Megjegyzések:The main goal of our research was to carry out a comprehensive study on the obligatorily myrmecophilous Maculinea alcon (Denis & Schiffermüller) whose taxonomy has remained highly debated despite the fact that Maculinea Van Eecke species are some of the most intensively studied insects in Europe. We aimed to elucidate the relationship between the different forms exploiting different ecological environments with specific regard to ♭Hirschke's rebeli' (M. rebeli (Hirschke)) from the high altitudes of the Styrian Alps which was studied in this respect for the first time. Molecular analyses including multiple gene sequences, microsatellites and allozymes completed with Wolbachia screening, together with geometric morphometry were applied on various sets of samples. Our results showed that ♭rebeli' from the type locality differed strongly from all other samples not only at the highly variable microsatellites, but also at standard genes and clearly represents a well?separated unit (potentially subspecies) within M. alcon. As all individuals were infected with a single Wolbachia strain, this is possibly an ancient infection with high maternal transmission rate. It also suggests that the separation of the topotypic populations was probably a relatively recent event which could have been promoted by genetic drift or specific conditions at high altitudes, and maintained by restricted gene flow. Both the genetic and the morphometric studies supported the proposition that the geographical origin of the populations has a higher effect on the structure of variation than the food plant use. Moreover, we detected more genetic lineages than could be defined by differential host plant usage.

Cím:Multilevel studies on the two phenological forms of Large Blue ( Maculinea arion ) (Lepidoptera: Lycaenidae) / Judit Bereczki, János P. Tóth, Gábor Sramkó, Zoltán Varga
Dátum:2014
ISSN:0947-5745
Megjegyzések:The main goal of our research was to study comprehensively the differences between the two phenological forms of the socially parasitic and globally threatened Large Blue (Maculinea arion)in the Carpathian Basin using four character sets (mitochondrial sequences, allozymes, male genitalia and wing morphometrics). Comparative analyses of distance matrices, phylogenetic trees and ordination patterns have been applied. The genetic and morphometric patterns revealed by our studies were disordant. While we experienced a significant differentiation between the 'spring' and 'summer type' of M. arion in both wing and genital traits, the two phenological forms did not show any genetic differentiation on two mitochondrial loci and in allozymes. At the same time, all individuals were infected by Wolbachia. Although certain wing traits may not represent reliable tracers of phylogeny because of the particular adaptive significance, the wing characteristics involved in our research are probably determined genetically. Additionally, the significant differentiation of male genitalia also indicates incipient prezygotic isolation arising from phenological differentiation between the 'spring' and summer arion'. It is possible that all extant differences between the two forms are attributable to (1) different host-ant use, (2) incipient speciation, (3) cytoplasmatic incompatibility (CI) by Wolbachia or the combination of these factors. In addition, discordant results indicate that the combined use of different approaches and data sets is strictly necessary to clarify systematic and evolutionary relationships.

Megjegyzések:Craniophora ligustri is the most widely distributed species within the genus Craniophora. We used geometric morphometry to assess the shape variation of male valvae in the Western Palaearctic and Russian Far East populations of C. ligustri including the subspecies C. l.carbolucana and C. l. hyrcanica. Outlines were transformed using Hangle Fourier method followed by shape analyses using multivariate statistics (CVA, UPGMA). Based on the results the Far Eastern sample is clearly separated from the Western Palaearctic populations. The West Palaearctic samples were split to two demes, a Northern and a Southern one. The probable refugia of the Southern deme may have extended from South Italy, the Balkan Peninsula across the Euxinic coast of Asia Minor to the Talysh and Northern Iran (Hyrcanian part of Elburs Mts), while the refugia of the Northern deme have been supposedly extra-Mediterranean.

Megjegyzések:Jelen dolgozatban a Mátra déli részének xerotherm tölgyeseiben végzett monitorozó jellegű lepkészeti kutatások eredményeit mutatjuk be. Vizsgálataink során néhány fokozottan védett, közösségi jelentőségű lepkefajnak jelentős egyedszámú állományát fedeztük fel; a magyar téliaraszolónak (Erannis ankeraria)európai jelentőségű állományai élnek a Mátra déli részének molyhos tölgyeseiben (Szurdokpüspöki, Kisnána). Eredményeink arra is rávilágítottak, hogy ezeken az élőhelyeken egy sor, a pontomediterrán faunakörhöz tartozó ritka, védelmet érdemlő molyhostölgyes és sztyep-sziklagyeplakó faj is megtalálható, melyek igen értékes lepkeközösséget alkotnak. Ezen közösségek élőhelyei jelenleg nem állnak védelem alat, fennmaradásukat több negatív hatás veszélyezteti. Hosszú távú megóvásuk csak fokozott területi védelemmel lehetséges. Ennek érdekében javasoljuk egy új Natura 2000 SCI terület kijelölését a Déli-Mátrában (Pálosvörösmarttól Kisnánáig).

Dátum:2016
ISSN:1439-6092
Megjegyzések:The pattern of genetic variation in a butterfly species depends on the past history of the given species and also on recent evolutionary processes affecting its populations. The aim of the present study was (i) to analyse the enzyme polymorphism in the Clouded Apollo populations of the Carpathian Basin to reveal the contemporary pattern of their genetic differentiation and (ii) to compare it with an expanded mitochondrial DNA (mtDNA) haplotype network of the SE European populations. Allozyme polymorphism was analysed in 22 populations of four geographic regions: Transdanubian (TM) and North Hungarian Mountains (NM), Körös (KÖR) and Bereg?Apuseni?East Carpathian regions (BEAC). The results of the Bayesian clustering analyses based on allozymes supported the presence of three main genetic lineages in the Carpathian Basin: One was typical for TM, another was characteristic for NM and the third cluster was predominant in KÖR. The populations of BEAC harboured a mixture of two clusters. The mtDNA haplotype network suggested a fairly similar distribution: The peri-Alpine clade together with the West Balkan clade was detected in TM, while the East Balkan clade occurred in NM, partly in TR and in the two eastern regions of the Basin (KÖR and BAEC). The incongruities between the results of the mtDNA and allozyme studies can be explained by the different timescales represented by the two markers. The mtDNA haplotype network provided strong evidence concerning the existence of two Balkan lineages, which probably formed a ♭zone of admixture' in the Transdanubian and North Hungarian Mountains. The possibility of Last Glacial survival of Parnassius mnemosyne in the Carpathian Basin and the conservation implications of these results are discussed.

Cím:Regional pattern of genetic variation in the Eastern Central European populations of Euphydryas maturna (Lepidoptera: Nymphalidae) / Katalin Pecsenye, Andrea Tóth, János P. Tóth, Judit Bereczki, Zoltán Varga
Dátum:2017
ISSN:1366-638X 1572-9753
Megjegyzések:The aim of this study was to delineate functional conservation units (FCUs) in Scarce Fritillary Euphydryas maturna (Linnaeus, 1758) populations in eastern Central Europe. We analyzed the level and structure of genetic variation using allozymes as markers in 26 Scarce Fritillary populations originating from nine geographic regions. Considering the reproductive strategy of E. maturna, we assumed that populations are strongly exposed to genetic drift. Accordingly, we hypothesized: (i) A low level of genetic variation within populations, and; (ii) A high level of differentiation among them with little evidence of geographic pattern. The results of this study mostly fulfilled our expectations: (i) Scarce Fritillary populations of eastern Central Europe exhibited a relatively low level of polymorphism at investigated enzyme loci, and; (ii) Comparatively strong differentiation was detected among populations. As opposed to our expectation, however, a more-or-less clear pattern of differentiation was revealed by the results of our Bayesian-clustering analysis. Four genetic regions were distinguished on the basis of their average cluster membership coefficients, specifically Dobrogea (Romania), North and East Hungary (composed of four geographic regions), Central and West Hungary (involving three geographic regions), and Transylvania. Three of these genetic regions were previously distinguished as separate subspecies, comprising E. maturna idunides (Central and West Hungary), E. maturna partiensis (North and East Hungary including Transylvania), and E. maturna opulenta (Dobrogea, Romania). These three putative subspecies can be considered as Scarce Fritillary FCUs in eastern Central Europe.

Megjegyzések:Since genetic variation is the basis of evolutionary potential of a species, its structure needs to be understood. Thus, the aim of this study was to analyze and contrast the structure of genetic and phenotypic variation in the Euphydryas aurinia populations of southeastern central Europe. Genetic variation was studied by two types of molecular genetic markers: mtDNA COI sequences and allozymes. As the great hiatus in the European distribution of E. aurinia is located in the central part of the Carpathian Basin, we expected that the populations East and West to this gap would be highly differentiated. Populations of Central Transdanubia actually represent the easternmost margin of the West European distribution of E. aurinia. In view of the peripheral position of these populations, we supposed to find some genetic sign of local adaptation, as a consequence of diversifying selection and an increased level of fluctuating asymmetry as a result of environmental stress. The analyses of the molecular genetic markers revealed a basic East-West differentiation among the populations of southeastern central Europe which was further structured in the western part of the study area. The results suggested that the genetic differentiation between the two western regions is probably the consequence of diversifying selection. The pattern of phenotypic differentiation among the western populations, however, was different. A geographic cline was revealed (decreasing wing size) toward the eastern margin of the distribution in parallel with increasing fluctuating asymmetry. The conservation inferences of the results are considered.

Cím:The genetic structure of phenologically differentiated Large Blue (Maculinea arion) populations (Lepidoptera: Lycaenidae) in the Carpathian Basin / Judit Bereczki, János P. Tóth, Andrea Tóth, Edit Bátori, Katalin Pecsenye, Zoltán Varga
Dátum:2011
Megjegyzések:The infraspecific taxonomy of the European populations of the Large Blue (Maculinea arion) is confusing. Several subspecies have been described mostly based on external morphological features. In the Carpathian Basin two subspecies have been distinguished. Maculinea arion arion flies from mid-May to mid-June and Maculinea arion ligurica is on the wing from the end of June to mid-August. The two forms show some differentiation in habitat use, but occasionally can also share habitats with two peaks in the appearance of butterflies. Our aim was to study the level and structure of genetic variation in a set of populations of the two phenologically different M. arion. Imagos were collected from 8 localities between 2000 and 2006. Enzyme polymorphism was analysed at 13 enzyme loci using polyacrylamide gel electrophoresis. In the analysis of the data, we estimated the parameters of polymorphism. To study the pattern of genetic differentiation F-statistics, hierarchical F-statistics and AMOVA were computed. GeneClass and Structure were both applied to analyse the differentiation between the two phenologically different sets of populations. Cavalli-Sforza and Edwards' arc distances were calculated and a UPGMA dendrogram was constructed on the basis of the distance matrix. PCA analysis was also carried out using the allele frequencies of the individuals. The level of polymorphism was relatively high in M. arion. The results of all analyses indicated that the differences between the two sets of phenologically different populations accounted for a low percentage of the total differentiation. In addition, a sizeable amount of variation could be attributed to the differences among the samples collected from the same population in consecutive years. Thus, we concluded that the "spring" and "summer arion" could not be considered as separate ESUs, although we could attribute conservation value to both forms on the basis of their phenological differentiation and habitat use.

Dátum:2019
ISSN:0307-6970
Megjegyzések:In this study we analysed for the first time the phylogenetic relationshipsamong the EuropeanLethrusspecies based on mitochondrial (COI, 16S) and nuclear(H3, wingless) markers. The phylogenetic inference based on these genes indicated thatthe EuropeanLethrusspecies form a monophyletic group with two main lineages. Oneof them is distributed in the central part of the Balkan Peninsula (North Macedonia, northwestern Greece and southwestern Bulgaria) and includes five species (L. perun,L. raymondi,L. strymonensis,L. halkidikiensisandL. elephas). Three species of theother clade (L. schaumii,L. aresandL. schneideri) occur in the eastern part of the BalkanPeninsula (southeastern Bulgaria and northeastern Greece) while its fourth member,L. apterus, could colonize much larger areas in central and eastern Europe. Despite itslarge geographic distribution,L. apterusproved to be a single species. By contrast, wefound strong intraspecific differentiation in bothL. schaumiiandL. halkidikiensisinspite of their restricted distribution. Surprisingly, two highly divergent mitogroups werealso detected within a singleL. schneideripopulation. As the microsatellite markersdid not show any differentiation between them, this finding is likely to be accountedfor by a unidirectional hybridization event. These findings highlight that phylogeneticstudies or species descriptions should not be based on mitochondrial markers alone. Ourstudy clearly demonstrates that allopatric speciation is the most important speciationmechanism in the European species of the genusLethrus, although we also hypothesizethat other well-known factors, such as hybridization or the glacial-interglacial dynamics, could significantly affect the speciation patterns of certain lineages.

Megjegyzések:Disjunct distribution patterns regularly resulted in the separation of different genetic lineages in glacial refugia. Recent patterns of survival and expansion have been often revealed by climatic niche modelling. We used the combination of genetic markers, geometric morphometry and climatic niche modelling to clear up the taxonomy and reconstruct the potential range of an endemic Iranian, taxonomically disputed Melitaea species in climatically different epochs. Our results show that this species (Melitaea abbas Gross and Ebert 1975, comb. n. = M. zagrosi T oth and Varga syn. nova) is clearly separated from all taxa of the Melitaea phoebe species group and only occurs in Iran and Azerbaijan but were also predicted for some adjacent regions. Molecular markers and distribution modelling show consistently that this species should have had a longterm survival in this area, and its range could have been slightly larger during the last glacial maximum than currently. Based on the studied molecular markers, three main groups in M. abbas can be recognized: those of steppic area of Azerbaijan, western Iran and northeastern Iran. Each group is characterized by own mitochondrial haplotypes, but also a high level of genetic diversity appears in the central part of the distribution area (Zagros Mts.).
