conditions. The simultaneous occurrence of physical, chemical and molecular biological regulation of the immune processes at cellular and network levels make them very good examples for focusing our interest also on similar processes in other systems and cells. The fast developing new measuring techniques and the rapidly accumulating experimental data make it relatively easy to provide interesting new aspects, and ideas in this field. Finally, the immune system itself has its great importance and after all, it has an obvious declination with aging, the immune-senescence.

Megjegyzések:Gerontological research has some past and sporadically also some highlights in Hungary, but its present state can be easily deduced from the following data. During the last 12 years and more, well over 10,000 Hungarian scientific papers have been published in well-recognized national or international journals. Altogether approximately 1% of them have been classified as gerontological publications from Hungary. This low figure shows that gerontology has low priority and--unfortunately low support--in Hungary. This statement does not intend to downgrade Hungarian gerontologists, however points out that the Hungarian trends are not far from those of European or world wide interest in aging. Despite the recognition that we have to accept the inevitable fact that industrial societies will have (they already have) an aging population with all the social and medical problems arising, the focus of interest is wide from this significant and interesting (sub)population, which is neglected (sometimes even despised); yet everybody is absolutely eager to join this club. The average of the Hungarian research achievements and publication activities are among the better European achievements. There are some highlights and new trends even initiated by some outstanding Hungarian scientists, yet the overall weight of gerontology research is still an orphan in the Hungarian scientific life. We deal in this short and far from complete summary almost exclusively with experimental gerontology. We have to apologize if we have not included everybody, who also contributed even significantly to this field because the time for the preparation of this overview was short.

Megjegyzések:Paraoxonase (PON1) is principally complexed to HDL and is responsible, at least in part, for its antioxidant properties. PON1 activity decreases in several pathologies associated with atherosclerosis. The aim of this study was to investigate the PON1 activity and factors influencing its activity as a function of age. One hundred and twenty nine healthy subjects aged between 22 and 89 years were recruited for the study. We found that serum PON1 activity significantly decreased with age (r=-0.38, p<0.0001) while its arylesterase activity as well as its concentration in the serum did not change significantly. HDL concentrations remained unchanged with age, however, Apo A1 concentration showed a slight negative but significant correlation with age (r=-0.19, p<0.027). Moreover, the total cholesterol concentration was positively and significantly correlated with age (r=0.40, p<0.001). Thus, our results suggest that the decrease in PON1 activity cannot be explained by the decrease in Apo A1 concentrations with age. HDL from elderly subjects was more susceptible to oxidation than HDL from young subjects measured by higher lipid peroxidation rate. Thus, the decrease in PON1 activity may contribute to this increased susceptibility of HDL to oxidation with aging. Altogether our results suggest that the decrease in PON1 activity may be related to the development of oxidative stress conditions with aging and the increased HDL susceptibility to oxidation in elderly subjects.
