Megjegyzések:Notch is a highly conserved transmembrane receptor that determines cell fate. Notch signaling denotes cleavageof the Notch intracellular domain, its translocation to the nucleus, and subsequent activation of targetgene transcription. Involvement of Notch signaling in several cancers is well known, but its role in melanomaremains poorly characterized. Here we show that the Notch1 pathway is activated in human melanoma.Blocking Notch signaling suppressed whereas constitutive activation of the Notch1 pathway enhanced primarymelanoma cell growth both in vitro and in vivo yet had little effect on metastatic melanoma cells. Activation ofNotch1 signaling enabled primary melanoma cells to gain metastatic capability. Furthermore, the oncogeniceffect of Notch1 on primary melanoma cells was mediated by ?-catenin, which was upregulated followingNotch1 activation. Inhibiting ?-catenin expression reversed Notch1-enhanced tumor growth and metastasis.Our data therefore suggest a ?-catenin?dependent, stage-specific role for Notch1 signaling in promoting the

Megjegyzések:Background: In the management of osteoporosis in addition to pharmaceutical therapy and diet, physiotherapy may also be included. Our research group developed a novel three-electrode iontophoretic apparatus involving the use of calcium- and phosphate-donating lattice suitable for local ion delivery into the bones. We performed in vivo studies on rats in order to compare the effects and efficacy of electrotherapy with that of systemic hormone replacement therapy.Materials and methods: Five months after the operation tibias of Sham and ovarectomized (OVX) Sprague-Dawley female rats were subjected to serial local iontophoretic treatment(IOP). Other OVX rats received systemic subcutaneous 17-beta-oestradiol therapy (E2,100ug/kg/day for 13 weeks) or were also treated with the combination of IOP and E2. Changes in bone density were detected for 90 days after IOP. At the end of the experiments, biomechanical studies were performed together with elemental analysis assessing the calcium and phosphate concentrations.Results: Serial iontophoretic treatment resulted in a longlasting increase in bone density in both the Sham and OVX animals. Similar changes could be detected after E2 monotherapy, further enhancement could be observed applyingE2 + IOP. The above improvement was also evident regardingthe biomechanical features of the tibias as reduced calcium and phosphate content of the bones caused by OVX could be reversed by IOP or E2 monotherapy, as well as combination therapy.Conclusions: Iontophoresis using calcium- and phosphate donating microparticles further enhances the efficacy of estrogen therapy by increasing bone density, restoring the calcium and phosphate balance and the biomechanical properties of the bone.

Megjegyzések:Hyper-IgM syndrome (HIGM) is a heterogeneous condition characterized by impaired Ig class-switch recombination (CSR). The molecular defects that have so far been associated with this syndrome - which affect the CD40 ligand in HIGM type 1 (HIGM1), CD40 in HIGM3, and activation-induced cytidine deaminase (AID) in HIGM2 - do not account for all cases. We investigated the clinical and immunological characteristics of 15 patients with an unidentified form of HIGM. Although the clinical manifestations were similar to those observed in HIGM2, these patients exhibited a slightly milder HIGM syndrome with residual IgG production. We found that B cell CSR was intrinsically impaired. However, the generation of somatic hypermutations was observed in the variable region of the Ig heavy chain gene, as in control B lymphocytes. In vitro studies showed that the molecular defect responsible for this new HIGM entity (HIGM4) occurs downstream of the AID activity, as the AID gene was induced normally and AID-induced DNA double-strand breaks in the switch micro region of the Ig heavy chain locus were detected during CSR as normal. Thus, HIGM4 is probably the consequence of a selective defect either in a CSR-specific factor of the DNA repair machinery or in survival signals delivered to switched B cells.

Megjegyzések:BACKGROUND: Osteoclasts play a critical role in bone resorption under basal conditions, but they also contribute to pathological bone loss during diseases including postmenopausal osteoporosis. Phospholipase C?2 (PLC?2) is an important signalling molecule in diverse haematopoietic lineages. Here, we tested the role of PLC?2 in basal and ovariectomy-induced bone resorption, as well as in in vitro osteoclast cultures using PLC?2-deficient (PLC?2(-/-) ) mice.MATERIALS AND METHODS: The trabecular architecture of long bone metaphyses was tested by micro-CT and histomorphometric analyses. Postmenopausal osteoporosis was modelled by surgical ovariectomy. Osteoclast development and function, gene expression and PLC?2 phosphorylation were tested on in vitro osteoclast and macrophage cultures.RESULTS: ? PLC?2(-/-) mice had significantly higher trabecular bone mass under basal conditions than wild-type mice. PLC?2 was required for in vitro development and resorptive function of osteoclasts, but not for upregulation of osteoclast-specific gene expression. PLC?2 was phosphorylated in a Src-family-dependent manner upon macrophage adhesion but not upon stimulation by M-CSF or RANKL. Surprisingly, ovariectomy-induced bone resorption in PLC?2(-/-) mice was similar to, or even more robust than, that in wild-type animals. CONCLUSIONS :Our results indicate that PLC?2 participates in bone resorption under basal conditions, likely because of its role in adhesion receptor signalling during osteoclast development. In contrast, PLC?2 does not appear to play a major role in ovariectomy-induced bone loss. These results suggest that basal and oestrogen deficiency-induced bone resorption utilizes different signalling pathways and that PLC?2 may not be a suitable therapeutic target in postmenopausal osteoporosis.

Megjegyzések:Morpholinosydnonimine, a nitric oxide (NO) donor, has been reported to inhibit insulin release in isolated pancreatic islets. We studied whether transdermal application of nitroglycerin, another NO donor widely used for angina prophylaxis, influenced glucose-stimulated insulin release in healthy, young, male volunteers.METHODS AND RESULTS:Oral glucose tolerance tests [(OGTT) 75 g glucose in 200 mL of water) were performed in the presence of placebo patches or nitroglycerin-releasing 'active' patches (approx. 0.4 mg hour-1 nitroglycerin) in the same patients with a 2-week intertest interval. Venous blood samples were taken before and 15, 30, 60, 90, 120 and 180 min after the glucose load and evaluated for plasma glucose level and immunoreactive insulin responses (radioimmunoassay). Glucose-stimulated maximum increase in plasma insulin immunoreactivity were 36.3 +/- 5 and 78.8 +/- 6.1 mU mL-1 (P < 0.05) in the presence of active and placebo patches, respectively. Nevertheless, both fasting and postload blood glucose levels were the same at either patch. Active patches significantly decreased blood pressure with a marginal increase in heart rate.CONCLUSION:We conclude that inhibition of glucose-stimulated insulin release by transdermal nitroglycerin without causing hyperglycaemia may serve as a novel component of the antianginal mechanism of action of nitrates.

Megjegyzések:The endocannabinoid system (ECS) regulates multiple physiological processes, including cutaneous cell growth and differentiation. Here, we explored the effects of the major nonpsychotropic phytocannabinoid of Cannabis sativa, (-)-cannabidiol (CBD), on human sebaceous gland function and determined that CBD behaves as a highly effective sebostatic agent. Administration of CBD to cultured human sebocytes and human skin organ culture inhibited the lipogenic actions of various compounds, including arachidonic acid and a combination of linoleic acid and testosterone, and suppressed sebocyte proliferation via the activation of transient receptor potential vanilloid-4 (TRPV4) ion channels. Activation of TRPV4 interfered with the prolipogenic ERK1/2 MAPK pathway and resulted in the downregulation of nuclear receptor interacting protein-1 (NRIP1), which influences glucose and lipid metabolism, thereby inhibiting sebocyte lipogenesis. CBD also exerted complex antiinflammatory actions that were coupled to A2a adenosine receptor-dependent upregulation of tribbles homolog 3 (TRIB3) and inhibition of the NF-KB signaling. Collectively, our findings suggest that, due to the combined lipostatic, antiproliferative, and antiinflammatory effects, CBD has potential as a promising therapeutic agent for the treatment of acne vulgaris.

Megjegyzések:This Review highlights selected frontiers in pruritus research and focuses on recently attained insights into the neurophysiological, neuroimmunological, and neuroendocrine mechanisms underlying skin-derived itch (pruritogenic pruritus), which may affect future antipruritic strategies. Special attention is paid to newly identified itch-specific neuronal pathways in the spinothalamic tract that are distinct from pain pathways and to CNS regions that process peripheral pruritogenic stimuli. In addition, the relation between itch and pain is discussed, with emphasis on how the intimate contacts between these closely related yet distinct sensory phenomena may be exploited therapeutically. Furthermore, newly identified or unduly neglected intracutaneous itch mediators (e.g., endovanilloids, proteases, cannabinoids, opioids, neurotrophins, and cytokines) and relevant receptors (e.g., vanilloid receptor channels and proteinase-activated, cannabinoid, opioid, cytokine, and new histamine receptors) are discussed. In summarizing promising new avenues for managing itch more effectively, we advocate therapeutic approaches that strive for the combination of peripherally active antiinflammatory agents with drugs that counteract chronic central itch sensitization.
