Megjegyzések:Vitamin A plays an essential role in the maintenance of gut homeostasis but its interplay with chemokines has not been explored so far. Using an in vitro model system we studied the effects of human colonic epithelial cells (Caco2, HT-29, and HCT116) derived inflammatory stimuli on monocyte-derived dendritic cells and macrophages. Unstimulated Caco2 and HT-29 cells secreted CCL19, CCL21, and CCL22 chemokines, which could attract dendritic cells and macrophages and induced CCR7 receptor up-regulation by retinoic-acid resulting in dendritic cell migration. The chemokines Mk, CXCL16, and CXCL7 were secreted by all the 3 cell lines tested, and upon stimulation by IL-1?? or TNF-?? this effect was inhibited by ATRA but+had no impact on CXCL1, CXCL8, and CCL20 secretion in response to IL-1??. In the presence of ATRA the supernatants of these cells induced CD103 expression on monocyte- derived dendritic cells and when conditioned by ATRA and cocultured with CD4 T-lymphocytes they reduced the proportion of Th17 T-cells. However, in the macrophage-T-cell cocultures the number of these effector T-cells was increased. Thus cytokine- activated colonic epithelial cells trigger the secretion of distinct combinations of chemokines depending on the proinflammatory stimulus and are controlled by retinoic acid, which also governs dendritic cell and macrophage responses.

Megjegyzések:It has been demonstrated that pollen grains contain NAD(P)H oxidases that induce oxidative stress in the airways, and this oxidative insult is critical for the development of allergic inflammation in sensitized mice. On the basis of this observation, we have examined whether pollen grain exposure triggers oxidative stress in dendritic cells (DCs), altering their functions. To test this hypothesis, human monocyte-derived DCs were treated with ragweed pollen grains. Our findings show that exposure to pollen grains induces an increase in the intracellular levels of reactive oxygen species in DCs. Our data also indicate that besides the NAD(P)H oxidases, other component(s) of pollen grains contributes to this phenomenon. Elevated levels of intracellular reactive oxygen species triggered the production of IL-8 as well as proinflammatory cytokines, such as TNF-alpha and IL-6. Treatment with pollen grains initiated the maturation of DCs, strongly upregulated the membrane expression of CD80, CD86, CD83, and HLA-DR, and caused only a slight increase in the expression of CD40. The pollen-treated DCs induced the development of naive T lymphocytes toward effector T cells with a mixed profile of cytokine production. Antioxidant inhibited both the phenotypic and functional changes of DCs, underlining the importance of oxidative stress in these processes. Collectively, these data show that pollen exposure-induced oxidative stress may contribute to local innate immunity and participate in the initiation of adaptive immune responses to pollen Ags.

Megjegyzések:The release of different cytokines and mediators in tears of patients with keratoconus (KC) wearing contact lenses (CLs) may contribute to the pathology of KC. METHODS: Cohort study was established in patients with KC wearing rigid gas permeable (RGP) CL (group I), patients with ametropia wearing silicone hydrogel (Si-Hi) CL (group II) and ametropic patients wearing RGP CL (group III). RESULTS: Our findings indicate that before CL wear, the release of epidermal growth factor (EGF) and tissue-type plasminogen activator (t-PA) was attenuated, whereas matrix metalloproteinase (MMP)-9, interleukin (IL)-6, chemokine (C-C motif) ligand 5 (CCL5), IL-13, and plasminogen activator inhibitor (PAI)-1 were enhanced in KC compared with ametropes. An increasing linear trend over time was found for MMP-9, EGF, and CXCL8 in KC and MMP-9, MMP-13, IL-6, and CXCL8 in group III. Significant differences were observed in the linear trend over time between groups I and III for MMP-13 and tissue inhibitor of metalloproteinases (TIMP)-1; between groups I and II for MMP-9 and CXCL8; and between groups III and II for MMP-9, CXCL8, and MMP-13. In KC, the release of MMP-9 at week 6 and nerve growth factor (NGF) at 10 min was higher, but NGF at week 2 was lower than that in group II. The release of MMP-13 and NGF at week 2 and 6 were lower in the KC group as compared with group III, and similarly, with IL-6 and CXCL8 at week 2 and PAI at all time points. CONCLUSIONS: Contact lens wear can influence the levels and dynamics of various mediators in the tears of patients with KC that might have an impact on the progression of the disease.

Megjegyzések:Célkitűzés: Különböző citokinek könnybe történő szekréciójának összehasonlítása perforáló kontralaterális autokeratoplasztika (AKP) és allokeratoplasztika után. Betegek és módszerek: AKP után könnymintát gyűjtöttünk két betegtől. Műtét után 1 évvel az autograft az "A" beteg esetében tiszta, a "B" beteg esetében parciálisan dekompenzált volt. Kontrollként 10 beteg komplikációmentes allograftja szolgált. A könnymintákban áramlási citométerrel mértük meg az IL-1/ß, IL-6, IL-8, IL-10, IL-13, IL-17, IL-18, TNF-a, INF-y, IP-10 és RANTES koncentrációját. A kontrollok citokinadataira empirikus trendvonalat illesztettünk. Eredmények: Az "A" betegnél az IL-1/ß, IL-10, IL-13, IL-17 és INF-y koncentrációja a trendvonal alatt, a "B" betegnél a fölött volt. Az IL-6, IL-8 és RANTES koncentrációja az "A" betegnél többnyire a trendvonal alatt, a "B" betegnél többnyire a fölött volt. Az IP-10 koncentrációja az "A" betegnél a trendvonal fölött, a "B" betegnél az alatt volt. Következtetések: A citokinkoncentrációk komplikációmentes AKP esetében jelentősen különböznek az allokeratoplasztikán átesett kontrollokétól. Komplikált AKP esetében azonban az allokeratoplasztikához hasonló citokinváltozásokat tapasztaltunk, ami bizonyítja, hogy az allogenetikus reakció nem az egyetlen faktor, mely a citokinszekréciót befolyásolja.

Megjegyzések:Pro- (Interleukin-6 (IL-6)) és anti-inflammatorikus (Interleukin-10 (IL-10)) citokinek meghatározása perforáló keratoplasticán átesett betegek könnyéből (különböző posztoperativ időszakban). Összehasonlítani ezen citokinek szintjét a komplikáció nélküli és a kilökődési reakción átesett betegek esetében. Betegek és módszerek: Prospektív módon, 12 szaruhártya-átültetésen átesett betegtől stimulálás nélkül könnyet gyűjtöttünk a posztoperativ 14 hónap során. A vizsgált időszakban 3 betegnél kilökődési reakció zajlott, míg 9 beteg esetében a transzplantátumok komplikáció mentesek voltak. A levett könnyminták IL-6 és IL-10 koncentrációját áramlási citométerrel mértük meg. Eredmények: Az IL-6 koncentrációja szignifikánsan magasabb (p=0,009), míg az IL-10 koncentrációja szignifikánsan alacsonyabb volt (p=0,008) a kilökődési reakción átesett betegeknél, a komplikációmentes esetekkel összehasonlítva. Az IL-6/IL-10 arány a vizsgált időszakban végig szignifikánsan magasabb volt (p=0,0231) a kilökődési reakción áteső betegeknél. Következtetések: A könnyben mért emelkedett IL-6 és az ezzel együtt járó csökkent IL-10 szint a szaruhártya kilökődés indikátora lehet. Ezek a könnyben kimutatott változások azonban a transzplantáció végső kimenetelét nem jelzik előre.

Megjegyzések:This was a kinetic study of inflammatory cytokine levels in postoperative tear samples from penetrating keratoplasty (PKP) patients with or without corneal rejection. In a prospective design, nonstimulated tears were collected from the affected eyes of 12 patients at regular intervals for 12-14 months following PKP. Nine patients retained clear grafts, whereas three suffered endothelial rejection of the corneal graft within 14 months. The concentrations of the cytokines IL-1beta, IL-6, TNF-alpha, IL-8, IL-10, and IL-12p70 were measured via cytometric bead array technology. The postoperative concentrations of the cytokines in the tears varied among the patients, but exhibited similar alteration patterns in each eye tested. The concentrations of IL-6 and IL-8 were significantly higher (P = 0.009 and P = 0.01, respectively), whereas those of IL-10, TNF-alpha, and IL-12p70 were significantly lower (P = 0.008, P = 0.006, and P = 0.0009, respectively) in the tear samples from the patients with corneal rejection as compared with those with uncomplicated corneal grafts. The ratios IL-6/IL-10 and IL-8/IL-10 were significantly higher (P = 0.0231 and P = 0.015, respectively), and TNF-alpha/IL-10 was significantly lower (P = 0.045) throughout the examination period in the patients with endothelial rejection. The enhanced release of IL-6 and IL-8 into the tears of patients with corneal graft rejection concomitant with decreased concentrations of IL-10, TNF-alpha, and IL-12p70 may possibly serve as an indicator of the rejection process. However, due to the large variation in the cytokine concentrations, the observed changes in tear composition do not categorically predict the final graft outcome.

Megjegyzések:The synthetic peptide comprising the 317-341 region of human influenza A virus (H1N1 subtype) hemagglutinin elicits peptide-specific antibody and helper T cell responses and confers protection against lethal virus infection. Molecular mapping of the 317-329 region, which encompasses the epitope recognized by peptide-specific T cells, revealed that the minimal size required for T cell activation was the 317-326 segment. The most likely peptide alignment, which placed 320Leu to pocket 1 of the I-E(d) peptide binding groove, was predicted by molecular mechanics calculations performed with the parental and with the Ala-substituted analogs. In line with the prediction data, the results of the peptide binding assay, where the relative binding efficiency to I-E(d) molecules expressed on the surface of antigen-presenting cells was monitored, identified the 320-326 core sequence interacting with the major histocompatibility class II peptide binding groove. Functional analysis of Ala-substituted variants by functional assays and by calculating the surface-accessible areas of the single peptidic amino acids in the I-E(d)-peptide complexes demonstrated that 324Pro is a primary contact residue for the T cell receptor. Our results show that this type of analysis offers a suitable tool for molecular mapping of helper T cell epitopes and thus provides valuable data for subunit vaccine design.Copyright 2000 Academic Press.

Megjegyzések:The cardinal role of dendritic cells (DC) in priming adaptive immunity and in orchestrating immune responses against all classes of pathogens and also against tumors is well established. Their unique potential both to maintain self-tolerance and to initiate protective immune responses against foreign and/or dangerous structures is based on the functional diversity and flexibility of these cells. Tissue DC lining antigenic portals such as mucosal surfaces and the skin are specialized to take up a wide array of compounds including proteins, lipids, carbohydrates, glycoproteins, glycolipids and oligonucleotides, particles carrying such structures and apoptotic or necrotic cells. This process is facilitated by specialized receptors with high endocytic capacity, which provides potential targets for delivering designed molecules. The best route for targeting B- and/or T cell epitopes, however, is still the subject of intense investigation. Immature DC, which reside in various tissues, can be activated by pathogens, stress and inflammation or modified metabolic products, which induce mobilization of cells to draining lymph nodes where they act as highly potent professional antigen presenting cells. This is brought about by the ability to present their accumulated intracellular content for both CD4+ helper (Th) and CD8+ cytotoxic/cytolytic T lymphocytes (Tc/CTL). Engulfed proteins are processed intracellularly and their peptide fragments are transported to the cell surface in the context of major histocompatibility complex encoded class I and II molecules for presentation to Th cells and CTLs, respectively. The T cell priming capacity of DC, however, depends not only on antigen presentation but also on other features of DC. Human monocyte-derived DC provide an excellent tool to study the internalizing, antigen-presenting and T cell-activating functions of DC at their immature and activated differentiation states. These biological activities of DC, however, are highly dependent on their migratory potential from the peripheral non-lymphoid tissues to the lymph nodes, on the expression of adhesion molecules, which support the interaction of DC with T lymphocytes, and the cytokines secreted by DC, which polarize immune responses to Th1-mediated cellular or Th2-mediated antibody responses. These results altogether demonstrate that monocyte-derived DC are useful candidates for in vitro or in vivo targeting of antigens to induce efficient adaptive immune responses against pathogens and also against tumors.

Megjegyzések:Accumulating data have shown that the microenvironment of dendritic cells modulates subtype differentiation and CD1 expression, but the mechanisms by which exogenous factors confer these effects are poorly understood. Here we describe the dependence of CD1a- monocyte-derived dendritic cell (moDC) development on lipids associated with the expression of peroxisome proliferator-activated receptor-gamma (PPARgamma). We also show the consecutive differentiation of immature CD1a-PPARgamma+ moDCs to CD1a+PPARgamma- cells limited by serum lipoproteins and terminated by proinflammatory cytokines. Immature CD1a- moDCs possess higher internalizing capacity than CD1a+ cells, whereas both activated subtypes have similar migratory potential but differ in their cytokine and chemokine profiles, which translates to distinct T-lymphocyte-polarizing capacities. CD1a+ moDCs stand out by their capability to secrete high amounts of IL-12p70 and CCL1. As lipoproteins skew moDC differentiation toward the generation of CD1a-PPARgamma+ cells and inhibit the development of CD1a+PPARgamma- cells, we suggest that the uptake of lipids results in endogenous PPARgamma agonists that induce a cascade of gene transcription coordinating lipid metabolism, the expression of lipid-presenting CD1 molecules, subtype dichotomy, and function. The presence of CD1a-PPARgamma+ and CD1a+PPARgamma- DCs in lymph nodes and in pulmonary Langerhans cell histiocytosis confirms the functional relevance of these DC subsets in vivo.
