Megjegyzések:Grapevine Trunk Diseases (GTD) are amongst the most important diseases in vineyards. GTD have been characterized only recently because of a worrying increase of the frequency of their symptom expression worldwide. Esca-, Petri- and black-foot disease, Eutypa- and Botryosphaeria- dieback are considered as major GTDs, are caused by a variety of pathogens which attack the woody perennial organs of the vine and ultimately lead to the death of the plant. Control of GTD is problematic, no grapevine cultivar is known to be resistant to GTD and there are no highly effective treatments available. The present knowledge of epidemiology, host-pathogen interaction and microbial ecology is still poor. The objective of this study was to isolate and identify fungi associated with symptoms of GTD and from canes of healthy grapevines in vineyards from the Tokaj wine region. The plantation studied is 21 years old of grapevine variety called Furmint. Four rows of grapevine were chosen for the collection of samples. Pure fungal cultures were used for morphological and molecular identifications of fungal species. Chips were cut, disinfected and incubated on potato dextrose agar medium at room temperature. Fungal DNA was obtained from pure fungal colonies after 3 days of growth on 50 ml potato dextrose broth on a rotary shaker (125 rpm). Mycelia were harvested by vacuum filtration. Total genomic DNA was extracted from freeze-dried mycelium and isolated using NucleoSpin Plant II (Macherey-Nagel) according to the protocol, followed the manufacturer's instructions. DNA concentrations were measured by NanoDrop (Thermo Scientific) and used in PCR amplification of the internal transcribed spacer region (ITS) and the 5.8S ribosomal subunit with primers ITS4 and ITS5 [1]. The PCR products were purified by using Nucleospin Gel and PCR Clean-up (Macherey-Nagel). Purified amplification products were sequenced by MWG Biotech Company in Germany. Sequences obtained were assembled and manually corrected. Reference sequences for the Botryosphaeriaceae were obtained from GenBank. Phylogenetic analyses were performed with MEGA. One thousand boostrap analyses were run to determine confidence levels at the branching points. The principal aim of this work is to assess the GTD types and their occurrence in the Tokaj wine region as well as the identification of the pathogenic fungi (isolated from symptomatic and asymptomatic grapevine trunks) on morphological and genetic basis. In this study, 101 fungal isolates were identified from grapevine woody tissues. The majority of them (72.3% ) were determined as member of Botryosphaeriaceae, 66 sample were determined as Diplodia seriata , 6 Botryosphaeria stevensii. Other fungi, like Alternaria, Mucor, Phoma, Phomopsis, Diaporthe and Xylaria also were identified from grapevine samples.

Megjegyzések:Grapevine trunk diseases (GTD) are one of the most important diseases in vineyards worldwide, which can be found in Hungarian vineyards as well. GTD are caused by a variety of pathogen fungi, like: Phaeoacremonium aleophilum, Phaeomoniella chlamydospora, Eutypa lata, Fomitiporia mediterranae, Diplodia seriata. There is very limited information about the occurrence of different pathogens causing GTD in the Tokaj wine region nowadays. Currently there is no effective fungicide, which could be used against the disease.The objective of the research were estimated the occurrence of grapevine trunk diseases in Tokaj wine region in cooperation with local specialists; furthermore, to isolate and identify fungi associated with symptoms of GTD in vineyards.Tokaj wine region have indigenous grape varieties (Hárslevelű and Furmint). The samples came from the Bakonyi area (101 samples). We cooperated with the staffs of the Research Institute for Viticulture and Oenology (Tokaj) in the collection of the grapevine trunk samples. Pure fungal cultures were used for morphological (colony formation, pigment production and the shape of the spore) and molecular identification of fungal species. The woody tissue samples were cut in a small part, and were disinfected with 10% Neomagnol (tosylchloramide sodium B) solution for 10 minutes, than were washed twice with sterile distilled water. Afterwards they were put on malt extract agar medium and incubated at room temperature (25 ?C) for 3-8 days. The isolates were maintained on potato-dextrose agar (PDA) at 4 ?C. Fungal DNA was extracted from three days old fungal mycelia growth on 50 ml potato dextrose broth on a rotary shaker (125 rpm) at room temperature, and was harvested by vacuum filtration. MagNaLyser (Roche) was used for disruption, and isolation was carried with NucleoSpin Plant II (Macherey-Nagel). The ITS4 and ITS5 primers were used for PCR amplification of the internal transcribed spacer regions. Purified amplicons were sequenced by MWG Biotech Company in Germany. The sequences were aligned with deponated reference sequences with Clustal X program, and manually corrected with GeneDoc. Phylogenetic analyses were performed with MEGA 5.05 (Kovács et al., 2014).The foliar symptoms could be observed on 0.46% (Szemere-dűlő), 3.49% (Dorgó-dűlő); 4.27% (Szarvas-dűlő) and 0.34% (Várhegy) of the examined trunks. However, dieback could be detected only in few cases. More than one hundred fungi were isolated in case of Bakonyi area from symptomatic woody tissues. In case of Bakonyi area the majority of species (72.3%) were determined as member of Botryosphaeriaceae, sixty-six samples were determined seventy-three samples as Diplodia seriata, and six samples as Botryosphaeria stevensii. Other fungi, like Alternaria (four), Mucor ramossimus (nine), Phoma sp. (one), Diaporthe (two) and Xylaria (seven),Gibberella zea (one),Pheniophora sp.(one), Xanthomendoza sp. (one) were also identified from grapevine trunk samples.

Megjegyzések:A tőkeelhalás napjainkban a szőlőültetvények egyik legsúlyosabb betegsége világszerte, mely a magyarországi szőlőkben is megtalálható.A magyarországi borvidékeken a tőkeelhalás kialakításában szerepet játszó kórokozók előfordulásáról azonban nagyon kevés adat található,a Tokaj-hegyaljai borvidéken pedig nincsenek ismereteink arról, hogy jelenleg a tárgyalt betegséggel összefüggésbe hozható kórokozók közülme lyek találhatóak meg az ültetvényekben.Kutatómunkánk célja a Tokaj-hegyaljai borvidéken észlelt fertőző tőkeelhalás néven összefoglalt egyes betegségtípusok helyzetének felmérése, a helyi szakemberek közreműködésével, továbbá a beteg tőkékből izolálható kórokozók morfológiai és genetikai alapon történő azonosítása.A vizsgált, elhalt tőkék mindegyikéből sikerült endofita gombákat kitenyészteni. Az izolátumok között döntő többségében a Botryosphaeriaceae családba tartozó Diplodia seriata volt kimutatható a tenyészetek morfológiai és az rDNS régió alapján történt molekuláris azonosítása alapján.

Megjegyzések:Zasmidium cellare (wine cellar mould, Syn.: Cladosporium cellare) was reported previously the most frequently occurred mould on the inner surfaces of the wine cellars , but other mould genus (e.g. Alternaria, Aspergillus, Aureobasidium, Cladosporium, Geosmithia, Penicillium, and Verticillium) have been identified, too. Since very few reports have been published in this topic in Hungary, we aimed to identify some isolates derived from the inner surface of some winery from the Tokaj wine region. Mycelial samples were collected from the inner surface of two cellars near to Tokaj in 2014 spring. The samples were collected with sterile forceps and were transported in sterile centrifuge tube to the Microbiological Laboratory of Institute of Food Science (University of Debrecen). Sample preparation and microbiological examination were carried out according to the protocol of Goto et al. (1989). Following the morphological tests, the molecular biological identification of the isolates was performed with the help of ITS1 and ITS2 sequences (Schoch et al., 2012). ITS4 and ITS5 primers were used for PCR amplification of the internal transcribed spacer regions (White et al., 1990). The PCR products were purified with using NucleoSpin Gel and PCR Clean-up. Purified amplicons were sequenced by Mycrosinth, Austria. The sequences were aligned with Clustal X program, and manually corrected with GeneDoc. Phylogenetic analyses were performed with MEGA 5.05.
