Megjegyzések:Although hybridization has long been recognized as a major force driving speciation in land plants, it has not yet been evidenced in Astragalus, the largest angiosperm genus. Here, we reveal the possible contribution of hybridization to speciation in Astragalus by employing cloning of the nrDNA ITS region and sampling three plastid regions (ycf1, ndhF?rpl32, and rpl32?trnL) in taxa belonging to sect. Dissitiflori. Phylogenetic network and tree analyses uncovered various levels of intra-individual and intraspecific polymorphism of ITS in most of the taxa investigated. Two distantly related ribotype groups were found to be shared by the closely related polyploids Astragalus pallescens M.Bieb., Astragalus peterfii Jáv., and Astragalus pseudoglaucus Klokov suggesting ancient hybridization followed by incomplete lineage sorting (i.e., shared ancestral polymorphism) in nrDNA ITS. Reticulation is also invoked as an underlying evolutionary process behind the statistically highly supported incongruent placement of A. pseudoglaucus and Astragalus vesicarius subsp. pastellianus (Pollini) Arcang. in nuclear versus plastid phylogenies. The phylogenetic results also shed light on taxonomic controversies in the section, such as the false synonimization ofA. peterfiiunderA. vesicarius s.l. Our results provide evidence for the (at least past) existence of speciation processes driven by hybridization in Astragalus.

Megjegyzések:Nymphaea lotus L. is a tropical and subtropical species of waterlilies with an African distribution. A conspicuous satellite occurrence can be found in Europe in a thermal spring in NW Romania. This population (treated as var. thermalis (DC.) Tuzson) received much attention as a potential Tertiary relict in the flora of Europe, although its relict vs. planted nature has been part of great debate among scholars during the last 200 years. We revisit this question by using molecular phylogenetic methods to estimate the timing of divergence of this species, and put this population into a phylogeographic context by comparing it to samples coming from the whole area of the species. By using sequences of the nuclear and the plastid genome, we reconstructed the phylogeographic relationships within N. lotus with a haplotype network building approach, that identified a genetically distant lineage in western Africa, which we relate to N. zenkeri Gilg, the sister species of N. lotus. All the populations of N. lotus s.str. displayed genetic distance between each other except for the Egyptian and Romanian populations. On our dated phylogeny of the subgenus Lotus the separation between N. lotus and its sister species was found to be younger than the end of the Tertiary (1.26 Mya). These results clearly refute the Tertiary relict status of the European population of N. lotus.

Megjegyzések:Background and Aims The terrestrial orchid genus Epipactis has become a model system for the study of speciation via transitions from allogamy to autogamy, but close phylogenetic relationships have proven difficult to resolve through Sanger sequencing. Methods We analysed with restriction site-associated sequencing (RAD-seq) 108 plants representing 29 named taxa that together span the genus, focusing on section Epipactis. Our filtered matrix of 12 543 single nucleotide polymorphisms was used to generate an unrooted network and a rooted, well-supported likelihood tree. We further inferred genetic structure through a co-ancestry heat map and admixture analysis, and estimated inbreeding coefficients per sample. Key Results The 27 named taxa of the ingroup were resolved as 11 genuine, geographically widespread species: four dominantly allogamous and seven dominantly autogamous. A single comparatively allogamous species, E. helleborine, is the direct ancestor of most of the remaining species, though one of the derived autogams has generated one further autogamous species. An assessment of shared ancestry suggested only sporadic hybridization between the re-circumscribed species. Taxa with the greatest inclination towards autogamy show less, if any, admixture, whereas the gene pools of more allogamous species contain a mixture alleles found in the autogams. Conclusions This clade is presently undergoing an evolutionary radiation driven by a wide spectrum of genotypic, phenotypic and environmental factors. Epipactis helleborine has also frequently generated many local variants showing inclinations toward autogamy (and occasionally cleistogamy), best viewed as incipient speciation from within the genetic background provided by E. helleborine, which thus becomes an example of a convincingly paraphyletic species. Autogams are often as widespread and ecologically successful as allogams.
