Megjegyzések:The fragments of rat amylin rIAPP(17-29) (Ac-VRSSNNLGPVLPP-NH2), rIAPP(17-22)(Ac-VRSSNN-NH2), rIAPP(19-22) (Ac-SSNN-NH2) and rIAPP(17?20) (Ac-VRSS-NH2) togetherwith the related mutant peptides (Ac-VASS-NH2 and Ac-VRAA-NH2) have been synthesized and theircopper(II) complexes studied by potentiometric, UV-Vis, CD and EPR spectroscopic methods. Despitethe lack of any common strongly coordinating donor functions some of these fragments are able tobind copper(II) ions in the physiological pH range. The longest fragment rat amylin(17-29) keeps oneequivalent copper(II) ion in solution in the whole pH range, while two other peptides Ac-VRSSNN-NH2and Ac-SSNN-NH2 are also able to interact with copper(II) ions in the slightly alkaline pH range.According to the spectral parameters of the complexes, the peptides can be classified into two differentcategories: (i) the tetrapeptides Ac-VRSS-NH2, Ac-VASS-NH2 and Ac-VRAA-NH2 can interact withcopper(II) only under strongly alkaline conditions (pH > 10.0) and the formation of only one specieswith four amide nitrogen coordination can be detected; (ii) the peptides Ac-VRSSNNLGPVLPP-NH2,Ac-VRSSNN-NH2 and Ac-SSNN-NH2 can form complexes above pH 6.0 with the majorstoichiometries [CuH-2L], [CuH-3L]- and [CuH-4L]2-. These data support that rIAPP(17?29) caninteract with copper(II) ions under physiological conditions and the SSNN tetrapeptide fragment canbe considered as the shortest sequence responsible for metal binding. Density functional theory (DFT)calculations provide some information on the possible coordination modes of Ac-SSNN-NH2 towardsthe copper(II) ion and suggest that for [CuH-2L], [CuH-3L]- and [CuH-4L]2-, the binding of two, threeand four deprotonated amide nitrogens, with NH- of the side chain of asparagine as anchoring group,is probable. Moreover, these data reveal that peptides can be effective metal binding ligands even in theabsence of anchoring groups, if more polar side chains are present in a specific sequence.

Megjegyzések:The tetrapeptides Ac-SKHM-NH2, Ac-TKHMNH2,Ac-MKHS-NH2, Ac-S(OMe)KHM-NH2, and Ac-MKHS(OMe)-NH2 and the nonapeptides Ac-KTNSKHMAG-NH2and Ac-KTNMKHSAG-NH2 were synthesized and theircopper(II) complexes were studied by potentiometric,UV?Vis, circular dichroism (CD), and electron paramagneticresonance (EPR) spectroscopic methods. Thesepeptides mimic the 109?112 and 106?114 residues of thesequence of human prion protein. The imidazole-N donoratoms of histidyl residues were found to be the primarymetal binding sites of all peptide fragments. This bindingmode provides a good possibility for the cooperative deprotonationand metal ion coordination of two amidefunctions preceding histidine. The (Nim,N-,N-)-bondedspecies predominate in the pH range 5.5?7.0 and the freecoordination sites of these species make possible the metalbinding of weakly coordinating side chains. Thecomparison of the potentiometric and spectroscopic resultsrevealed the stabilizing role of the oxygen donors of seryl,threonyl, or methoxyseryl residues of Ac-SKHM-NH2, Ac-TKHM-NH2, Ac-S(OMe)KHM-NH2, and Ac-KTNSKHMAG-NH2 containing the mutations in position 109. Theseinteractions were, however, not observed in the peptidescontaining the specific amino acids in other locations of thepeptide sequence.

Megjegyzések:The hexa- and hepta-peptides CSSACS-NH2 and ACSSACS-NH2 have been synthesized by solid phasepeptide synthesis and their zinc(II) and cadmium(II) complexes studied by potentiometric, NMRspectroscopic and ESI MS techniques. Both peptides have outstanding zinc(II) and cadmium(II) bindingaffinity but their coordination chemistry is different. In the case of the hexapeptide, the amino terminusis the primary metal binding site in the form of a stable (NH2,S ) 5-membered chelate supported bymacrochelation via the distant cysteinyl residue. The heptapeptide ACSSACS-NH2 is a slightly lesseffective metal binder but its coordination chemistry is more versatile. The thiolate groups are theprimary binding sites for both metal ions and an 18-membered (S ,S ) macrochelate is the favoredcoordination mode of the peptide. In slightly basic samples the deprotonated amino group can alsocontribute to metal binding. Moreover, the interaction of the terminal amino-N and the thiolate-S ofthe Cys(2) moiety can promote the deprotonation and metal ion coordination of the amide groupbetween these residues. This reaction results in the formation of the (NH2,N ,S ) fused chelatessupported by the thiolate of the distant cysteinyl residue. Zinc(II) induced deprotonation andcoordination of amide groups have already been described in various peptides of histidine, but in thecase of cadmium(II) this is the first example for the formation of a Cd-N(peptide amide) bond.

Megjegyzések:Copper(II) and nickel(II) complexes of an octapeptide (Ac-MKHMGTHS-NH2) and its N-terminally lengthened derivatives (Ac-GGMKHMGTHS-NH2 and Ac-GDMKHMGTHS-NH2) have been studied by potentiometric and spectroscopic measurements. The amino acid sequences of the peptides correspond to the metal binding domains of 96H and 111H residues in the native prion fragments (GT96HS and MK111HM) but these domains are in the opposite order in the model peptides. The results revealed that both histidyl moieties can be effective metal binding sites. Evaluation of CD spectra made it possible to assess the ratio of coordination isomers of mononuclear complexes. The preference both for copper(II) and nickel(II) binding at the GTHS site was obtained in all cases. In the case of nickel(II) this is in a good agreement with the previous results reported for the prion fragments but the opposite distribution was obtained for copper(II). These data strongly support that nickel(II) prefers the binding to the specific sites of peptides best suited for square planar coordination, while the overall stability of copper(II) peptide complexes is finely tuned by the secondary effects of distant side chains. Moreover, the formation of heteropolynuclear copper(II)-nickel(II)-peptide complexes has also been detected and it was found that the addition of nickel(II) to the mononuclear copper(II) species can redistribute the coordination of copper(II) among the available metal binding sites.

Megjegyzések:Nickel(II), cobalt(II) and cadmium(II) complexes of terminally protected multihistidine peptides including Ac-HGH-OH, Ac-HGH-NHMe, Ac-HHGH-OH, Ac-HAHVH-NH2, Ac-HVHGH-NH2, Ac-HGHVH-NH2 and Ac-(His-Sar)(n)-His-NH2 (n = 1, 2 or 3) were studied by potentiometric, UV-Vis, CD and H-1 NMR spectroscopic techniques. It was found that the complexes in which the histidine imidazole nitrogens coordinate with ML stoichiometry are the main species in the physiological pH-range in all cases. The stability of these complexes is determined by the number of bound imidazole rings, the presence of the carboxylate group and the quality of the metal ion centre. The larger the number of coordinated imidazole-N donor atoms, the higher the stability of the complex. The stability constants of the ML complexes follow the Ni(II) > Co(II) similar to Cd(II) order. Cobalt(II) and cadmium(II) are not, but nickel(II) is able to promote the deprotonation and the coordination of amide nitrogens and NiH_L-2 and NiH_L-3 (Ni2H_L-4) species predominate in basic solutions. For the pentapeptides with the exception of the sarcosine containing ligand the presence of coordination isomers is supported by spectroscopic methods. These data reveal that the favoured isomers are coordinated on the C-termini, but the ratio of isomers depends on the sequence of peptides.
