Megjegyzések:Due to several byproducts derived from Jerusalem artichoke (JA), it has several ecological and economical benefits in bioenergy production, functional foods and human health. These bio-based multi-products include fructose, inulin, antioxidants, natural fungicides and bio-ethanol. This study included the assessment of some biochemical components of in vitro callus and in vivo leaves and tubers of three JA cultivars and their significant role for human health. Inulin as a functional food ingredient, fructose, protein and polyphenols were selected. The most important findings demonstrated that the inulin content in tubers of any studied JA cultivar was 4-5 folds that in callus. The lowest values of fructose among JA fractions were recorded for callus of the three studied cultivars. Both callus and leaves of all studied JA cultivars showed the highest crude protein comparing with the tubers. The total soluble phenols were higher in callus of all JA cultivars than both leaves and tubers. This study may open a window on the potential of plant tissue culture techniques for increasing such important bioactive compounds for human health. This is the first report handles the comparison among callus, tubers and leaves of three JA cultivars in their content of the bioactive compounds. Further investigations are required not only to determine the fractionations of the bioactive compounds but also different approaches of tissue culture techniques should be employed to increase the studied bioactive components particularly the inulin and fructose in the investigated JA cultivars.

Megjegyzések:It is necessary to develop and deploy novel protein production to allow the establishment of a sustainable supply for both humans and animals, given the ongoing expansion of protein demand to meet the future needs of the increased world population and high living standards. In addition to plant seeds, green biomass from dedicated crops or green agricultural waste is also available as an alternative source to fulfill the protein and nutrient needs of humans and animals. The development of extraction and precipitation methods (such as microwave coagulation) for chloroplast and cytoplasmic proteins, which constitute the bulk of leaf protein, will allow the production of leaf protein concentrates (LPC) and protein isolates (LPI). Obtained LPC serves as a sustainable alternative source of animal-based protein besides being an important source of many vital phytochemicals, including vitamins and substances with nutritional and pharmacological effects. Along with it, the production of LPC, directly or indirectly, supports sustainability and circular economy concepts. However, the quantity and quality of LPC largely depend on several factors, including plant species, extraction and precipitation techniques, harvest time, and growing season. This paper provides an overview of the history of green biomass-derived protein from the early green fodder mill concept by Károly Ereky to the state-of-art of green-based protein utilization. It highlights potential approaches for enhancing LPC production, including dedicated plant species, associated extraction methods, selection of optimal technologies, and best combination approaches for improving leaf protein isolation.

Dátum:2020
Megjegyzések:Long-term predictable and reliable protein supply for food and feed production is one of the great challenges of today's agriculture both in Hungary and other EU countries. Considering economical, ethical, environmental and ecological aspects of food/feed proteins it is urgent to find alternative sources with technologies besides soy. Among alternative plantbased protein the green biomass has gained an intensive attention. Hungary is self-sufficient in alfalfa and grasses as dedicated green protein source. At the same time green biomass of alfalfa is more than only protein source, it is a rich repository of essential fatty acids and phytonutrients. The quality and quantity of phytochemicals partially depending on the processing of green biomass. After wet fractionation leaf protein concentrate (LPC) as potenital final product can be obtained from the green juice fraction using any coagulation method. The theoretical basis of this process is included in the concept of green biorefinery. In our work the phytochemical composition of fractionated alfalfa green biomass was evaluated, with special regard to LPC fraction as directly exploitable, protein-rich (40 ? 45 m/m% N) feed/food source. UHPLC-ESI-MS was applied to identify different phytonutrients. The phenolic compounds were significant part of the identified compounds. Among flavonoids 298 flavones were dominant mostly in the forms of glucosides. Just few flavone aglycones could be identified such as apigenin and luteolin. Hydroxylated methoxyflavone could be isolated from green juice and fiber fractions. Three isoflavones were found such as ononin; alfalone and formonetin. Nine saponins could be identified and seven unknown saponin aglcyons.

Megjegyzések:Jerusalem artichoke is widely known for its inulin-enriched tubers. Recently the opportunity has been arisen to involve the whole plant in biorefinery concept due to its high lignocellulose biomass and tuber production. This paper focuses on the repeatedly harvestable green biomass of Jerusalem artichoke. Ultra-High Performance Liquid Chromatography-Electrospray Ionization/Mass Spectrometry (UHPLC-ESI-MS) was applied to identify the phytochemicals in Jerusalem artichoke leaf protein concentrate (JAPC) thermally extracted from green biomass of three clones, i.e., Alba, Fuseau and Kalevala. Amino acid and fatty acid profiles as well as yield of JAPC were also analyzed. The UHPLC-ESI-MS analyses showed that no toxic phytochemicals were identified in JAPC. The results revealed, also, that JAPC is not only essential-amino acids-rich but also contains substantial amounts of polyunsaturated fatty acids (66-68%) such as linolenic and linoleic acids. Linolenic acid represented 39-43% of total lipid content; moreover, the ratio between {omega}-6 and {omega}-3 essential fatty acids in JAPC was ~0.6: 1. Using UHPLC-ESI-MS, the following hydroxylated methoxyflavones were for the first time identified in JAPC, i.e., dimethoxy-tetrahydroxyflavone, dihydroxy-methoxyflavone, hymenoxin and nevadensin. These compounds are medically important since they are referred to as anti-cancer, anti-inflammatory and antioxidants. Also, liquiritigenin - estrogenic-like compound - was identified in JAPC alongside the following terpenes, i,e., loliolide and dihydroactinidiolide. However, no remarkable differences of phytochemicals, fatty acids and amino acids composition were seen among Jerusalem artichoke clones. The green biomass of tested clones ranged between 5 to 5.6 kg m-2 and JAPC yield varied from 28.6 to 31.2 g DM kg-1 green biomass with total protein content, on average, of 33.3%. According to our knowledge, this paper is the first scientific report highlighting bioactive substances in JAPC such as PUFA phytochemicals. These results clearly prove that JAPC is a valuable product which can direct towards human and animal nutrition as well as it can serve as basic material for different industrial purposes.

Megjegyzések:The present study evaluates the green biomass of Jerusalem artichoke (JA) as an alternative green protein. A leaf protein concentrate (LPC) was prepared from leafy shoots using biotechnological methods. Seven clones were compared to assess the importance of the genetic basis of JA, and alfalfa served as the control. The LPC content of JA was an average 39 g kg?1 of fresh biomass, while that of alfalfa was 32 g kg?1 . The JA can produce up to 936 kg of protein ha?1 year?1 without fertilization under rainfed conditions. The crude protein content of the LPC varied from 24.2 to 31.4 m/m%, depending on clones and harvesting time, which was comparable to that of alfalfa LPC (~ 32.3%). The amino acid profile of the LPC of JA, particularly of the essential amino acids, was similar to alfalfa and soybean. In addition, our results confirmed that the polyunsaturated fatty acid (PUFA) content varied between 64 and 68% in the LPC fraction, regardless of the clone that was used, with linoleic acid and linolenic acid being the predominant PUFAs. In addition, unlike alfalfa, the content of arachidonic acid was 0.5% in the JA LPC. The tuber yield was significantly reduced because of the repeated harvesting of the shoot parts; however, the tubers obtained were sufficient to regenerate the plantation in the subsequent year, thus ensuring the renewable ability and sustainability of the green biomass of JA.
