Megjegyzések:In addition to producing sebum, sebocytes link lipid metabolism with inflammation at a cellular level and hence, greatly resemble adipocytes. However, so far no analysis was performed to identify and characterize the adipocyte-associated inflammatory proteins, the members of the adipokine family in sebocytes. Therefore, we determined the expression profile of adipokines (adiponectin, interleukin [IL] 6, resistin, leptin, serpin E1, visfatin, apelin, chemerin, retinol-binding protein 4 [RBP4] and monocyte chemoattractant protein 1 [MCP1]) in sebaceous glands of healthy and various disease-affected (acne, rosacea, melanoma and psoriasis) skin samples. Sebaceous glands in all examined samples expressed adiponectin, IL6, resistin, leptin, serpin E1 and visfatin, but not apelin, chemerin, RBP4 and MCP1. Confirming the presence of the detected adipokines in the human SZ95 sebaceous gland cell line we further characterized their expression and secretion patterns under different stimuli mimicking bacterial invasion (by using Toll-like receptor [TLR] 2 and 4 activators), or by 13-cis retinoic acid (13CRA; also known as isotretinoin), a key anti-acne agent. With the exception of resistin, the expression of all of the detected adipokines (adiponectin, IL6, leptin, serpin E1 and visfatin) could be further regulated at the level of gene expression, showing a close correlation with the secreted protein levels. Besides providing further evidence on similarities between adipocytes and sebocytes, our results strongly suggest that sebocytes are not simply targets of inflammation but may exhibit initiatory and modulatory roles in the inflammatory processes of the skin through the expression and secretion of adipokines.

Megjegyzések:Epidermal energy metabolism is relevant to skin physiology, aging, and photodamage. While selected hormones stimulate epidermal keratinocyte mitochondrial activity, its negative regulation remains unknown. In several cell types, cannabinoid receptor 1 (CB1) is expressed both in the cell membrane (cmCB1), and in the mitochondrial outer membrane (mtCB1), where its stimulation directly suppresses mitochondrial functions. In the current pilot study, we investigated if CB1 is a negative regulator of human epidermal energy metabolism under physiological conditions. Using organ-cultured full-thickness human skin specimens of healthy individuals, we showed that antagonizing the homeostatic CB1 signaling by the administration of the CB1 inverse agonist AM251 increased activity of respiratory chain complex I and II/IV activity in a CB1-dependent manner, since the CB1-selective agonist arachidonyl-2'-chloroethylamide could prevent the effect. Moreover, the phenomenon was also reproduced by siRNA-mediated down-regulation of CB1. As revealed by the unaltered expression of several relevant markers (TFAM, VDAC1, MTCO1, and NDUFS4), modulation of CB1 signaling had no effect on the epidermal mitochondrial mass. Next, by using immunoelectron microscopy, we found that human epidermal keratinocytes express both cmCB1 and mtCB1. Finally, by using equipotent extracellularly-restricted (hemopressin) as well as cell-permeable (AM251) inverse agonists, we found that mitochondrial activity is most likely exclusively regulated by mtCB1. Thus, our data identify mtCB1 as a negative regulator of keratinocyte mitochondrial activity in intact human epidermis, and highlight the question, whether topical therapeutic interventions capable of selectively activating mtCB1 can reduce excessive mitochondrial ROS production resulting from dysregulated mitochondrial activity during skin aging or photodamage.

Megjegyzések:Acne is a common skin disease characterized by elevatedsebum production and inflammation of the sebaceous glands. Wehave previously shown that a non-psychotropic phytocannabinoid((?)-cannabidiol [CBD]) exerted complex anti-acne effects bynormalizing ♭pro-acne agents'-induced excessive sebaceous lipidproduction, reducing proliferation and alleviating inflammation inhuman SZ95 sebocytes. Therefore, in this study we aimed to explorethe putative anti-acne effects of further non-psychotropic phytocannabinoids((?)-cannabichromene [CBC], (?)-cannabidivarin[CBDV], (?)-cannabigerol [CBG], (?)-cannabigerovarin [CBGV]and (?)-D9-tetrahydrocannabivarin [THCV]). Viability andproliferation of human SZ95 sebocytes were investigated by MTTand CyQUANT assays; cell death and lipid synthesis weremonitored by DilC1(5)-SYTOX Green labelling and Nile Redstaining, respectively. Inflammatory responses were investigated bymonitoring expressions of selected cytokines uponlipopolysaccharide treatment (RT-qPCR, ELISA). Up to 10 lM, thephytocannabinoids only negligibly altered the viability of thesebocytes, whereas high doses (?50 lM) induced apoptosis.Interestingly, basal sebaceous lipid synthesis was differentiallymodulated by the substances: CBC and THCV suppressed it, andCBDV had only minor effects, whereas CBG and CBGV increased it.Importantly, CBC, CBDV and THCV significantly reducedarachidonic acid (AA)-induced ♭acne-like' lipogenesis. Moreover,THCV suppressed proliferation, and all phytocannabinoids exertedremarkable anti-inflammatory actions. Our data suggest that CBGand CBGV may have potential in the treatment of dry-skinsyndrome, whereas CBC, CBDV and especially THCV showpromise to become highly efficient, novel anti-acne agents.Moreover, based on their remarkable anti-inflammatory actions,phytocannabinoids could be efficient, yet safe novel tools in themanagement of cutaneous inflammations.

Megjegyzések:Polyols (e.g. glycerol, xylitol) are implicated as moisturizers of the skin and other epithelial tissues. However, we lack information about their exact cellular mechanisms and their effects on the gene expression profiles. Therefore, in this study, we aimed at investigating the effects of glycerol and xylitol on human epidermal keratinocytes. The polyols (identical osmolarities; xylitol: 0.0045%-0.45%; glycerol: 0.0027%-0.27%)did not alter cellular viability or intracellular calcium concentration. However, they exerted differential effects on the expression of certain genes and signalling pathways. Indeed, both polyols up-regulated the expression of filaggrin, loricrin, involucrin and occludin; yet, xylitol exerted somewhat more profound effects. Moreover, while both polyols stimulated the MAPK pathway, only xylitol induced the activation-dependent translocation of protein kinase C?, a key promoter of epidermal differentiation. Finally, in various keratinocyte inflammation models, both polyols (albeit with different efficacies) exerted anti-inflammatory effects. Taken together, these data strongly suggest that glycerol and xylitol differentially modulate expressions of multiple genes and activities of signalling pathways in epidermal keratinocytes. Thus, our findings invite clinical trials to explore the applicability and the impact of a combined glycerol-xylitol therapy in the management of various skin conditions.

Megjegyzések:Echinacea purpurea extracts (purple coneflower) are known to have immunemodulatory effects. Several alkamides, as the major lipophilic constituents, bind to cannabinoid receptors 1 and 2. Since the endocannabinoid system is of importance in inflammatory skin diseases, anti-inflammatory activity of alkamides was investigated. Therefore the new lipophilic root extract of Echinacea purpurea was tested in cultured human keratinocytes. A significant (P < 0.05) reduction of lipoteichoic acid-induced mRNA expression of IL-1a, IL-1b, and IL-6 was observed. Moreover, the mixture was also able to reduce expression of IL-8 and showed significant anti-inflammatory effects in vitro. The overall impact of the new developed water-in-oil (W/O) emulsion containing the new lipophilic root extract of Echinacea purpurea was investigated in different clinical studies. Long-term efficacy and safety of the Echinacea purpurea root extract (W/O) emulsion was evaluated in a 3 month half-side trial against comparator (30.2 15.9 years; n = 60). The emulsions were applied at least twice daily on two comparable and contralateral located skin areas on the crooks of arms, hollow of the knees, on the trunk, on the wrist or on the shin with slight lesions of atopic dermatitis. Erythema reduced significantly after 1, 2 and 3 months after application of Echinacea purpurea root extract (W/O) emulsion, as well as comparator. Interestingly, Echinacea purpurea root extract (W/O) emulsion is superior after prolonged application, indicated by a significant difference to comparator after 2 and 3 months. A significant reduction of pruritus could be measured after 1, 2 and 3 months. Furthermore Echinacea purpurea root extract (W/O) emulsion was significant superior compared to comparator after 2 months and nearly significant after 3 months. The same was observed for the local SCORAD. A significant reduction could be measured after 1, 2 and 3 months. Furthermore Echinacea purpurea root extract (W/O) emulsion was significant superior compared to comparator after 3 months. To gain further insight in underlying mechanisms, electron microscope analyses of the skin barrier as well as the lipid analysis by HPTLC will be performed and data will be presented. In summary, application of an Echinacea purpurea root extract (W/O) emulsion reduced significantly the local SCORAD, erythema and pruritus without irritation, very likely by improved functions of the epidermal barrier.

Megjegyzések:Fluoxetine is a safe antidepressant with remarkable anti-inflammatory actions; therefore, we aimed to investigate its effects on immortalized (HaCaT) as well as primary human epidermal keratinocytes in a polyinosinic-polycytidylic acid (p(I:C))-induced inflammatory model. We found that a non-cytotoxic concentration (MTT-assay, CyQUANT-assay) of fluoxetine significantly suppressed p(I:C)-induced expression and release of several pro-inflammatory cytokines (Q-PCR, cytokine array, ELISA), and it decreased the release of the itch mediator endothelins (ELISA). These effects were not mediated by the inhibition of the NF-?B or p38 MAPK pathways (western blot), or by the suppression of the p(I:C)-induced elevation of mitochondrial ROS production (MitoSOX Red labeling). Instead, unbiased activity profiling revealed that they were most likely mediated via the inhibition of the phosphoinositide 3-kinase (PI3K) pathway. Importantly, the PI3K-inhibitor GDC0941 fully mimicked the effects of fluoxetine (Q-PCR, ELISA). Although fluoxetine was able to occupy the binding site of GDC0941 (in silico molecular docking), and exerted direct inhibitory effect on PI3K (cell-free PI3K activity assay), it exhibited much lower potency and efficacy as compared to GDC0941. Finally, RNA-Seq analysis revealed that fluoxetine deeply influenced the transcriptional alterations induced by p(I:C)-treatment, and exerted an overall anti-inflammatory activity. Collectively, our findings demonstrate that fluoxetine exerts potent anti-inflammatory effects, and suppresses the release of the endogenous itch mediator endothelins in human keratinocytes, most likely via interfering with the PI3K pathway. Thus, clinical studies are encouraged to explore whether the currently reported beneficial effects translate in vivo following its topical administration in inflammatory and pruritic dermatoses.
