Megjegyzések:Pituitary adenylate cyclase-activating polypeptide(PACAP), a neuropeptide with survival-promotingactions, has been observed in endocrine organs and isthought to play a role in reproductive functions, includingpregnancy. PACAP occurs in two forms, 27 and 38 aminoacid residues, with PACAP38 being the predominant formin human tissues. In the present study, we determined theconcentrations of PACAP38 and PACAP27 in firsttrimesterand full-term human placentas using radioimmunoassay.We found high levels of PACAP38 and lowerlevels of PACAP27 in different parts of the full-term humanplacenta. PACAP38 content increased in the placentaduring pregnancy, both on the maternal side and on thefetal side. The effects of PACAP on the survival of JARhuman choriocarcinoma cells were investigated using flowcytometry and MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) cell viability assay in cellsexposed to the widely used chemotherapeutic agentmethotrexate (MTX). It was found that PACAP neitherinfluenced the survival of JAR cytotrophoblast cells noraffected cellular response to the death-inducing effect of thechemotherapeutic agent MTX. The present observationsfurther support the significance of PACAP in the humanplacenta. The observation that PACAP did not influence theeffects of MTX may have future clinical importance,showing that PACAP does not decrease the effects ofcertain chemotherapeutic agents.Keywords JAR . Megjegyzések:The aim of the present article was to investigate the influenceof gonadectomy on pituitary adenylate cyclase-activating polypeptide(PACAP) levels in different brain areas. In males, there seems tobe an inverse relationship between gonadotropins and PACAP in thebrain in the acute phase of castration: PACAP levels decreased in almostall brain areas examined within the first week after castration. Infemales, such pattern was observed in the hypothalamus, brain stem,and temporal cortex. In the pituitary, levels decreased only on the firstday after ovariectomy, and later, as in the thalamus, increases were observed.Although the pattern of change showed gender differences, ourresults provide further evidence that levels of gonadotropins and possiblygonadotropin-releasing hormone influence PACAP levels and thatPACAP is involved in the regulation of gonadal functions.

Megjegyzések:PACAP (pituitary adenylate cyclase-activating polypeptide) occurs in two biologicallyactive forms, with 38 and 37 amino acid residues (PACAP38 and PACAP27). Inmammalian tissues, PACAP38 is the dominant form. Diverse effects have been describedin the cardiovascular, respiratory, gastrointestinal, and urogenital systems. PACAP isknown for its strong cytoprotective effects, which are present endogenously as well, asproven by knockout studies and results showing that PACAP is up-regulated followingdiverse injuries. Little is known about such effects in the kidney. We have previously shownthat PACAP is protective in renal ischemia-reperfusion injury. Therefore, the aim of thepresent study was to investigate the changes of endogenous PACAP following 60-minuterenal ischemia using radioimmunoassay. Changes were observed within 24 hours followingrenal vessel clamping. In the cortex, an acute decrease was followed by an increase on theintact side, and levels returned to original ones on the operated side. In the medulla,changes were only observed on the clamped side: a marked up-regulation was detected inPACAP38-like immunoreactivity within the first 24 hours. The present study showed thatPACAP38- and PACAP27-like immunoreactivities sensitively react to renal ischemiareperfusion,the physiological importance of which awaits further investigation.
