Megjegyzések:The ontogeny of the interleukin 2 receptor (IL 2 R) expression and of the IL 2 responsiveness has been investigated in the rat thymus. In tissue sections, IL 2 R-bearing cells were first detected at day 16 of gestation using the anti-IL 2 R mAb ART-18. In contrast to mice, IL 2 R-bearing cells of the rat are localized mainly in the thymic medulla from the first day of the corticomedullary compartimentalization, and not in the cortex. They are found in regions with a high expression of MHC class II antigens. The proportion of IL 2 R-bearing cells increases during gestation, reaches a peak at the first day after birth and declines to the adult level in the following weeks. The appearance of medullary localized IL 2 R-bearing cells is paralleled with the capacity of the thymocytes to proliferate in response to IL 2 without any additional stimuli.

Megjegyzések:Monoclonal antibodies were raised to a crude photoreceptor-membrane suspension from chicken retinas. Clones producing antibodies against cone outer segments were selected by screening with immunocytochemistry on semithin sections of the retina. One monoclonal antibody, called COS-1, specifically labelled outer segments of double cones and one type of single cones; outer segments of rods and several single cones were not stained. On immunoblots of retinal photoreceptor membranes, this antibody recognized a protein with an apparent molecular mass of 33,000. The visual pigment character of the 33,000 protein was indirectly established by another monoclonal antibody, OS-2, which labelled all outer segments on semithin sections and four bands (33,000-, 36,000-, 38,000-40,000- and a composite band between 66,000-72,000 MW) on immunoblots. Of these, the 36,000- and the 72,000 MW protein bands were identified with an anti-rhodopsin polyclonal antibody as rhodopsin monomer and dimer. Monoclonal antibody OS-2 is assumed therefore to represent an antibody against a common epitope of all visual pigments of the chicken. The monoclonal antibody COS-1 was found to bind to certain cone outer segments of many other vertebrate species as well.

Megjegyzések:During embryonic development of the mouse, before expressing classical T cell markers, the blast cells colonizing the thymus react with the monoclonal antibody AMT-13 shown previously to detect interleukin 2 receptors. The proportion of AMT-13+ cells decreases as gestation time increases. On the other hand, the proportion of Thy-1+, Lyt-1+ and Lyt-2+ cells increases during ontogenesis. On the 19th day of gestation when the thymus architecture is comparable to the adult thymus, the AMT-13+ cells become localized in the subcapsular area of the cortex. In the adult thymus after cortison treatment the regenerating cells express the AMT-13 antigen. The AMT-13 antigen presumably the interleukin 2 receptor is the first marker of the early embryonic thymocytes reported until now that may be related to cellular function.

Megjegyzések:Recently, species specific monoclonal antibodies, directed against the interleukin-2 (IL-2) receptor of murine, rat and human origin, have been produced. In this study we demonstrate with immunohistological methods that cells reacting with these antibodies are present in normal primary and secondary lymphatic organs. The cells are exclusively localized in T cell-dependent areas, and their number increased as a result of immunization.

Megjegyzések:Recently we observed the expression of MHC class II antigens on bile duct epithelium in human liver transplantation rejection. In this report we present evidence for the expression of MHC class II antigens on bile duct epithelium in experimental GVHD. These findings support the idea, that the target tissue elements in liver transplantation rejection and GVHD are the bile ducts, on the other hand this is a new experimental model for the analysis of MHC class II antigen expression on epithelial tissue.

Megjegyzések:The proportion and anatomical localization in murine thymus of T cell subpopulations, including those which are defined by the monoclonal anti-interleukin 2 (anti-IL-2) receptor antibody AMT-13, were studied by flow cytofluorometry and by immunohistochemical methods, both in irradiated and in normal mice. As a consequence of irradiation the proportion of AMT-13 positive cells and that of Lyt-1 positive cells were markedly enhanced, while the proportion of Lyt-2 positive cells was reduced. The vast majority of the AMT-13 positive cells both in normal and in irradiated thymi were located in the subcapsular area of the thymic cortex, whereas the irradiation resistant Lyt-1 positive cells were located in the medulla. These findings are compatible with the view that, similar to the developing thymus in the mouse embryo, in the regenerating adult thymus, AMT-13+ cells include the activated pro-thymocytes that repopulate the irradiated thymus.

Megjegyzések:IL-2R alpha-chain is expressed on a subset of mouse CD4- and CD8-, double negative (DN) thymocytes. This expression of IL-2R alpha-chain on some DN thymocytes in the mouse has led to the proposal that IL-2 might serve as a principal growth and/or differentiation factor for immature thymocytes. However, previous histologic observations have indicated that IL-2R alpha-chain is not expressed on the subcapsular thymic blasts (an area rich in DN cells) in either huma or rat thymus, whereas all three species display IL-2R expression on a few cells in the thymic medulla. Therefore, we characterized rat DN thymocytes to determine whether they contained an IL-2R+ population. The results show that rat thymic DN cells share several characteristics with mouse DN cells. However, most of the rat strains do not express the IL-2R on DN cells as shown either by immunofluorescence or by IL-2 binding and receptor cross-linking. Thus, the rare medullary IL-2R+ cells were not found in the DN cells. Only in the exceptional F344 rat strain is the IL-2R alpha-chain expressed on a major proportion of thymocytes, including both DN cells and small cortical-type thymocytes. Furthermore, rat DN cells do not contain detectable IL-2 mRNA or cytoplasmic IL-2 activity, thus supporting the conclusion that it is unlikely that IL-2 and IL-2R serve to maintain the proliferation of rat DN thymocytes in vivo. The possible significance of in vivo expression of IL-2R alpha-chain on immature thymocytes in the mouse and in a single rat strain is discussed.
