Megjegyzések:IL-1? is a "master" cytokine regulating a wide variety of physiologic and immunologic processes. The most frequently studied models for NLRP3 inflammasome-mediated IL-1? production are the macrophages; however, depending on their microenvironment, they can develop into functionally different cells. Several protocols have been developed to model the diversity of these cells in vitro. Here, we report for the first time, to our knowledge, a comparative study about the dynamics and molecular mechanisms of NLRP3 inflammasome priming and activation in LPS-stimulated, human, monocyte-derived GM- or M-macrophages, differentiated in the presence of GM-CSF or M-CSF, respectively. Our results show that IL-1? production by LPS-stimulated M-macrophages is a rapid and short event that requires ATP supplementation and is attenuated, in part, by the presence of IL-10, which reduces Akt signaling. However, IL-1? production by GM-macrophages develops gradually, and these cells produce IL-1?, even in the absence of ATP supplementation, because of the constitutively active caspase-1 enzyme. We show that the membrane-bound ectonucleotidases have an important regulatory role on the IL-1? secretion in GM-macrophages. Furthermore, we provide evidence that adenosine treatment enhances LPS-primed IL-1? secretion by GM-macrophages, but not by M-macrophages. These results show that, because of the different activation status and expression levels of the NLRP3 inflammasome components, as well as the signaling activity of the pathways, the two subtypes of macrophages respond very differently to the same stimuli. For this reason, the molecular composition of the microenvironment that shapes macrophage development should be considered when research or therapeutic methods are planned to control IL-1? production.

Megjegyzések:Type I and III IFNs are crucial, soluble components of potent antiviral responses. It has been explored recently that mTOR is involved in the regulation of IFN-?/? production by pDCs, albeit its role in the induction of IFN responses in cDCs remained unrevealed. In this study, we demonstrate that the PI3K/mTOR pathway is indispensable for eliciting intact type I and III IFN responses in moDCs stimulated with polyI:C. The inhibition of mTOR functionality by rapamycin impairs the pIRF3 and also a few members of the MAPK family, suggesting that mTOR contributes to the activation of multiple signaling pathways in the presence of viral antigens. Furthermore, rapamycin-treated moDCs show decreased capacity to prime IFN-? secretion by naive CD8+ T-lymphocytes. As in moDCs, mTOR-mediated regulation is also essential for the production of type I and III IFNs in circulating CD1c+ DCs. To our best knowledge, these results demonstrate for the first time that mTOR has an impact on the functional activities of cDCs via modulating the outcome of IFN secretion.

Megjegyzések:Glucocorticoids are powerful anti-inflammatory compounds inhibiting inflammatory cell recruitment and production of pro-inflammatory cytokines. We have recently found that dendritic cells, the key-players of T cell priming and polarization respond to allogeneic apoptotic neutrophils with proinflammatory cytokine release and Th1 cell activation. Here we show that monocyte-derived human dendritic cells develop their capacity to engulf apoptotic cells by up-regulating a set of apopto-phagocytic genes. This gene expression pattern was reprogrammed when differentiation took place in the presence of the synthetic glucocorticoid dexamethasone (Dex) which increased the expression of phagocytosis receptors MERTK and CD14, the bridging molecule C1QA, DNASE2 and the adenosine A3 receptor (ADORA3). The increased phagocytosis was attenuated by the addition of ADORA3 antagonist and could not be observed when bone marrow derived dendritic cells of ADORA3 knockout mice were treated with Dex. The glucocorticoid treated human dendritic cells loaded with allogeneic apoptotic neutrophils secreted, in response to LPS and IFN, the inflammatory cytokine TNFα. Furthermore, the Dex treated dendritic cells could activate autologous T lymphocytes toward Th1 effector cells and this was enhanced by their exposure to allogeneic apoptotic neutrophils.

Megjegyzések:The alternatively activated macrophage phenotype induced by IL-10 is called M2c. Adenosine is an endogenous purine nucleoside that accumulates in the extracellular space in response to metabolic disturbances, hypoxia, inflammation, physical damage, or apoptosis. As adenosine is known to regulate classically activated M1 and IL4- and IL-13-activated M2a macrophages, the goal of the present study was to explore its effects on M2c macrophages. We found that adenosine augmented the IL-10-induced expression of TIMP-1 and arginase-1 by the mouse macrophage cell line RAW 264.7 and by mouse BMDMs. The effects of AR stimulation on IL-10-induced TIMP-1 or arginase-1 expression were lacking in A2BAR KO macrophages. The role of A2BAR on TIMP-1 production of RAW 264.7 cells was confirmed with specific agonist BAY606583 and antagonist PSB0788. AR stimulation augmented IL-10-induced STAT3 phosphorylation in macrophages, and pharmacological inhibition or silencing of STAT3 using siRNA reduced the stimulatory effect of AR stimulation on TIMP-1 production. In contrast to its stimulatory effect on IL-10-induced STAT3 activation, adenosine inhibited IL-6-induced STAT3 phosphorylation and SAA3 expression. In conclusion, adenosine enhances IL-10-induced STAT3 signaling and M2c macrophage activation.

Megjegyzések:Regulated production of ROS is mainly attributed to Nox family enzymes. In neutrophil granulocytes and macrophages, Nox2 has a crucial role in bacterial killing, and the absence of phagocytic ROS production leads to the development of CGD. Expression of Nox2 was also described in B lymphocytes, where the role of the enzyme is still poorly understood. Here, we show that peritoneal B cells, which were shown recently to possess phagocytic activity, have a high capacity to produce ROS in a Nox2-dependent manner. In phagocytosing B cells, intense intraphagosomal ROS production is detected. Finally, by studying 2 animal models of CGD, we demonstrate that phagocyte oxidase-deficient B cells have a reduced capacity to kill bacteria. Our observations extend the number of immune cell types that produce ROS to kill pathogens.? Society for Leukocyte Biology.

Megjegyzések:Adipose tissue macrophages (ATMs) play key roles in metabolic inflammation, insulin resistance, adipose tissue fibrosis, and immune disorders associated with obesity. Research on ATM biology has mostly been conducted in the setting of adult obesity, since adipocyte hypertrophy is associated with a significant increase in ATM number. Signals that control ATM activation toward a proinflammatory or a proresolving phenotype also determine the developmental program and lipid metabolism of adipocytes after birth. ATMs are present at birth and actively participate in the synthesis of mediators, which induce lipolysis, mitobiogenesis, and mitochondrial uncoupling in adipocytes. ATMs in the newborn and the infant promote a lipolytic and fatty acid oxidizing adipocyte phenotype, which is essential to support the lipid-fueled metabolism, to maintain nonshivering thermogenesis and counteract an excessive adipose tissue expansion. Since adipose tissue metabolism in the early postnatal life determines obesity status in adulthood, early-life ATM functions may have a life-long impact.

Megjegyzések:Cytosolic RIG-I-like helicases (RLR) are PRRs involved in type I IFN production and antiviral immunity. This study focuses to the comparison of the expression, function, and signaling cascades associated to RLR in the previously identified CD14(-)DC-SIGN(+)PPARγ(low)CD1a(+) and CD14(low)DC-SIGN(+)PPARγ(high)CD1a(-) human moDC subsets. Our results revealed that the expression of RLR genes and proteins as well as the activity of the coupled signaling pathways are significantly higher in the CD1a(+) subset than in its phenotypically and functionally distinct counterpart. Specific activation of RLR in moDCs by poly(I:C) or influenza virus was shown to induce the secretion of IFN-beta via IRF3, whereas induction of proinflammatory cytokine responses were predominantly controlled by TLR3. The requirement of RLR-mediated signaling in CD1a(+) moDCs for priming naïve CD8(+) T lymphocytes and inducing influenza virus-specific cellular immune responses was confirmed by RIG-I/MDA5 silencing, which abrogated these functions. Our results demonstrate the subset-specific activation of RLR and the underlying mechanisms behind its cytokine secretion profile and identify CD1a(+) moDCs as an inflammatory subset with specialized functional activities. We also provide evidence that this migratory DC subset can be detected in human tonsil and reactive LNs.
