Megjegyzések:Pituitary adenylate cyclase activating polypeptide (PACAP) is a multifunctional and pleiotropic neuropeptide. PACAP hasdiverse effects in the endocrine system, among others, it plays important roles in oogenesis, implantation and development ofthe nervous system.However, it is not knownwhether PACAP is present in the fluids of the human reproductive organs. The aimof the present study was to determine, by means of mass spectrometry and radioimmunoassay, whether PACAP is present inhuman amniotic fluid, ovarian follicular fluid and cervico-vaginal fluid. Samples were obtained from healthy adult volunteers.Our MALDI TOF and MALDI TOF/TOF spectrometry results show that PACAP38 is present in all of the follicular fluid samples,and PACAP-like immonoreactivity was also measured by radioimmunoassay. However, we did not find the characteristic peakrepresenting the unmodified 38 amino acid form of the peptide in normal cervico-vaginal smear and amniotic fluid samples.Furthermore, we analyzed other body fluids for comparison, such as human nasal fluid, saliva and aqueous humor. PACAPwas not found in these latter samples. In summary, the present study provides evidence for the presence of PACAP in humanfollicular fluid, suggesting a role in oocyte function, but determination of the exact physiological significance awaits furtherinvestigation.

Megjegyzések:Pituitary adenylate cyclase-activating polypeptide(PACAP), a neuropeptide with survival-promotingactions, has been observed in endocrine organs and isthought to play a role in reproductive functions, includingpregnancy. PACAP occurs in two forms, 27 and 38 aminoacid residues, with PACAP38 being the predominant formin human tissues. In the present study, we determined theconcentrations of PACAP38 and PACAP27 in firsttrimesterand full-term human placentas using radioimmunoassay.We found high levels of PACAP38 and lowerlevels of PACAP27 in different parts of the full-term humanplacenta. PACAP38 content increased in the placentaduring pregnancy, both on the maternal side and on thefetal side. The effects of PACAP on the survival of JARhuman choriocarcinoma cells were investigated using flowcytometry and MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) cell viability assay in cellsexposed to the widely used chemotherapeutic agentmethotrexate (MTX). It was found that PACAP neitherinfluenced the survival of JAR cytotrophoblast cells noraffected cellular response to the death-inducing effect of thechemotherapeutic agent MTX. The present observationsfurther support the significance of PACAP in the humanplacenta. The observation that PACAP did not influence theeffects of MTX may have future clinical importance,showing that PACAP does not decrease the effects ofcertain chemotherapeutic agents.Keywords JAR . Megjegyzések:Tissue injury caused by cold preservation and reperfusion during small bowel transplantation remains an unsolved problem. Increasing evidence suggests that pituitary adenylate cyclaseactivating polypeptide (PACAP) has protective effects in several ischemia-reperfusion (I/R) models. This study investigated the effect of PACAP-38 on oxidative stress in autotransplanted intestine. We established sham-operated, I/R, and autotransplanted groups in Wistar rats (n = 55). We applied ischemia for 1 (GI), 2 (GII), or 3 hours (GIII). In autotransplanted groups, we performed total orthotopic intestinal autotransplantation. Grafts were preserved in University of Wisconsin (UW) solution for 1 (GIV), 2 (GV), 3 (GVI), or 6 (GVII) hours and in PACAP-38-containing UW for 1 (GVIII), 2 (GIX), 3 (GX), or 6 (GXI) hours. Reperfusion lasted 3 hours in each group. Endogenous PACAP-38 values were measured by radioimmunoassay. Oxidative stress parameters malondialdehyde (MDA), reduced glutathione (GSH), and superoxide dismutase (SOD) were measured in tissue homogenates. Concentration of endogenous PACAP-38 significantly decreased in GI to GIII compared with the sham-operated animals following I/R periods (P < .05). Cold preservation in UW and reperfusion of the intestine increased the level of tissue MDA in GIV to GVII, which correlated with the duration of cold storage. The content of GSH decreased in GIV to GVII to levels that were significantly different between GIV and GVIII and between GVII and GXI. SOD activity decreased dramatically in GIV to GVII with significantly higher activity in GIX to GXI. Our findings confirmed that I/R decreased endogenous PACAP-38 concentration. Administration of PACAP-38 to UW solution mitigated the oxidative injury during intestinal autotransplantation.

Megjegyzések:Follicular growth, ovulation, and luteinization areinfluenced by interactions of peptide and steroid hormonesignalingcascades in the ovary. Pituitary adenylate cyclaseactivatingpolypeptide (PACAP) plays an important role in theregulation of several endocrine processes and is present inovarian follicular fluid (FF). However, little is known aboutPACAP in FF with regard to maturation, ovulation, fertilization,and successful pregnancy. The aim of this pilot study wasto investigate whether there is a correlation between PACAPconcentration in FF and ovarian response to superovulationtreatment in infertile women, performed in volunteers (n0132; aged between 20 and 35). After treatment, the numberof harvested oocytes was recorded and PACAP immunoreactivityin FF was measured by radioimmunoassay. All thecorresponding PACAP concentrations were below 290 fmol/ml in cases when the number of harvested oocytes exceeded 14per patient, while in all cases above 290 fmol/ml, the number ofoocytes was below 14. Using these cutoff values, we determinedthree study groups: high-PACAP concentration, highoocytenumber, and low-PACAP concentration?low-oocytenumber groups. Median values of PACAP concentration inthese groups were 411.2, 106.5, and 101.0 fmol/ml, respectively,while themedian values of harvested oocyteswere 5.5, 19.0,and 5.0, respectively. Differences were significant, indicating acorrelation between concentration of PACAP in FF and thenumber of recruited oocytes. Higher concentrations of PACAPin FF might be associated with lower number of developingoocytes, while low concentrations of PACAP might correlatewith a markedly higher number of ova retrieved, thus predictinga higher chance for ovarian hyperstimulation. Our presentstudy is among the first few human clinical studies with directconclusions drawn for possible clinical impact of PACAP.

Megjegyzések:Pituitary adenylate cyclase-activating polypeptide (PACAP)was discovered from the hypothalamus and it belongs tothe vasoactive intestinal peptide (VIP)/secretin/glucagonspeptide family (1). It occurs in two biologically active forms,with 38 and 27 amino-acid residues. The first studies reportedon its adenylate cyclase activation in pituitary cellsand described its distribution in hypothalamic nuclei influencingpituitary hormone secretion (1). Although the last 20years since its discovery have revealed numerous distincteffects in the nervous system and various peripheral organs,its endocrine functions still represent a major focusin PACAP research. PACAP influences, among others, therelease of GnRH and FSH in different species (2, 3) andthe secretion of PRL (4-6). PACAP also influences sexualbehavior, ovarian follicular maturation, and fertility (3, 7,8), uteroplacental circulation (9), and trophoblast survival(10). Furthermore, PACAP is a very important trophic factor,especially during the development of the nervous system(1, 11). Radioimmunoassay (RIA) analysis revealed thatPACAP38 concentration is higher in the hypophyseal portalblood than in the peripheral circulation in the rat (12). Inspite of the vast literature on PACAP, there are only fewhuman studies. The presence of PACAP has been shownin human blood and some other tissues (1, 13). The half-lifeof PACAP in the circulation is relatively short, only a fewminutes, due to rapid enzymatic degradation by dipeptidyl-peptidase IV (14). We have recently shown the presenceof PACAP38 in human milk by mass spectrometry,and we have also found that PACAP38-like immunoreactivity(PACAP38-LI) is 5-20 times higher in the milk than inthe plasma (15). In humans, PACAP has also been shownto influence PRL, vasopressin, and oxytocin secretion (5,16). Immunohistochemical studies have revealed the presenceof PACAP and its receptors in the female reproductivesystem, including the placenta, uterus, and mammarygland (9, 17). Recent studies have suggested the involvementof PACAP in the pathophysiological processes ofstress urinary incontinence and the prolapse of pelvic organsin female patients (18).Regarding the role of PACAP in reproductive functionsand the urging need to obtain further human data, theaim of the present study was to investigate whether theconcentration of PACAP is altered during normal pregnancy,delivery, and after childbirth. We also measuredthe levels of endogenous PACAP with RIA in the peripheralblood and of newborn babies, as well as in the bloodobtained from the umbilical arteries and veins.

Recently, we have shown the presence ofPACAP in human plasma, milk, placenta, and follicularfluid samples. However, relatively few data are availableon PACAP in human tissues from patients with differentdisorders. The aim of the present study was to determine, byradioimmunoassay, the tissue level of PACAP38-like immunoreactivity(LI) and PACAP27-LI in different primary nonsmallcell lung cancer, colon tumor samples, and in cardiacmuscle samples from patients suffering from ischemic heartdisease and valvular disorders. We also labeled the PAC1receptors in human cardiac cells. All samples showed significantlyhigher PACAP38-LI compared with PACAP27-LI. We found significantly lower levels of PACAP38-LI andPACAP27-LI in tumoral and peripheral samples comparedwith normal healthy tissue in both lung and colon cancers.Further investigations are necessary to describe the exactfunction of PACAP in oncogenesis. We showed thatPACAP38-LI and PACAP27-LI are significantly higher inischemic heart diseases compared with valvular abnormalities,suggesting that PACAP might play a role in ischemicheart disorders.
