Megjegyzések:We isolated full-length cDNA clones for human matrilin-2, an oligomeric protein, which forms filamentous networks in the extracellular matrices of various tissues. The human matrilin-2 precursor is encoded by a 4.0-kb mRNA, it consists of 956 amino acids and shows 93% similarity to the mouse protein. Out of the two von Willebrand factor type A-like domains, the 10 epidermal growth factor-type modules, one unique sequence and the oligomerization module, the first A domain is the most conserved. RT-PCR demonstrated wide expression of the gene in human cell lines of fibroblastic or epithelial origin. Alternative splicing affected only 19 amino acids in a 75-moiety-long segment, unique to matrilin-2. Isolation and analysis of the 3' end of the gene revealed that the reason for alternative splicing is alternative 3' splice site selection. Further, we identified in the human matrilin-2 gene a U12 type AT-AC intron between the last two exons encoding the oligomerization domain. We mapped the matrilin-2 gene (MATN2) by fluorescence in situ hybridization at chromosome position 8q22.

Megjegyzések:The role of major cellular serine/threonine-specific protein phosphatases,protein phosphatase 1 and 2A, was investigated during chicken cartilagedifferentiation under in vitro conditions. Activity of protein phosphatase 2Adecreased parallel to differentiation of chondrogenic cells, whereas activity ofprotein phosphatase 1 remained unchanged as assayed in the supernatants of thehomogenised chicken limb bud micromass cell cultures. When okadaic acid, a potentinhibitor of protein phosphatase 1 and 2A was applied in 20 nM concentration for4 h during the second and third culturing days, it significantly increased thesize of metachromatic cartilage areas measured in 6-day-old colonies. Followingokadaic acid treatments, a significant inhibition in the activity of proteinphosphatase 2A was found, while the activity of protein phosphatase 1 wasunaffected as measured an days 2 and 3. TRITC-phalloidin labelling demonstratedthat okadaic acid disorganised actin filaments and induced rounding ofchondrogenic cells. This deterioration of actin filaments was reversible.Electron microscopy and biochemical analysis of colonies revealed that theultrastructure and major components of cartilage matrix remained unchanged underthe effect of okadaic acid. Okadaic acid-treatment applied to cultures containingpredominantly differentiated chondrocytes (after day 4) did not influence thecartilage formation. 3H-thymidine and bromodeoxyuridine incorporation-assaysdemonstrated enhanced cell proliferation in the okadaic acid-treated coloniescompared to that of the untreated ones. Our results indicate, for the first time,that protein phosphatase 2A is involved in the regulation of chondrogenesis.Inhibition of protein phosphatase 2A with okadaic acid may result in increasedchondrogenesis via modulation of proliferation and cytoskeletal organisation, aswell as via alteration of protein kinase A-signaling pathway of the chondrogeniccells.

Megjegyzések:aWe have evaluated the importance of the Ser/Thr protein phosphorylation anddephosphorylation for chondrogenesis in high-density chicken limb bud mesenchymalcell cultures (HDCs) by using H89, a cell-permeable protein kinase inhibitor, andokadaic acid (OA), a phosphoprotein phosphatase (PP)-specific inhibitor molecule.When 20 nM OA was applied to the HDCs on Days 2 and 3 of culturing, itsignificantly inhibited protein phosphatase 2A (PP2A), enhanced cartilageformation, and elevated the activity of cAMP-dependent protein kinase (PKA).Application of 20 microM H89 significantly decreased the activity of PKA andblocked the chondrogenesis in HDCs. Furthermore, OA enhanced cartilage formationand elevated the suppressed activity of PKA even in the H89-pretreated HDCs.cGMP-dependent protein kinase was not detected in HDCs, while protein kinase Cmu(PKCmu), which is also inhibited by nanomolar concentrations of H89, was presentthroughout the culturing period. Neither OA nor H89 influenced the expression ofthe catalytic subunit of PKA or the cAMP response element binding protein, CREB.However, a significantly elevated amount of Ser-133-phosphorylated-CREB (P-CREB)was detected following addition of OA, while H89 treatment resulted in a decreaseof the amount of P-CREB. Our results demonstrate that PP2A plays a role in theregulation of the PKA signaling pathway and that the phosphorylation level ofCREB is influenced by the activity of both enzymes during in vitrochondrogenesis.

Megjegyzések:Calcineurin was found as a positive regulator of chondrogenesis in chondrifying chicken micromass cultures (HDCs), as cyclosporine A (CsA) reduced both the amount of cartilage and the expression of mRNAs of aggrecan and the chondrogenic transcription factor Sox9. Cartilage formation was inhibited by H(2)O(2) in a concentration-dependent manner without loss of cellular viability or severe decrease of cell number. Expression of both the mRNA and the unphosphorylated protein Sox9 was decreased, while its phosphorylation was stimulated by either H(2)O(2) or CsA. Oxidative stress decreased the activity of calcineurin but the phosphorylation of the member of MAPK family ERK1/2 was extremely elevated either by 1 mM H(2)O(2) or 2 muM CSA. The ERK inhibitor PD098059 attenuated the depletion of cartilage matrix as well as decreased the expression and phosphorylation of Sox9 in cultures treated with H(2)O(2) or CsA. Our results suggest that the chondrogenesis-inhibiting effect of H(2)O(2) is mediated, at least partly, by inhibition of calcineurin and by activation of ERK1/2. We also propose a regulatory role of calcineurin in the phosphorylation level of either ERK1/2 or Sox9 and a positive role of ERK1/2 in regulating both the expression level and the phosphorylation state of Sox9 in chicken HDCs.

Megjegyzések:Oxidative stress has been implicated in the pathogenesisof various diseases affecting chondrogenesis or the function of articular cartilage. DNA damage caused byoxidative stress may trigger the activation of the nuclearenzyme, poly(ADP-ribose) polymerase-1 (PARP-1) whichmay contribute to tissue injury. We aimed at investigating the effects of peroxynitrite (100-600 ?M) and hydrogen peroxide (0.1-4 mM) on PARP activation and extracellular matrix production of high density micromass cultures (HDC)prepared from chick limb bud mesenchymal cells. We foundthat both oxidative species strongly inhibited matrix formation of HDCs treated on day 2 but not on day 5. The PARP inhibitor 3-aminobenzamide (3-AB) stimulated matrix production in non-stressed cells and prevented suppressed matrix production in oxidatively stressed cells. Both hydrogen peroxide and peroxynitrite induced PARP activation and poly(ADP-ribose) accumulation. Decreased proliferation, viability and NAD+ content were not or only slightly improved by 3-AB, indicating that 3-AB directly affects matrix formation. In conclusion, oxidative stress stimulates poly(ADP-ribose) metabolism and inhibits extracellular matrix production of HDCs in a PARP-dependent manner. Our findings may have implications for potential therapeutic approaches aimed at restoring the matrix production capacity of chondrogenic cells.
