Megjegyzések:White oaks (Quercus section, Quercus subgenus) are widely distributed in Europe. Quercus petraea (sessile oak), an economically important species is predicted to be affected by climate change. Q. pubescens (pubescent oak) and Q. virgiliana (Italian pubescent oak) are economically less important, drought tolerant species. Frequent hybridization of white oaks was observed and currently the introgression of Q. pubescens and Q. virgiliana in non-mediterranean regions of Europe has been reported. Our goal was to use tissue cultures established from individual trees of the above taxa and their putative hybrids, all present in the forest stand of Síkfőkút LTER Research Area (NE Hungary) as simple experimental model systems for studying drought/osmotic stress tolerance. Tissue cultures are more suitable models for such studies, than seedlings, because they are genetically identical to the parent plants. Polyethylene glycol (PEG6000) treatments were used for this purpose. The identification of taxa was based on leaf morphological traits and microsatellite analysis and showed that Q. petraea is genetically distinct to all other taxa examined. We established six callus lines of Quercus. As expected, in Q. petraea cultures PEG6000 induced severe loss of fresh weight and the ability to recover after removal of the osmoticum, which was not characteristic for Q. pubescens and Q. virgiliana. Putative hybrids exhibited an intermediate response to osmotic stress. Activity gels showed the increase of single-strand preferring (SSP) nuclease and no significant change of guaiacol-peroxidase activities in drought-sensitive genotypes/cultures and no significant increase of SSP nuclease activities accompanied with increases of guaiacol-peroxidase activities in drought-tolerant ones. This indicates that drought/osmotic stress tolerance is associated to increased capacity of scavenging reactive oxygen species and hence less susceptibility to DNA damage. Our results confirm that tissue cultures of oak are suitable model systems for studying drought/osmotic stress responses.

Megjegyzések:Crocus heuffelianus belongs to the C. vernus(Iridaceae) species aggregate. In the Carpathian Basin andparticularly in Hungary it is considered an endangered spe-cies. Therefore our aim was to establish a tissue culturesystem with potential of germplasm preservation of thistaxon. For in vitro culture experiments, shoot primordia fromcorms were the most suitable. We induced an embryogeniccallus line from those explants on basal Murashige-Skoog(MS) medium supplemented with Gamborg's vitamins, 2%(w/v) sucrose, 10 mg l-1(53.7 lM) a-naphthaleneaceticacid (NAA) and 1 mg l-1(4.44 lM) 6-benzyladenine (BA).Globular stage embryos developed on this medium andseveral culture conditions were used in an attempt to obtainmature embryos and plant regeneration. Firstly a decrease ofauxin/cytokinin concentration and ratio, then secondly adecrease in the strength of culture medium and the concen-tration of carbon source was used, which was effective inembryogenesis and the production of plants. Regenerationmedium used in the second step was fourfold diluted MSmedium and Gamborg's vitamins supplemented with 1%(w/v) sucrose, 0.05 mg l-1(0.26 lM) NAA and 0.5 mg l-1(2.22 lM) BA, with a 14/10 h photoperiod. Under theseconditions we could detect all the stages of somatic embryodevelopment characteristic for Iridaceae. This is the?rst report demonstrating the production of stable tissueculture of C. heuffelianus with potential use in germplasmpreservation via plant regeneration. This study could alsocontribute to a better understanding of somatic embryogen-esis in the Crocus genus.

Megjegyzések:We present a list of tissue culture systems developed at the Department of Botany,University of Debrecen. Several of tissue cultures were developed for the first time in our laboratory. These include micropropagation of Elatine hungarica, callus cultures of several oak genotypesof Hungarian origin and of Crocus species characteristic for the Carpathian Basin. Calluscultures were either organogenic (e.g. Crocus scepusiensis) or embryogenic (Quercus petraea,Galanthus and Crocus species, Phragmites australis). In case of embryogenic cultures, somaticembryos showed either normal, bipolar development (e.g. Crocus heuffelianus, Phragmitesaustralis) or they have lost this bipolarity at the maturation of embryos (C. banaticus, C. sativus,Galanthus). The type of callus is yet to be identified for several cultures (e.g. Plantago lanceolata,Vicia faba). Most of our in vitro cultures proved to have plant regeneration potential. Severalof them derived from endangered/ red list plants (Crocus species, Elatine hungarica, Galanthusnivalis), therefore they are suitable for germplasm preservation. Others (Quercus, Phragmitesaustralis) proved to be suitable for stress physiology and/ or cell biology experiments. Culturessuch as Crocus sativus or Plantago lanceolata derived from plants of medicinal importance, therefore are of potential pharmacological use.
