Megjegyzések:Although biparental care is thought to be rare among insects, it has evolved independently numerous times resulting in diverse forms and varying levels of parental involvement. Several theoretical studies address potential factors promoting biparental care in beetles; however, empirical studies target principally only species of Scarabaeidae and Silphidae families. Here, we investigated the behavioural repertoire of the breeding pairs in two natural populations of a geotrupid beetle Lethrus apterus with parental care. Changes in female care as a response to the male`s absence were also investigated by experimentally removing males from pairs. We found that males spend the majority of their time at the nest entrance, while females leave their nest and collect leaves at a significantly higher rate than males. We also showed that males often desert their nests and were usually replaced by new males in a short time. The desertion of males, however, does not seem to influence females` behaviour. These findings indicate that L. apterus may have a dynamic mating system with typically short-term mate fidelity. Furthermore, our results have advanced our understanding of parental roles and nesting behaviour and contributed to the description of the distinct roles of parents in different phases of nesting events in L. apterus beetles.

Megjegyzések: Hormones play an important role in the regulation of physiological, developmental and behavioural processes. Many of these mechanisms in insects, however, are still not well understood. One way to investigate hormonal regulation is to analyse gene expression patterns of hormones and their receptors by real-time quantitative polymerase chain reaction (RT-qPCR). This method, however, requires stably expressed reference genes for normalisation. In the present study, we evaluated 11 candidate housekeeping genes as reference genes in samples of Lethrus apterus, an earth-boring beetle with biparental care, collected from a natural population. For identifying the most stable genes we used the following computational methods: geNorm, NormFinder, BestKeeper, comparative delta Ct method and RefFinder. Based on our results, the two body regions sampled (head and thorax) differ in which genes are most stably expressed. We identified two candidate reference genes for each region investigated: ribosomal protein L7A and RP18 in samples extracted from the head, and ribosomal protein L7A and RP4 extracted from the muscles of the thorax. Additionally, L7A and RP18 appear to be the best reference genes for normalisation in all samples irrespective of body region. These reference genes can be used to study the hormonal regulation of reproduction and parental care in Lethrus apterus in the future.

Megjegyzések:In this study we attempted to isolate 13 additional microsatellites from the beetle Lethrus apterus and then to use them to screen for polymorphisms in 35 specimens collected from the northern Hungarian mountains. The newly developed markers presented in this paper are polymorphic, including between two and seven detected alleles per locus. The observed and expected heterozygosity per locus ranged between 0.057 and 0.686 and between 0.056 and 0.756, respectively, while two loci showed significant deviations from Hardy-Weinberg equilibrium. These deviations can be explained by the presence of null alleles. All loci cross-amplified in at least two out of four congeneric Lethrus species (L. bituberculatus, L. scoparius, L. strymonensis and L. perun). The polymorphic microsatellite markers we present for L. apterus will be valuable for future population genetic studies.

Megjegyzések:Fifteen new microsatellite markers for the beetle Lethrus apterus were developed and tested in 45 specimens from the North Hungarian Mountains. Fourteen of the markers developed were polymorphic, and the number of alleles per locus ranged from two to nine. The observed and expected heterozygosities of the polymorphic markers ranged from 0.178 to 0.578, and 0.201 to 0.698, respectively. One locus showed significant deviation from the Hardy-Weinberg equilibrium, probably due to null alleles. The primers were tested on four other Lethrus species (L. bituberculatus, L. scoparius, L. strymonensis and L. perun) and six other coleopteran species (Copris hispanus, Geotrupes stercorarius, Melolontha melolontha, Onthophagus taurus, Oryctes nasicornis and Protaetia affinis). Thirteen loci showed cross-amplification in the Lethrus species and only three loci could be amplified in some of the six other coleopteran species. The developed markers will be valuable in investigating the population genetic structure, behaviour and reproductive biology of L. apterus.

Megjegyzések:The Eurasian Steppe belt is one of the largest biomes in the Northern Hemisphere. We provide here a range-wide phylogeography of the fightless steppe beetle Lethrus apterus that inhabits the western part of the Steppe belt through the study of population-level variance of mitochondrial cytochrome oxidase I sequences and nuclear microsatellites. We detected a concordant geographic structure of genetic data with a signifcant isolation-by-distance pattern. We found more genetic variation in the western part of the area and identifed Northern Bulgaria and the Pannonian Basin as possible refugia. Genetic clusters were separated by main rivers in the eastern part of the area. This implies west-to-east colonisation and argues for an evolutionarily recent arrival of this species to its current main distribution area, the Pontic Steppes. This contradicts the classical biogeographical wisdom that assumed an east-to-west colonisation pattern.
