Megjegyzések:Free amino acid and free D-amino acid contents of milksamples with different microorganism numbers and composition of dairyproducts produced from them were examined. Total microorganism num-ber of milk samples examined varied from 1.25-106 to 2.95-106. It wasestablished that with an increase in microorganism number concentrationof both free D-amino acids and free L-amino acids increased, however,increase in D-amino acid contents was bigger considering its proportion.There was a particularly significant growth in the microorganism num-ber range of 1.5-106 to 2.9-106. Based on analysis of curds and cheesesamples produced using different technologies we have come to the con-clusion that for fresh dairy products and for those matured over a short time there was a close relation between total microorganism number andfree D-amino acid and free L-amino acid contents. At the same timeit was found that the ratio of the enantiomers was not affected by thetotal microorganism number. For dairy products, however, where aminoacid production capability of the microbial cultures considerably exceedsproduction of microorganisms originally present in the milk raw material,free amino acid contents of the milk product (both D- and L-enantiomers)seem to be independent of the composition of milk raw material.

Megjegyzések:Free amino acid and total amino acid content, water solublevitamin content (vitamin C, B1, B2, B6 and B12) and utilizable lysine,lysinoalanine and hydroxymethyl furfurol content of milk were examinedafter the samples were pasteurized using different heat treatment.They analyzed the effect of microwave treatment on the amino acids,free amino acid content and biological value compared to the conventionalheat treatment technology. In was established that the two applied heattreatments caused practically no change in the amino acid compositionof the milk protein neither in case of the essential nor in the case ofthe non-essential amino acids. The total free amino acid content of theraw milk (20.67 mg/100 g milk) reduced in the milk pasteurized in thetraditional way to 8.02, whereas in the microwave pasteurized milk to 8.96mg amino acid/100 g milk.

Megjegyzések:Performic acid oxidation of cysteine and methionine result-ing in the formation of cysteic acid and methionine sulphon has beenapplied in order to avoid the loss of these two sulphur containing aminoacids during the acidic hydrolysis of proteins that is necessary prior toamino acid analysis. The aim of the research was assigned by the increas-ing demand for the determination of the amount amino acid enantiomers:the applicability of performic acid oxidation was evaluated in this pointof view. Racemization of L-cysteine and L-methionine was found not significant during oxidation with performic acid, therefore this processcan be applied before hydrolysis during quantification of cysteine andmethionine enantiomers. Additionally, the quantification of cysteic acidand methionine sulphon enantiomers was accomplished in the form oftheir diastereoisomer derivatives via the development of a reversed phasehigh performance liquid chromatography method.

Megjegyzések:Six different dairy breeds kept under the same farm and fedunder the same conditions were evaluated with respect to the fat content and the fatty acid composition of their milk drawn on the 3-5th months of lactation.The fat content of the milk of the breeds did not differ significantly (p = 0.076) owing to the high variance of samples in the examined time period. The mean value of the sum of saturated fatty acids was the highest in the case of Jersey (77.8%) while this value ranged from 71.8% to 74.8% for the other breeds. An inversely proportional tendency can be seen for the ratio of monounsaturated fatty acids: in this case, the meanvalue for Jersey was the lowest (18.9%) and the values of the other breeds ranged from 21.9 to 24.9%. The saturated fatty acid/unsaturated fatty acid ratio was significantly (p < 0.05) higher in the milk fat of Jerseythan that of Swedish red, Holstein-Friesian, Norwegian red, and Brown Swiss. The n6/n3 ratio of fatty acids in the milk fat of the breeds under investigation did not differ significantly.

Megjegyzések:Prebiotics are such indigestible food ingredients that enter the colon and serve as nutrient for bifidobacteria and lactobacilli. Since fibres and oligosaccharides are the typical prebiotics, we produced prebiotics in our experiments with the reaction of lactose and malic acid as well as citric acid, where these reactions made use of an appropriate concentration of these substances, had an adequate duration, and were carried out under optimal temperature conditions. We determined the optimal parameters of the reaction, measured the loss of the starting materials as well as the increase in concentration of the end-product, and analysed the total sugar content of the hydrolysed prebiotics after hydrolysis by hydrochloric acid. In vitro experiments were performed to demonstrate our end-product's resistance to carbohydrate-degrading enzymes, which is a fundamental requirement for a prebiotic so that upon reaching the colon it can serve as nutrient for the probiotic bacteria found there.

Megjegyzések:have developed methods for the production of protected methionine and protected lysine, making use of the reaction between citric acid and malic acid as well as methionine and lysine, on the one hand, and of the interaction between swollen bentonite and the two amino acids, on the other hand. Our in vivo and in vitro experiments have demonstrated that one part of the amino acids transformed during the reaction, while another part bound on the bentonite's surface to a significant degree. Assisted by the reaction between hydroxycarboxylic acids and amino acids, we achieved a protection of about 75% for methionine and 60% for lysine, that is, 25% of the methionine and 40% of the lysine appeared in the free amino acid fraction. The swollen bentonite bound 75% of the added methionine and 60% of the added lysine. Our chemical analyses have demonstrated that through the time{temperature combinations applied by us the methionine and lysine do not undergo signi cant degradation and can be fully released from the protected form. Further, our in vitro experiments using rumen uid from stulated cattle showed that during the average retention time of the fodder in the rumen the protected amino acids will resist microbial enzymes and maintain their protected status during their presence in the rumen.

Megjegyzések:We have developed a new procedure for reducing soy trypsin inhibitor activity by means of heat treatment combined with chemical methods, through which soy trypsin inhibitor activity decreases to the tenth or twentieth part of the original value. We determined the optimal concentration of the applied chemicals (hydrogen-peroxide, ammoniumhydroxide) as well as the optimal temperature and duration of the treatment. The chemical procedure combined with heat treatment results in lower energy consumption as compared to the original heat treatment methods.

Megjegyzések:Mercaptoethanesulfonic acid (MES-OH) can be applied forhydrolyzing proteins. The aim of our research was to examine if it can beused also as a derivatization reagent for fluorescens detection of aminoacids together with OPA (o-phthaldialdehyde) instead of ME (mercap-toethanol) owning to its thiol-group. Corn, soybean and meatmeal sam-ples were hydrolized with hydrochloric acid or MES-OH, derivatized withninhydrin or OPA/ME or OPA/MES-OH and analysed with IEC (ion ex-change chromatography) or RP-HPLC (reversed phase high performanceliquid chromatography). There were no significant differences amongthe amino acid composition results of samples irrespective of choice of the hydrolysis or derivatization methods. MES-OH can be applied notonly for hydrolysis but also for derivatization of amino acids. In caseof samples with high protein content (>50%), due to the dilution afterhydrolysis the MES-OH concentration could be insufficient for derivativeformation, in these cases an extra MES-OH addition is required prior toderivatization.

Megjegyzések:The authors examined the vitamins B1, B2 and C con-tent of meatballs and also the D-aspartic acid and D-glutamic acidcontent plotted against the cooking time. The vitamin B1 mixed in 50 mg/100 g concentration to the normal-sized meatball changed mini-mal in 10 minutes, but during 20 minutes baking, 70% decomposed. Thevitamin B2 seems to resist more against microwave treatment since itsconcentration in 10 minutes reduced from 50 mg/100 g to 43 mg/100 g,and after 20 minutes of microwave treatment it reduced to 35 mg/100 g.The vitamin C content of the meatballs (50 mg/100 g initial concentra-tion), applying the two methods of our own was 20-22 mg/100 g after 10 minutes and 13-14 mg/100 g after 20 minutes. No significant differ-ences were found in the vitamin content of inside and outside of themeatball. The D/L aspartic acid ratio was 0.048 in the non-treated sam-ple, the D/L glutamic acid ratio was 0.027. This ratio concerning theaspartic acid increased to 0.069 in 10 minutes, 0.151 in 20 minutes, andconcerning the glutamic acid increased to 0.042 in 10 minutes, and 0.047 in 20 minutes.

Megjegyzések:The biological role of selenium was discovered in the sec-ond half of the last century. It was established that selenium was apart of the antioxidant system of the organism, and it protected theorganism from the attack of the harmful free radicals. The researchesmade it clear that the organically bound selenium, like selenomethio-nine (SeMet) and selenocysteine (SeCys), had an especially importantrole. It is very important for experts dealing with both food science andfeeding stuffs to determine the concentration of these two substances infoods and feeding stuffs. We have no information on whether the enan-tiomers of these two seleno-amino acids have ever been examined beforeby anyone else; therefore, we have elaborated new methods and amendedold ones, respectively, for the exact determination of these compounds byion-exchange column chromatography (IEC) and high-performance liquidchromatography (HPLC). In the first step, we carried out the separationof the seleno-amino acids along with protein-components amino acids byion-exchange column chromatography. In the next step, we attemptedthe separation of the oxidized derivatives of these amino acids by IEC,utilizing our experiences gained during the determination of methion-ine (Met) and cysteine (Cys) in oxidized form. Afterwards, we triedto increase the sensibility by applying HPLC after precolumn deriva-tization with OPA/2-mercaptoethanol and OPA/2-mercaptoethane sul-phonic acid, respectively, as the seleno-amino acids are contained in thebiological samples, in a several orders of magnitude lower concentrationas compared to the protein-component amino acids. For the separa-tion of the seleno-amino acid enantiomers, the OPA/TATG derivatiza-tion method was used, which we applied for amino acid enantiomersbeforehand, but derivatization with 1-(9-fluorenyl)-ethyl chloroformate(FLEC) was also attempted.
