Colostrinin (TM) (CLN), isolated from mothers' pre-milk fluid (colostrum), is a uniform mixture of low-molecular-weight, proline-rich polypeptides. CLN induces neurite outgrowth of pheochromocytoma cells, extends the lifespan of diploid fibroblast cells, inhibits beta-amyloid-induced apoptosis and improves cognitive functions when administered to Alzheimer's disease patients. Objective: The aim of this study was to investigate potential allergic responses to CLN and its impact on allergic sensitization and inflammation caused by common allergens. Methods: We used a well-characterized mouse model of allergic airway inflammation. Changes in IgE/IgG1 and mucin levels, airway eosinophilia and hyperreactivity to methacholine were determined by ELISA, differential cell counting and whole-body plethysmography, respectively. Results: CLN did not increase IgE/IgG1 levels or induce cutaneous hypersensitivity reaction, airway inflammation and mucin production. Importantly, CLN significantly (p < 0.001) decreased IgE/IgG1 production, airway eosinophilia, mucin production and hypersensitivity induced by allergenic extracts from ragweed pollen grains and house dust mites. Conclusion: CLN itself is non-allergenic; however, it is effective in preventing allergic responses to known indoor and outdoor allergens. These data support the safe application of CLN and its potential use in the prevention of allergic inflammation in humans.

The aetiology of chronic urticaria is heterogeneous. Physical urticaria (PU) is estimated at around 35 %, autoimmune urticaria (AIU) at 25 % and chronic idiopathic urticaria (CIU) at 35% of all chronic urticaria cases. Methods: Differences in clinical and laboratory parameters among AIU, PU and CIU groups were examined. AIU was diagnosed if basophil CD63 assay was positive. Demographic data, severity of symptoms, and association with allergic and autoimmune diseases were analysed by the aid of an internationally accepted questionnaire a questionnarie. Immunoassays were carried out and the effectiveness of therapy was also investigated. Results: Female dominance in the AIU group was prominent. Concerning the urticaria score, there were significant differences between the AIU and PU patients (p=0.015) and between AIU and CIU cases (p=0.013). Between the CIU and PU patients, we found an insignificant difference in the urticaria score (p=0.78). AIU was more frequently associated with autoimmune diseases in the personal (p<0.001), and with other types of urticaria in the family history (p<0.001). Also anti-thyroid antibodies were more frequently detected in the AIU group. Antihistamine therapy was less effective in the AIU group (12.8%) than in the PU (70.3%) and CIU groups (68.6 %), but there were no significant differences between the CIU and PU groups regarding the effectiveness of antihistamine therapy. Conclusion: The autoimmune subgroup represents the most severe form of chronic urticaria. On the other hand between the CIU and PU groups there were no significant differences in urticaria scores as well as in response to antihistamine therapy. Background: Interleukin (IL)-16 has been characterized as animmunomodulatory cytokine. Besides its chemotactic properties,IL-16 amplifies inflammatory processes and possessesimmunoregulatory functions. Our aim was to investigate theassociation between serum IL-16 levels and the degree of allergicsensitization in patients with atopic dermatitis (AD).Methods: The serum level of IL-16 was measured by immunoenzymaticassays. Eosinophil cell count, serum total andspecific IgE levels were assessed; prick tests were also carriedout. Based on specific IgE levels and prick tests, AD patientswere divided into sensitized and nonsensitized subgroups,and correlations among serum IL-16, total IgE levels and eosinophilcell counts were measured in the total patient groupand in subgroups. Results: In the total patient group, significantlyhigher levels of IL-16 were found in the sera of patientswith AD, compared to healthy individuals and patientswith psoriasis. A significant correlation was detected betweenserum levels of IL-16 and total IgE, total IgE andeosinophil counts, but not between IL-16 and eosinophils.

Carotenoids are important derivatives of the human diet and occur in high concentrations in the human organism. Various carotenoids are also present in human breast milk and are transferred to breast-fed children. The alternative to breastfeeding is supplementation with an infant milk formula, but these formulas contain only a limited variety of carotenoids. Our question is: 'What is the function of various carotenoids in human nutrition with a special emphasis on child development and the development of atopy?' In this review, the mechanisms of action of the most important non-pro-vitamin A and pro-vitamin A carotenoids: α-carotene, β-carotene, β-cryptoxanthin, lutein, zeaxanthin, lycopene and retinoids are discussed. In summary, the combination of carotenoids, especially lycopene, seems to be of great importance, and exclusive usage of β-carotene in infant formula may yield in an increased atopy prevalence mediated in various target organs like the skin, lungs and immune competent cells. We conclude that the determination of novel bioactive metabolites of various carotenoids, at various stages in different organs during atopy development, might be the key to understanding the potential importance of carotenoids on atopy development.
