Megjegyzések:The plausibility of sympatric speciation is still debated despite increasing evidence, such as host races in insects. This speciation process may be occurring in the case of the two phenological forms of the obligatorily myrmecophilous Phengaris arion. The main goal of our research was to study the nature and causes of difference between these forms focusing primarily on the incipient speciation via host races. Molecular analyses based on highly variable microsatellites together with Wolbachia screening, male genitalia morphometrics and host ant studies were carried out on four syntopic sample pairs. Our results show that the two phenological forms of P. arion may meet the criteria for host plant races. They coexist in sympatry in certain parts of the species range which is allowed by the adaptation to the distinct phenology of the host plants. Negative selection acts against the intermediate individuals which are on the wing in the inappropriate time frame. Thus, disruptive selection affects and produces bimodal distributions of phenotypes. However, the phenology of food plants is not entirely distinct and fluctuates year by year. Therefore, the two forms can exchange genes occasionally depending on the length of the time slot when they can meet with each other. Consequently, the reproductive isolation could not be completed and the existence of the two arion forms may represent only an incipient stage of sympatric speciation. It is also clear that Wolbachia is likely not a driver of sympatric speciation in this case.

Megjegyzések:Fifteen new microsatellite markers for the beetle Lethrus apterus were developed and tested in 45 specimens from the North Hungarian Mountains. Fourteen of the markers developed were polymorphic, and the number of alleles per locus ranged from two to nine. The observed and expected heterozygosities of the polymorphic markers ranged from 0.178 to 0.578, and 0.201 to 0.698, respectively. One locus showed significant deviation from the Hardy-Weinberg equilibrium, probably due to null alleles. The primers were tested on four other Lethrus species (L. bituberculatus, L. scoparius, L. strymonensis and L. perun) and six other coleopteran species (Copris hispanus, Geotrupes stercorarius, Melolontha melolontha, Onthophagus taurus, Oryctes nasicornis and Protaetia affinis). Thirteen loci showed cross-amplification in the Lethrus species and only three loci could be amplified in some of the six other coleopteran species. The developed markers will be valuable in investigating the population genetic structure, behaviour and reproductive biology of L. apterus.

Megjegyzések:Insulin/insulin-like growth factor signaling (IIS) is thought to be a central mediator of life history traits, but the generality of its role is not clear. Here, we investigated mRNA expression levels of three insulin-like peptide genes, the insulin-like receptor htk7, as well as several antioxidant genes, and the heat-shock protein hsp70 in the freshwater cnidarian Hydra vulgaris. Hydra polyps were exposed to a combination of different levels of food and perceived population density to manipulate life history traits (asexual reproduction and oxidative stress tolerance). We found that stress tolerance and the rate of asexual reproduction increased with food, and that these two effects were in significant interaction. Exposing animals to high perceived density resulted in increased stress tolerance or reduced reproduction only on lower food levels, but not on high food. The insulin-like receptor htk7 and the antioxidant gene catalase were significantly upregulated in the high density treatments. However, the expression level of insulin-like peptide genes, most antioxidant genes, and hsp70 were not affected by the experimental treatments. The higher expression level of htk7 may suggest that animals maintain a higher level of preparedness for insulin-like ligands at high population densities. However, the lack of difference between food levels suggests that IIS is not involved in regulating asexual reproduction and stress tolerance in hydra, or that its role is more subtle than a simple model of life history regulation would suggest.
