Megjegyzések:A 1-isopropyl-3-phospholene 1-oxide was prepared and the two enantiomers were isolated from the racemate by resolution using optically active TADDOL derivatives or the acidic Ca2+ salts of (?)-O,O-diaroyl-(2R,3R)-tartaric acids. The single crystal X-ray structure of the 1-isopropyl-3-phospholene oxide ? spiro-TADDOL 1:2 associate revealed the mode of binding between the host and guest molecules. The role of the interactions between the three molecules was supported not only by the contact data, but also force field and semiempirical calculations. Beside X-ray analysis, the absolute configuration of the P-stereogenic center was also determined on the basis of CD spectroscopy and high level quantum chemical calculations. The racemic and optically active 1-isopropyl-3-phospholenes obtained after deoxygenation were converted to the corresponding borane complexes and Pt(II) complexes. Stereostructure of the latter species was evaluated by high level quantum chemical calculations and the Pt complexes were tested as catalysts in the hydroformylation of styrene.

Megjegyzések:An efficient resolution method was elaborated for the preparation of (+)-4-chloro-5-methyl-1-phenyl-1,2,3,6-tetrahydrophosphinine oxide using the acidic Ca2+ salt of (?)-O,O-di-p-toluoyl-(2R,3R)-tartaric acid. Crystal structure of the diastereomeric complex was evaluated by single crystal X-ray analysis. Beside this, the absolute P-configuration was also determined by a CD spectroscopic study including theoretical calculations. The tetrahydrophosphinine oxide was then converted to the corresponding platinum complex whose stereostructure was investigated by high level quantum chemical calculations. The Pt-complex was tested as catalyst in the hydroformylation of styrene.

Megjegyzések:Endomorphins (EM-1 and EM-2) are selective, high affinity agonists of the ?-opioid (MOP) receptor, an important target in pain regulation. Their clinical use is impeded by their poor metabolic stability and limited entry to the central nervous system. In this study the Pro2 residue of EM-2 was modified systematically through substitution by hydroxyproline (Hyp), (S)-?-homoproline (?Pro), 2-aminocyclopentene-1-carboxylic acid (?Acpc) or 2-aminocyclohexene-1-carboxylic acid (?Achc) to obtain stable MOP active compounds. Both Hyp2 and ?Pro2 substitution decreased receptor affinity. Analogues incorporating alicyclic ?-amino acids exhibited diverse receptor binding properties, depending on the configuration of the substituent side-chain. (1S,2R)?Acpc2-EM-2 was shown to have MOP affinity and selectivity comparable to those of EM-2 and proved to act as agonist, while being resistant to proteolysis. NMR and molecular dynamics (MD) studies revealed that bent backbone structures are predominant in the most potent analogues, while their presence is less pronounced in ligands of lower receptor affinity.

Megjegyzések:Among the pathogen-associated carbohydrate patterns, the Man(a1?2)Man a disaccharide motif is of particular interest because its multivalent derivatives are considered as potential antiviral or antibacterial agents through interaction with mannose-binding lectins. We present a straightforward synthesis of amphiphilic compounds containing a hydrolytically stable S-linked 1,2-mannobioside residue, a tetraethylene glycol linker to ensure water solubility and various lipophilic carriers such as a hexadecyl chain and two pyrrolidinofullerene derivatives. According to a dynamic light scattering study, the obtained amphiphiles form nanoscale aggregates in water producing multivalent presentation of the thiomannobioside residue.

Megjegyzések:The NMR pulse sequence design strategy of NORD (NO Relaxation Delay) is extended to design of two new three-module experiments, NORD {HMBC}-{HSQC-TOCSY}-{TOCSY} and NORD {HMBC}-{2BOB}-{TOCSY}, each delivering four spectra - HMBC, HSQC, TOCSY, and either HSQC-TOCSY or H2BC. Compared to individual recording of these spectra particularly the sensitivity of the least sensitive mod-ule, HMBC, is enhanced by designing the homonuclear TOCSY module to allow buildup of magnetization pertinent to HMBC during its execution. Effectively, the sensitivity of the heteronuclear modules is boosted at the expense of the inherently much higher TOCSY sensitivity, thus resulting in a significant saving in spectrometer time.

Megjegyzések:Anuroctoxin (AnTx), a 35-amino-acid scorpion toxin containing four disulfide bridges, is a high affinity blocker of the voltage-gated potassium channel Kv1.3, but also blocks Kv1.2. To improve potential therapeutic use of the toxin, we have designed a double substituted analog, [N17A/F32T]-AnTx, which showed comparable Kv1.3 affinity to the wild-type peptide, but also a 2500-fold increase in the selectivity for Kv1.3 over Kv1.2. In the present study we have achieved the chemical synthesis of a Sec-analog in which all cysteine (Cys) residues have been replaced by selenocysteine (Sec) forming four diselenide bonds. To the best of our knowledge this is the first time to replace, by chemical synthesis, all disulfide bonds with isosteric diselenides in a peptide/protein. Gratifyingly, the key pharmacological properties of the Sec-[N17A/F32T]-AnTx are retained since the peptide is functionally active. We also propose here a combined experimental and theoretical approach including NOE- and 77Se-based NMR supplemented by MD simulations for conformational and dynamic characterization of the Sec-[N17A/F32T]-AnTx. Using this combined approach allowed us to attain unequivocal assignment of all four diselenide bonds and supplemental MD simulations allowed characterization of the conformational dynamics around each disulfide/diselenide bridge.

Megjegyzések:Resonance assignment is a pivotal step for any nuclear magnetic resonance (NMR) analysis, such as structure elucidation or the investigation of protein-ligand interactions. Both H-1-C-13 heteronuclear single quantum correlation (HSQC) and H-1 -H-1 correlation spectroscopy (COSY) two-dimensional (2D) experiments are invaluable for H-1 NMR assignment, by extending the high signal dispersion of C-13 chemical shifts onto H-1 resonances and by providing a high amount of through-bond connectivity information, respectively. The recently introduced HSQC-CLIP(Clean In-Phase)-COSY method combines these two experiments, providing COSY correlations along the high-resolution C-13 dimension with clean in-phase multiplets. However, two experiments need to be recorded to unambiguously identify COSY cross-peaks. Here, we propose novel variants of the HSQC-CLIP-COSY pulse sequence that edit cross-peak signs so that direct HSQC responses can be distinguished from COSY relay peaks, and/or the multiplicities of the C-13 nuclei are reflected, allowing the assignment of all the peaks in a single experiment. The advanced HSQC-CLIP-COSY variants have the potential to accelerate and simplify the NMR structure-elucidation process of both synthetic and natural products and to become valuable tools for high-throughput computer-assisted structure determination.

Megjegyzések:Novel CLIP-COSY based homo-and heteronuclear correlation experiments are reported for the rapid,semi-automated NMR assignment of small to medium-sized molecules. The homonuclear CLIP-COSY and corresponding relayed experiment semploythe perfect-echo based mixing sequence for in-phase coherence transfer between directly and/or indirectly coupled proton spins.The combined analysis of the resulting CLIP-COSY and relayed spectra made it possible to easily track down,layer by layer,the proton?proton connectivity network.In larger molecules the narrow chemical shift range of protonsmay,however,compromise the efficacy of the homonuclear correlation based assignment strategy.To overcomet his limitation,an HSQC variant of the CLIP-COSY experimenthas now been devised.Combined treatment of HSQC-CLIP-COSY (relayed)and standard HSQC spectra facilitates simultaneous and semi-automatic assignment of 1H and 13C resonances of medium-sizedmolecules,such as pentasaccharides.The recently introduced PSYCHE broadband homonuclear decoupling scheme has been also implemented in to the devised homo-and heteronuclear CLIP-COSY based experiments,resulting in fully decoupled high-resolution pure-shift correlation spectra.

Megjegyzések:Their emerging nature as multifunctional effectors explains the large interest to monitor glycan binding to galectins and to define bound-state conformer(s) of their ligands in solution. Basically, NMR spectroscopy facilitates respective experiments. Towards developing new and even better approaches for these purposes, extending the range of exploitable isotopes beyond 1H, 13C, and 15N offers promising perspectives. Having therefore prepared selenodigalactoside and revealed its bioactivity as galectin ligand, monitoring of its binding by 77Se NMR spectroscopy at a practical level becomes possible by setting up a 2D 1H, 77Se CPMG-HSQBMC experiment including CPMG-INEPT long-range transfer. This first step into applying 77Se as sensor for galectin binding substantiates its potential for screening relative to inhibitory potencies in compound mixtures and for achieving sophisticated epitope mapping. The documented strategic combination of synthetic carbohydrate chemistry and NMR spectroscopy prompts to envision to work with isotopically pure 77Se-containing ?-galactosides and to build on the gained experience with 77Se by adding 19F as second sensor in doubly labeled glycosides.
