Megjegyzések:Grewia lasiocarpa E. Mey. Ex Harv., Malvaceae (forest raisin) is a tropical small tree or shrub valued for its ecological importance as well as its nutritional, antioxidant, antibacterial, and anti-cancer properties as well as its ecological and ornamental importance. Glandular and non-glandular trichomes are present on the fruits, stem bark and leaves of G. lasiocarpa and these trichomes are the first line of defense. They are important structures that plants use to combat biotic and abiotic stress. The development of G. lasiocarpa trichomes and the biomechanics of the exudates present in the glandular (capitate) trichome were investigated for the first time using advanced microscopy techniques [Scanning electron microscope (SEM) and Transmission electron microscope (TEM)]. The pressurized cuticular striations may play a role in the exudates' biomechanics, i.e., releasing secondary metabolites present in the capitate trichome, which was observed to be multidirectional. The presence of many glandular trichomes on a plant implies an increase in the amount of phytometabolites. A common precursor for the development of trichomes (non-glandular and glandular) was observed to be DNA synthesis associated with a periclinal cell division, thus the final fate of the cell is determined by cell cycle regulation, polarity, and expansion. The glandular trichomes of G. lasiocarpa are multicellular and polyglandular, while the non-glandular (glandless) trichomes are either single-celled or multicellular. Since, trichomes ♭house' phytocompounds of medicinal, nutritional, and agronomical benefits; the molecular and genetic study of the glandular trichomes of Grewia lasiocarpa will be beneficial to humanity.

Megjegyzések:Harsh climates, i.e., drought, extreme temperatures, and toxic gases, pose issues to agriculture by altering plants` growth and yield. Biotechnology with biochemical defense approaches is beneficial for generating new plants/varieties with extra resilience to adverse conditions. In response to stress, cultures show an enriched level of secondary metabolite synthesis. Here, an efficient in vitro propagation method using axillary shoot proliferation, along with callus formation, was established in Glycyrrhiza glabra L. The phytochemical composition of in vitro and in vivo grown tissues was analyzed using a gas chromatography-mass spectrometry (GC-MS) technique, and the biochemical attributes were measured and compared in different investigated tissues. Callus formation from root explants was achieved with a frequency of 88.89% on MS medium containing 2.0 mg/L BAP and 0.5 mg/L 2,4-D. Axillary shoot proliferation was obtained from dormant buds when cultured onto MS supplemented with BAP alone, or in combination with, IAA. The maximum shoot proliferation (94.44%) was recorded on MS with 1.0 mg/L BAP with an average shoot length of 10.5 cm. The regenerated shoots were subcultured and transferred to the root induction medium, supplemented with various concentrations of IAA/IBA, wherein 2.0 mg/L IBA resulted in the best rooting frequency (88.89%). The GC-MS-based phytocompounds analysis of the methanolic extracts of root-derived callus and in vivo- and in vitro- grown root tissues was conducted. These samples revealed the presence of more than 35 therapeutically important bioactive compounds, such as methylglabridin, sitosterol, lupeol, squalene, stearic acid, linoleic acid, etc. The biochemical parameters, like total phenolic content, flavonoid content, DPPH scavenging activity, superoxide dismutase, and peroxidase activity were also measured. All the biochemical attributes were found to be higher in in vitro derived roots than the callus and in vivo grown root (donor) samples. These findings demonstrated that callus (root derived) and in vitro roots are a stable and potent source of multiple phytocompounds, encompassing medical significance with wide applications. This study may serve as an alternative opportunity in the sustained and continuous synthesis of important compounds without harming natural vegetation and normal environment.

Megjegyzések:Barleria albostellata C.B. Clarke (Acanthaceae) is a plant native to South Africa and relatively few studies have been performed on it. Species in this genus are known for their ethnopharmacological and phyto-medicinal values. In this study, the total flavonoid and phenolic contents and the antioxidant and cytotoxic activities of hexane, chloroform, and methanol extracts were evaluated at five different concentrations (15, 30, 60, 120, and 240 mu g/mL). The antioxidant activity of the extracts of B. albostellata was assessed in vitro using the 2,2 '-diphenyl-1-picrylhydrazyl (DPPH) scavenging and ferric reducing antioxidant power (FRAP) assays, while the phenolic content was determined using a Folin-Ciocalteu assay. The extracts' cytotoxicity was established using a 3-[(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] (MTT) assay in human embryonic kidney (HEK293), cervical cancer (HeLa), and breast adenocarcinoma (MCF-7) cell lines. Methanolic leaf extracts had the highest total flavonoid content (42.39 +/- 1.14 mg GAE/g DW) compared to other solvents. Additionally, the total phenolic content was the greatest in the methanol leaf extract (6.05 +/- 0.09 GAE/g DW), followed by the methanol stem extracts (2.93 +/- 0.73 GAE/g DW). The methanolic leaf and stem extract concentrations needed for 50% inhibition (DPPH) were 16.95 mu g/mL and 14.27 mu g/mL, respectively, whereas for FRAP, the reducing powers of all extracts were considerably lower than the ascorbic acid standard. The IC50 values of extracts tested in the three cell lines were >63 mu g/mL. According to the findings of our study, the leaves and stems of B. albostellata are rich in several bioactive compounds that may be a possible source of natural antioxidants and may have the potential to treat certain diseases. The extraction of the bioactive compounds from the leaves and stems of B. albostellata using bioassay-guided fractionation and the assessment of their safety will be essential for further investigations into this species in the search of potential novel therapeutic drug leads. To the best of our knowledge, this is the first report of the cytotoxic activities of leaf and stem extracts of Barleria albostellata.

Megjegyzések:Cyphostemma hypoleucum (Harv.) Desc. ex Wild & R.B. Drumm is a perennial climber, indigenous to Southern Africa, and belongs to the Vitaceae. Although there have been many studies of Vitaceae micromorphology, only a few taxa have been described in detail. This study aimed to characterize the micro-morphology of the leaf indumentum and determining its possible functions. Stereo microscope, scanning electron microscope (SEM), and transmission electron microscope (TEM) were used to produce images. Micrographs of stereomicroscopy and SEM showed the presence of non-glandular trichomes. In addition, pearl glands were observed on the abaxial surface using a stereo microscope and SEM. These were characterized by a short stalk and a spherical- shaped head. The density of trichomes decreased on both surfaces of leaves as the leaf expanded. Idioblasts that contained raphide crystals were also detected in tissues. The results obtained from various microscopy techniques confirmed that non-glandular trichomes serve as the main external appendages of the leaves. Additionally, their functions may include serving as a mechanical barrier against environmental factors such as low humidity, intense light, elevated temperatures, as well as herbivory and insect oviposition. Our results may also be added to the existing body of knowledge with regard to microscopic research and taxonomic applications.

Megjegyzések:An effective and reproducible micropropagation protocol was developed for Centratherum punctatum Cass. Successful in vitro initiation of callus and subsequent plant regeneration were obtained on nodal explants cultured on MS medium supplemented with plant growth regulators (PGRs). The maximum frequency of callus formation (98.3%) was noted on MS containing 4.0 mg/L 6-Benzylaminopurine (BAP) and 3.5 mg/L Kinetin with a maximum callus weight of 2.02 g. The best shoot induction frequency (100%) with an average of 30.2 shoots per explant was achieved when 4.5 mg/L BAP and 4.0 mg/L Kinetin were added to the MS. The same PGR combination resulted in the best callus-mediated shoot formation (8.3 shoots/callus mass). The highest rhizogenic response (95.3%) with an average 26.1 roots per shoot and root length of 6.2 cm was obtained with 1.0 mg/L Indole-3-acetic acid (IAA)-supplemented MS medium. The gas chromatography-mass spectrometry (GC-MS) technique was applied in the present study to analyze the methanolic extracts of the leaf, callus, and root of regenerated C. punctatum shoots to detect the different phytochemical constituents. The leaf extract of the regenerated C. punctatum showed 37 phytocompounds; some important bioactive compounds were the Phytol,1,6-Octadien 3,5-Dimethyl-Cis, 4,8-Dimethylnona-3,8-dien-2-one, 2,6-Octadiene, Stigmasterol, Chondrillasterol, Lanosteryl acetate, etc. In the callus, the extract had a total of 57 phytocompounds; among them, the Stigmasterol, Guanosine, and Tri-decanoic acid were the major ones. In the root extract, the GC-MS revealed a low number of 23 phytocompounds, the important compounds of which were Stigmasterol, Trimethylsilyl (TMS) derivative, Chrysantenyl 2-methuylbutanoate, 4-tert-Butoxybutan-1-ol, etc. The order in terms of numbers of phytocompounds present in tissue sources are callus > leaf > root.
