Megjegyzések:Spiraea crenata L. is a deciduous shrub distributed across the Eurasian steppe zone. The species is of cultural and horticultural importance and occurs in scattered populations throughout its westernmost range. Currently, there is no genomic information on the tribe of Spiraeeae. Therefore we sequenced and assembled the whole genome of S. crenata using second- and third-generation sequencing and a hybrid assembly approach to expand genomic resources for conservation and support research on this horticulturally important lineage. In addition to the organellar genomes (the plastome and the mitochondrion), we present the first draft genome of the species with an estimated size of 220 Mbp, an N50 value of 7.7 Mbp, and a BUSCO score of 96.0%. Being the first complete genome in tribe Spiraeeae, this may not only be the first step in the genomic study of a rare plant but also a contribution to genomic resources supporting the study of biodiversity and evolutionary history of Rosaceae.

Megjegyzések:Different versions of Restriction-site-Associated DNA sequencing (RADseq) have become powerful and popular tools in molecular ecology. Although RADseq datasets are generally regarded as representative of the nuclear genome, reduced representation genomic libraries may also sample the organellar (i.e. the mitochondrial and, in the case of plants, the plastid) DNA. This cytoplasmic genetic variance provides a better understanding of evolutionary history by uncovering past hybridisation and identifying maternal or, rarely, paternal lineage due to rapid lineage sorting. We developed a pipeline that is based on existing bioinformatic tools to automatically mine and genotype organellar loci from RADseq libraries. The efficacy of our pipeline is tested on eight publicly available datasets spanning different phylogenetic levels (i.e. from family-level phylogenies to phylogeography) and RADseq methods (sdRAD, ddRAD, ezRAD, GBS) for genotyping mitochondrial and plastid loci, which were subject to phylogenetic tree reconstruction. In all cases, organellar phylogenies adequately supplemented the original studies by corroborating the large-scale picture based on RADseq or by bringing additional evidence on past or contemporary hybridisation. RADseq methods designed to achieve larger horizontal coverage (i.e. ddRAD, ezRAD) yielded longer organellar alignments, but sdRAD and GBS still provided valuable polymorphic organellar loci at no additional sequencing effort.

Megjegyzések:At the westernmost distribution of the steppe herbaceous plant, Galatella villosa, in Hungary, Serbia and Ukraine, we recently observed intermediate specimens between this species and its close relative, G. linosyris. We were able to demonstrate the hybrid origin of these individuals by sequencing the biparentally inherited nuclear ribosomal internal transcribed spacer (nrITS) region and checking additive polymorphism in the hybrids. In addition, examination of the maternally inherited plastid regions (trnH-psbA and trnL-trnF intergenic spacers) revealed that G. villosa is likely to be the maternal parent in the Hungarian and Ukrainian populations and G. linosyris in the Serbian population. The intermediate forms produced only sterile seeds. The alleged hybrid between the above two species has already been described as G. xsubvillosa based on a very brief diagnosis. Still, the analysis of the morphological characters using linear discriminant analyses clearly separated the holotype of G. xsubvillosa from individuals of G. linosyris x G. villosa. The latter appeared to be morphologically intermediate between populations of G. villosa and G. linosyris. Contrary to the originally stated hybrid origin of the type plants of G. xsubvillosa, morphological evidence indicates the involvement of G. divaricata not G. linosyris. The hybrid G. linosyris x G. villosa is thus described here, as a new nothospecies G. xfeketegaborii. This study highlights the power of easily available molecular phylogenetic tools for demonstrating the hybrid origin of plants and illustrates how additive polymorphism can be distinguished from other types of intraindividual polymorphism in nuclear DNA sequences.

Megjegyzések:For the effective control of an invasive species, gathering as much information as possible on its ecology, establishment and persistence in the affected communities is of utmost importance. We aimed to review the current distribution and characteristics of Sporobolus cryptandrus (sand dropseed), an invasive C4 grass species of North American origin recently discovered in Hungary. We aimed to provide information on (i) its current distribution paying special attention to its invasion in Eurasia; (ii) the characteristics of the invaded habitats in Central Europe; (iii) seed bank formation and germination characteristics, crucial factors in early establishment; and (iv) the effects of its increasing cover on vegetation composition. Finally, we aimed to (v) point out further research directions that could enable us to understand the invasion success of this potential invasive species. Field surveys uncovered large stands of the species in Central and Eastern Hungary with most of the locations in the former, especially in the Kiskunsag region. The species invaded disturbed stands of dry and open sand grasslands, closed dune slack grasslands and it also penetrates natural open sand grasslands from neighboring disturbed habitats. Increasing cover of Sporobolus cryptandrus was associated with a decline in species richness and abundance of subordinate species both in the vegetation and seed banks, but a low density of Sporobolus cryptandrus can even have a weak positive effect on these characteristics. Viable seeds of Sporobolus were detected from all soil layers (2.5 cm layers measured from the surface to 10 cm in depth), which indicates that the species is able to form a persistent seed bank (1114 to 3077 seeds/m(2) with increasing scores towards higher abundance of the species in vegetation). Germination of Sporobolus cryptandrus was negatively affected by both litter cover and 1 cm deep soil burial. To sum up, Sporobolus cryptandrus can be considered as a transformer invasive species, whose spread forms a high risk for dry sand and steppe grasslands in Eurasia. We can conclude that for the effective suppression of the species it is necessary: (i) to clarify the origin of the detected populations; (ii) to assess its competitive ability including its potential allelopathic effects; (iii) to assess its seed bank formation potential in habitats with different abiotic conditions; and (iv) to assess the possibility of its suppression by natural enemies and management techniques such as mowing or livestock grazing.
