Megjegyzések:Interaction of copper(II) and nickel(II) ions with the Ac-PHAAAGTHSMKHM-NH2 tridecapeptide containing the His85, His96 and His111 binding sites of human prion protein has been studied by various techniques. pH-potentiometry, UV?Vis and circular dichroism spectroscopy were applied to study the stoichiometry, stability and structure of the copper(II) and nickel(II) complexes, while HPLC-MS and MS/MS were used for identifying the products of copper(II) catalyzed oxidation. The copper binding ability of shorter fragments, namely the nonapeptide Ac-PHAAAGTHS-NH2 and pentapeptide Ac-PHAAA-NH2 have also been studied. The tridecapeptide is able to bind three equivalent of copper(II) ion, since the histidine residues behave as independent metal binding sites. Nevertheless, the metal binding ability of histidine residue mimicking the octarepeat domain (His85) is decreased, while the other parts of the peptide mimicking the histidines outside the octarepeat domain bind the copper ions in comparable concentration. On the other hand, this peptide is able to coordinate only two equivalents of nickel ion on the domains outside the octarepeat region. Furthermore the His96 binding site is more effective for the nickel ions. Both histidine and methionine residues are sensitive for oxidation, the oxidation of these residues are proved, and in the case of the histidine residues follows the order His96?>His85>>His111.

Megjegyzések:Munkám során a patkány amilin 17-22-es szakaszát modellező fragmensek és mutánsok Cu(II)-komplexeit vizsgáltam. Igazoltuk, hogy a terminális aminocsoport szerepe messze fölülmúlja az aszparagin oldallánc szerepét a fémion megkötésében. A CD-, UV- és ESR-spektroszkópiai adatok elemzésével alátámasztottuk, hogy az aszparagin-oldallánc még szabad N-terminus mellett is részt vesz a Cu(II)-ion koordinálásában.Bizonyítottuk, hogy az ?SSNN? domén még a terminális aminocsoport mellett is másodlagos kötőhelyként funkcionálhat.

Megjegyzések:Copper(II), nickel(II) and zinc(II) complexes of two N-terminally free and C-terminally blocked hexapeptides, NH2-ADAAAH-NH2 and NH2-AADAAH-NH2, containing separate aspartyl and histidyl residues have been studied by potentiometric, UV-Vis, CD and ESR spectroscopic methods. The amino termini were found as the primary anchoring sites of both ligands for the complexation with all three metal ions. The β-carboxylate function of the second or third aspartyl residue enhances the thermodynamic stability of the copper(II) and nickel(II) complexes and shifts the deprotonation of the subsequent amide functions into a more alkaline pH range. In the case of NH2-AADAAH-NH2 the imidazole-N donor of the histidyl residue does not have a significant contribution to the overall stability of the mononuclear complexes. The side chain imidazole, however, can be an independent metal binding site resulting in the formation of dinuclear or even mixed metal complexes. The stabilizing role of the histidyl residue is much more pronounced in the complexes of the NH2-ADAAAH-NH2 peptide. In this case a tridentately (NH2,N?,β-COO?)-coordinated species is formed and its stability is significantly enhanced by the macrochelation of the side chain imidazole. The presence of two anchoring sites (terminal amino and side chain imidazole) in one molecule enhances the stability of the corresponding zinc(II) complexes, too, but the amide nitrogens are not involved in metal binding in this case.

Megjegyzések:Nickel(II), zinc(II) and cadmium(II) complexes of two N-terminally free hexapeptides AAHAAC andAHAAAC containing separate histidyl and cysteinyl residues have been studied using potentiometric andvarious spectroscopic (UV-Vis, CD, 1H-NMR) techniques. Both peptides have outstanding metal bindingability but the speciation of the systems and the binding sites of peptides reveal a significant specificity.In the nickel(II)?AAHAAC system the amino terminus is the primary nickel(II) binding site in the form ofthe (NH2,N-,N-,Nim) chelate. However, the C-terminal thiolate function can bind another nickel(II) ionwith the involvement of amide nitrogens in metal binding. Zinc(II) and cadmium(II)ions are not ableto promote deprotonation and coordination of the peptide amide groups of AAHAAC and onlymononuclear complexes are formed, in which imidazole-N and thiolate-S- donors are the primarymetal binding sites. In the case of AHAAAC both nickel(II) and zinc(II) ions can induce deprotonation andcoordination of the first amide bond in the sequence. This results in the enhanced stability of thecorresponding species, [MH-1L], containing a tridentate (NH2,N-,Nim) binding mode at the aminoterminus supported by a macrochelate from the distant thiolate group.

Megjegyzések:Nickel(II) complexes of A beta(1-16)Y10A and its smaller fragments including A beta(1-4), A beta(1-6), Ac-A beta(1-6) and Ac-A beta(8-16)Y10A have been studied by potentiometric, UV-Vis and circular dichroism spectroscopic measurements. The formation of mixed metal complexes and the distribution of metal ions among the possible coordination sites in the Cu(II)-Ni(II)-A beta(1-16)Y10A and Cu(II)-Ni(II)-Zn(II)-A beta(1-16)Y10A systems have also been evaluated. It was found that the hexadecapeptide and its fragments are effective nickel(II) binding ligands and complex formation processes of nickel(II) ions are quite similar to those of copper(II). Formation of mono-and di-nuclear complexes was detected in the nickel(II)-A beta(1-16)Y10A system suggesting the existence of two separated metal binding motifs: the N-terminus and internal histidyl residues. The preference for the coordination at the N-terminus was supported by the spectroscopic measurements but in equilibrium with the metal binding at the internal histidyl sites. Neither zinc(II) nor nickel(II) can, however, substitute copper(II) in the mixed metal complexes of A beta(1-16)Y10A, but both metal ions are able to alter the distribution of copper(II) ions among the various binding sites. Both N-terminus (amino and His6) and internal histidyl residues (His13 and His14) can work as dinuclear binding motifs, preferably accommodating copper(II) and zinc(II), respectively, while nickel(II) can occupy the remaining free coordination sites.

Megjegyzések:Terminally protected pentapeptides with 2 histidines (Ac-HHVGD-NH2 and Ac-HVGDH-NH2) and the terminally free peptides containing both internal aspartyl and C-terminal histidyl residues (FDAH and VIDAH) have been synthesized, and copper(II) complexes studied by potentiometric, UV-Vis, CD, and EPR spectroscopic techniques in solution. Both thermodynamic and spectroscopic data reveal that side chain donor atoms of aspartyl and histidyl residues have a significant contribution to the metal binding affinity of peptide molecules. In the case of terminally protected peptides, the role of the imidazole-N donor functions is reflected in the enhanced stability of the 3N and 4N coordinated copper(II) complexes. The amino and ?-carboxylate groups of FDAH and VIDAH create a very effective metal binding site with the (NH2, N?, ?-COO?) and (NH2, N?, N?, ?-COO?) coordination modes including the N-termini, while the histidine sites are available for the formation of the (Nim, N?, N?) binding mode resulting in the preference of dinuclear complex formation.

Megjegyzések:Copper(II) complexes of the pentapeptides Ac-HisAlaHisValHis-NH2, Ac-HisValHisAlaHis-NH2, Ac-HisProHisAlaHis-NH2, Ac-HisAlaHisProHis-NH2, Ac-HisGlyHisValHis-NH2 and Ac-HisValHisGlyHis-NH2 have been studied by potentiometric, UV-Vis, CD and EPR spectroscopic methods. It has been found that the pentapeptides are efficient ligands for the complexation with copper( II) and exhibit an outstanding versatility in the co-ordination geometry of complexes. The presence of three histidyl residues provides a high possibility for the formation of macrochelates via the exclusive binding of imidazole-N donor atoms. The macrochelation suppresses, but cannot preclude the deprotonation and metal ion co-ordination of amide functions and the species [CuH-2L] and [Cu2H-4L] predominate at physiological pH in equimolar solutions and in the presence of excess metal ions, respectively. It is also clear from the data that both C-terminal and internal histidyl residues can work as the anchoring sites for metal binding and subsequent amide deprotonation resulting in the formation of co-ordination isomers and dinuclear species in equimolar solutions and in the presence of excess metal ions, respectively. In more alkaline solutions ( pH similar to 10) a third amide function can be deprotonated and co-ordinated in the species [CuH-3L](-) with (N-, N-, N-, N-im) co-ordination. The dinuclear species [Cu2H-5L](-) and [Cu2H-6L](2-) containing hydroxide ions and/or imidazolato bridges are formed at high pH in the presence of excess of metal ions. The insertion of one proline into the sequence preceding histidyl residues hinders the deprotonation of amide functions at that site and the formation of only mononuclear complexes was observed with these peptides.

Megjegyzések:Copper(II), nickel(II) and zinc(II) complexes of the peptides GlyLys(Gly) and Asp-epsilon-Lys, containing the amide functions at the c-amino groups of the lysyl residues were studied by potentiometric, UV-VIS and EPR spectroscopic methods. The stoichiometry of the major species formed in the copper(II)-GlyLys(Gly) system is [CuH-1L] and the EPR spectroscopic data indicate the existence of two isomeric forms of the complex. The nickel(II) and zinc(II)-GlyLys(Gly) systems have been characterised by the formation of stable [ML](+) complexes containing bis(NH2,CO) co-ordination and a macrochelate. Deprotonation and co-ordination of the amide groups were detected in the copper(II) and nickel(II) complexes. The co-ordination chemistry of Asp-epsilon-Lys is best described by the metal binding ability of the amino acid residues and it's high affinity for dimerisation. The stoichiometry of the dinuclear complexes can be given as [M2L2]. containing only amino acid binding sites. Deprotonation and co-ordination of the amide functions were suggested only in the copper(II)-containing systems, resulting in the formation of the [Cu2H-2L2](2-) dinuclear complex.
