Cylindrospermopsin (CYN) is a cyanobacterial toxin known as a eukaryotic protein synthesis inhibitor.We aimed to study its effects on growth, stress responses and mitosis of a eukaryotic model, Vicia faba(broad bean). Growth responses depended on exposure time (3 or 6 d), cyanotoxin concentration, cultureconditions (dark or continuous light) and V. faba cultivar (♭♭Standard'' or ♭♭ARC Egypt Cross''). At 6 d ofexposure, CYN had a transient stimulatory effect on root system growth, roots being possibly capableof detoxification. The toxin induced nucleus fragmentation, blebbing and chromosomal breaks indicatingdouble stranded DNA breaks and programmed cell death. Root necrotic tissue was observed at0.1?20 lgmL 1 CYN that probably impeded toxin uptake into vascular tissue. Growth and cell death processesobserved were general stress responses. In lateral root tip meristems, lower CYN concentrations(0.01?0.1 lgmL 1) induced the stimulation of mitosis and distinct mitotic phases, irrespective of cultureconditions or the cultivar used. Higher cyanotoxin concentrations inhibited mitosis. Short-term exposureof hydroxylurea-synchronized roots to 5 lgmL 1 CYN induced delay of mitosis that might have beenrelated to a delay of de novo protein synthesis. CYN induced the formation of double, split and asymmetricpreprophase bands (PPBs), in parallel with the alteration of cell division planes, related to theinterference of cyanotoxin with protein synthesis, thus it was a plant- and CYN specific alteration.

Microcystin-LR (MCY-LR), a cyanobacterial toxin alters subcellular organization in common reed (Phragmites australis), an ecologically important aquatic plant. MCY-LR co-occurs with common reed populations. It is an inhibitor of type 1 and 2A protein phosphatases (PP1 and PP2A). They play crucial roles in eukaryotic cellular events including regulation of cell cycle, and developmental events like auxin-mediated root morphogenesis in plants. Our further experiments were done in Arabidopsis and Vicia faba. Inhibition of PP1 and PP2A activities by MCY-LR induced (i) chromatin and spindle alterations in mitotic cells and (ii) hyperphosphorylation of histone H3 that delayed metaphase/ anaphase transition, highlighting the essential role of PP1 and PP2A in plant cell cycle regulation. Subcellular changes led to altered elongation growth, thus abnormal root morphology. Primordia of lateral roots develop prematurely, but they are not able to emerge from mainroots. Such anomalies show direct correlation with abnormal distribution of auxin efflux carriers (PINs). MCY-LR can be a tool for the study of PP1/PP2A mediated developmental processes.

Cylindrospermopsin (CYN) is a tricyclic guanidino derivative, an alkaloid toxin produced by several cyanobacterial genera. It alters cellular functioning in eukaryotes, including animal and plant organisms. Over the past decades, more and more evidence shows its potential hazardous effects on animal and human health. In this review, we give a critical survey and interpretation of data currently available on its biochemical and consequently, cellular effects. CYN is considered to be a cytotoxin. Several reports suggest that it is a potent inhibitor of eukaryotic protein synthesis, though the exact mechanisms are far from not completely understood. Here we show that the biochemical changes induced by CYN suggest are complex, and possibly involving multiple modes of action. Glutathione metabolism and pyrimidine nucleotide synthesis is affected besides the proposed protein synthesis inhibition. Biochemical alterations lead to the following cellular/subcellular alterations both in animals and plants: (i) changes in cell division rates due to perturbations in chromatin and cytoskeleton; (ii) perturbations of structure and functioning of endomembranes including endoplasmic reticulum; (iii) general metabolic alterations leading to genotoxicity and programmed cell death/apoptosis. The underlying mechanisms and possible health consequences are discussed.
