Megjegyzések:Pituitary adenylate cyclase activating polypeptide (PACAP) is a multifunctional and pleiotropic neuropeptide. PACAP hasdiverse effects in the endocrine system, among others, it plays important roles in oogenesis, implantation and development ofthe nervous system.However, it is not knownwhether PACAP is present in the fluids of the human reproductive organs. The aimof the present study was to determine, by means of mass spectrometry and radioimmunoassay, whether PACAP is present inhuman amniotic fluid, ovarian follicular fluid and cervico-vaginal fluid. Samples were obtained from healthy adult volunteers.Our MALDI TOF and MALDI TOF/TOF spectrometry results show that PACAP38 is present in all of the follicular fluid samples,and PACAP-like immonoreactivity was also measured by radioimmunoassay. However, we did not find the characteristic peakrepresenting the unmodified 38 amino acid form of the peptide in normal cervico-vaginal smear and amniotic fluid samples.Furthermore, we analyzed other body fluids for comparison, such as human nasal fluid, saliva and aqueous humor. PACAPwas not found in these latter samples. In summary, the present study provides evidence for the presence of PACAP in humanfollicular fluid, suggesting a role in oocyte function, but determination of the exact physiological significance awaits furtherinvestigation.

Megjegyzések:Pituitary adenylate cyclase activating polypeptide (PACAP), a multifunctional neuropeptide,has 2 active forms, PACAP38 and PACAP27. It is now well-established that PACAPhas several actions also in peripheral organs, including renoprotective effects. The peptideitself has not been previously identified in the rat kidney. The first aim of our study was toidentify PACAP in the rat kidney using mass spectrometry and radioimmunoassay (RIA).Receptor mRNA and binding studies revealed the existence of all 3 PACAP receptors(PAC1, VPAC1, and VPAC2) in the kidney, but their exact localization in histologicsections was not evident. Because most of the cytoprotective effects of PACAP relate to itsspecific PAC1 receptor, our second aim was to identify the cell types wherein the PAC1receptor is expressed in the rat kidney. Mass spectrometry revealed the presence ofPACAP38 in the kidney. RIA measurements showed both PACAP38- and PACAP27-likeimmunoreactivities in kidney homogenates, with PACAP38 being dominant. Immunohistochemistryrevealed PAC1 receptor-like immunoreactivity in kidney sections, mainlyexpressed in cortical tubular epithelial cells. These results showed PACAP to beendogenously present in the kidney. The tubular localization of the PAC1 receptorprovides the basis for the renal effects of the peptide under physiologic and pathologicconditions.

Megjegyzések:Pituitary adenylate cyclase-activating polypeptide(PACAP), a neuropeptide with survival-promotingactions, has been observed in endocrine organs and isthought to play a role in reproductive functions, includingpregnancy. PACAP occurs in two forms, 27 and 38 aminoacid residues, with PACAP38 being the predominant formin human tissues. In the present study, we determined theconcentrations of PACAP38 and PACAP27 in firsttrimesterand full-term human placentas using radioimmunoassay.We found high levels of PACAP38 and lowerlevels of PACAP27 in different parts of the full-term humanplacenta. PACAP38 content increased in the placentaduring pregnancy, both on the maternal side and on thefetal side. The effects of PACAP on the survival of JARhuman choriocarcinoma cells were investigated using flowcytometry and MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) cell viability assay in cellsexposed to the widely used chemotherapeutic agentmethotrexate (MTX). It was found that PACAP neitherinfluenced the survival of JAR cytotrophoblast cells noraffected cellular response to the death-inducing effect of thechemotherapeutic agent MTX. The present observationsfurther support the significance of PACAP in the humanplacenta. The observation that PACAP did not influence theeffects of MTX may have future clinical importance,showing that PACAP does not decrease the effects ofcertain chemotherapeutic agents.Keywords JAR . Megjegyzések:Milk contains a variety of proteins and peptides that possess biological activity. Growth factors, such as growth hormone, insulin-like, epidermal and nerve growth factors are important milk components which may regulate growth and differentiation in various neonatal tissues and also those of the mammary gland itself. We have recently shown that pituitary adenylate cyclase-activating polypeptide (PACAP), an important neuropeptide with neurotrophic actions, is present in the human milk in much higher concentration than in the plasma of lactating women. Investigation of growth factors in the milk of domestic animals is of utmost importance for their nutritional values and agricultural significance. Therefore, the aim of the present study was to determine the presence and concentration of PACAP in the plasma and milk of three ruminant animal species. Furthermore, the presence of PACAP and its specific PAC1 receptor were investigated in the mammary glands. Radioimmunoassay measurements revealed that PACAP was present in the plasma and the milk of the sheep, goat and the cow in a similar concentration to that measured previously in humans. PACAP38-like immunoreactivity (PACAP38-LI) was 5-20-fold higher in the milk than in the plasma samples of the respective animals, a similar serum/milk ratio was found in all the three species. The levels did not show significant changes within the examined 3-month-period of lactation after delivery. Similar PACAP38-LI was measured in the homogenates of the sheep mammary gland samples taken 7 and 30 days after delivery. PAC1 receptor expression was detected in these udder biopsies by fluorescent immunohistochemistry suggesting that this peptide might have an effect on the mammary glands themselves. These data show that PACAP is present in the milk of various ruminant domestic animal species at high concentrations, the physiological implications of which awaits further investigation.

The neuroprotective effectsof PACAP are well-established in different neuronal systems against noxious stimuli in vitro and in vivo.Recently, its general cytoprotective actions have been recognized, including renoprotective effects. However,the effect of endogenous PACAP in the kidneys is not known. The main aim of the present studywas to investigate whether the lack of this endogenous neuropeptide influences survival of kidney cellsagainst oxidative stress. First, we determined the presence of endogenous PACAP from mouse kidneyhomogenates by mass spectrometry and PACAP-like immunoreactivity by radioimmunoassay. Second,primary cultures were isolated from wild type and PACAP deficient mice and cell viability was assessedfollowing oxidative stress induced by 0.5, 1.5 and 3mM H2O2. Our mass spectrometry and radioimmunoassayresults show that PACAP is endogenously present in the kidney. The main part of our studyrevealed that the sensitivity of cells from PACAP deficient mice was increased to oxidative stress: bothafter 2 or 4 h of exposure, cell viability was significantly reduced compared to that from control wild typemice. This increased sensitivity of kidneys from PACAP deficient mice could be counteracted by exogenouslygiven PACAP38. These results show, for the first time, that endogenous PACAP protects againstoxidative stress in the kidney, and that PACAP may act as a stress sensor in renal cells. These findingsfurther support the general cytoprotective nature of this neuropeptide.

Pituitary adenylate cyclaseactivatingpolypeptide (PACAP) plays an important role in theregulation of several endocrine processes and is present inovarian follicular fluid (FF). However, little is known aboutPACAP in FF with regard to maturation, ovulation, fertilization,and successful pregnancy. The aim of this pilot study wasto investigate whether there is a correlation between PACAPconcentration in FF and ovarian response to superovulationtreatment in infertile women, performed in volunteers (n0132; aged between 20 and 35). After treatment, the numberof harvested oocytes was recorded and PACAP immunoreactivityin FF was measured by radioimmunoassay. All thecorresponding PACAP concentrations were below 290 fmol/ml in cases when the number of harvested oocytes exceeded 14per patient, while in all cases above 290 fmol/ml, the number ofoocytes was below 14. Using these cutoff values, we determinedthree study groups: high-PACAP concentration, highoocytenumber, and low-PACAP concentration?low-oocytenumber groups. Median values of PACAP concentration inthese groups were 411.2, 106.5, and 101.0 fmol/ml, respectively,while themedian values of harvested oocyteswere 5.5, 19.0,and 5.0, respectively. Differences were significant, indicating acorrelation between concentration of PACAP in FF and thenumber of recruited oocytes. Higher concentrations of PACAPin FF might be associated with lower number of developingoocytes, while low concentrations of PACAP might correlatewith a markedly higher number of ova retrieved, thus predictinga higher chance for ovarian hyperstimulation. Our presentstudy is among the first few human clinical studies with directconclusions drawn for possible clinical impact of PACAP.

Megjegyzések:The effects of pituitary adenylate cyclase activating polypeptide (PACAP) are mediated through Gprotein-coupled receptors, the specific PAC1 receptor and VPAC1 and VPAC2 receptors which bind vasoactive intestinal peptide with similar affinity. Based on binding affinity studies, PACAP6-38 was discovered as a potent antagonist of PAC1 and it has been used by hundreds of studies as a PACAP antagonist. Recently, we have found that in certain cells/tissues, PACAP6-38 does not antagonize PACAP-induced effects, but surprisingly, it exerts similar actions to PACAP1-38, behaving as an agonist. In the present study, we report on the agonistic behavior of PACAP6-38 on neuropeptide release from sensory nerves of the isolated rat trachea and on the MAPK signaling pathways in cytotrophoblast cells. In isolated rat tracheae, PACAP6-38, similarly to PACAP1-38, induced significant inhibitory effects on the release of three simultaneously measured sensory neuropeptides, substance P, calcitonin gene-related peptide, and somatostatin evoked by both chemical excitation and electrical field stimulation of capsaicin-sensitive afferents. Effects of PACAP6-38 were the same as those of PACAP1-38 on MAPK signaling in human cytotrophoblast cells. Western blot analysis showed that both peptide forms stimulated ERK1/2 and JNK phosphorylation, while they both inhibited p38 MAPK phosphorylation. The most pronounced effects were observed when both peptides were present. In summary, our results show that PACAP6-38, which is a PACAP receptor antagonist in most cells/tissues, can behave as an agonist in other systems. The increasing interest in the effects of PACAP requires further studies on the pharmacological properties of the peptide and its analogues.

Megjegyzések:Pituitary adenylate cyclase activating polypeptide (PACAP) is present in the cranial arteries and trigeminalsensory neurons. We therefore examined the alterations in PACAP-like immunoreactivity (PACAP-LI) ina time-dependent manner in two rat models of trigeminovascular system (TS) activation. In one groupchemical stimulation (CS) was performed with i.p. nitroglycerol (NTG), and in the other one the trigeminalganglia (TRG) were subjected to electrical stimulation (ES). The two biologically active forms, PACAP-38and PACAP-27, were determined by means of radioimmunoassay (RIA) and mass spectrometry (MS) inthe plasma, the cerebrospinal fluid (CSF), the trigeminal nucleus caudalis (TNC), the spinal cord (SC) andthe TRG. The tissue concentrations of PACAP-27 were 10 times lower than those of PACAP-38 in the TNCand SC, but about half in the TRG. PACAP-38, but not PACAP-27, was present in the plasma. Neither formcould be identified in the CSF. PACAP-38-LI in the plasma, SC and TRG remained unchanged after CS, butit was increased significantly in the TNC 90 and 180 min after NTG injection. In response to ES of the TRG,the level of PACAP-38 in the plasma and the TNC was significantly elevated 90 and 180 min later, but notin the SC or the TRG. The alterations in the levels of PACAP-27 in the tissue homogenates in responseto both forms of stimulation were identical to those of PACAP-38. The selective increases in both formsof PACAP in the TNC suggest its important role in the central sensitization involved in migraine-likeheadache.
